Dysfunction of the brain glutamate system has been associated with alcoholism Ionotropic glutamatergic alpha-amino-3-hydroxy-5-methylisoxazole-4-propionic acid receptors (AMPARs) play an important role in both neurotransmission and post synaptic plasticity. Alterations in AMPAR densities may also play a role in the neurobiological changes associated with alcoholism. In the present study, [H-3] AMPA binding density was evaluated in the nucleus accumbens (NAc), frontal cortex, anterior cingulate cortex (ACC), dentate gyros and hippocampus of Cloninger type 1 (n=9) and 2 (n=8) alcoholics, and compared with non-alcoholic control subjects (n=10) by post-modem whole hemisphere autoradiography. The [3H] AMPA binding density was significantly higher in the ACC of early onset type 2 alcoholics when compared with controls (p=0.011). There was also a significant negative correlation between [3H] AMPA binding and previously published results of dopamine transporter (DAT) density in the ACC in these same brain samples (R=-0.95, p=0.001). Although preliminary, and from a relatively small diagnostic group, the present results help to further explain the pathology of alcohol dependence and impulsive behaviour in type 2 alcoholics. Mortality in psychotic depression: 18-year follow-up study

Tapio Paljarvi, Jari Tiihonen, Markku Lahteenvuo, Antti Tanskanen, Seena Fazel, Heidi Taipale

Summary: This study used routine data from nationwide health registers in Finland to estimate the risk of cause-specific mortality in psychotic depression compared with severe non-psychotic depression while controlling for comorbid psychiatric disorders. The results showed that individuals with psychotic depression had a higher risk of all-cause mortality, suicides, and fatal accidents during the subsequent 5-year period after the index diagnosis compared with those with non-psychotic depression. Prompt treatment and enhanced monitoring for psychotic symptoms are necessary to prevent deaths because of suicides and other external causes in patients with severe depression.

Women with Schizophrenia-Spectrum Disorders After Menopause: A Vulnerable Group for Relapse

Iris E. Sommer, Bodyl A. Brand, Shiral Gangadin, Antti Tanskanen, Jari Tiihonen, Heidi Taipale

Summary: Women with schizophrenia have a higher risk of hospitalization for psychosis compared to men, especially after the age of 45. The effectiveness of antipsychotic medications also declines in women after this age. This highlights the importance of providing special attention and care to older women with schizophrenia.

Summary: This study aimed to establish the risk of suicide associated with incident psychotic depression (PD) compared to incident non-psychotic severe depression (NPD). The results showed that PD was associated with a twofold increased risk of suicide compared to NPD, with the highest relative risks for impact-related and suffocation-related suicides. This study revealed some characteristics of PD patients in terms of the severity of suicidal ideation and choice of self-harm methods.

Johan Lundberg, Thomas Cars, Sven-ake Loeoev, Jonas Soederling, Johan Sundstroem, Jari Tiihonen, Amy Leval, Anna Gannedahl, Carl Bjoerkholm, Mikael Sjaelin, Clara Hellner

Summary: This cohort study estimated the burden of treatment-resistant depression (TRD) and developed a prognostic model for clinical use. The study found that TRD is associated with a high burden for both patients and society, and the time between initiation of new antidepressant treatments is longer than recommended in current treatment guidelines.

Summary: Six rare SLCO1B1 single nucleotide variants (SNVs) were identified in Finnish individuals with a psychotic disorder, which may affect the expression and functionality of the OATP1B1 protein. Functional studies showed that one of these SNVs (c.629G>T) resulted in the loss of OATP1B1 function and decreased membrane protein expression.

Impacts of maternal microbiota and microbial metabolites on fetal intestine, brain, and placenta

Aleksi Husso, Tiina Pessa-Morikawa, Ville Mikael Koistinen, Olli Karkkainen, Hyuk Nam Kwon, Leo Lahti, Antti Iivanainen, Kati Hanhineva, Mikael Niku

Summary: Maternal microbiota has a significant impact on fetal development, with gene expression differences in the fetal intestines, brains, and placenta associated with microbial metabolite concentrations. Male fetuses may be more susceptible to microbial modulation, affecting genes important for immune system, neurophysiology, translation, and energy metabolism before birth. Several microbial metabolites have been identified in this context, with many important metabolites yet to be discovered.

The majority of patients did not adhere to their prescribed medications, with factors such as young age, recent diagnosis of bipolar disorder, multiple hospitalizations, and use of benzodiazepines or antidepressants being associated with an increased risk of non-adherence.

Comparative steroid profiling of newborn hair and umbilical cord serum highlights the role of fetal adrenals, placenta, and pregnancy outcomes in fetal steroid metabolism

Johanna Koskivuori, Raimo Voutilainen, Markus Storvik, Merja R. Hakkinen, Lauri Uusitalo, Leea Keski-Nisula, Katri Backman, Seppo Auriola, Marko Lehtonen

Summary: This study compared the concentrations of 17 steroids in newborn hair and umbilical cord serum. The results showed high individual variations in steroid hormone concentrations in both sample matrices. The concentrations of cortisol, corticosterone, estrone, estradiol, dehydroepiandrosterone, 118-hydroxyandostenedione, 5α-androstanedione, and 17α-hydroxypregnenolone were positively correlated between cord serum and newborn hair samples. In addition, cortisol and 11bOHA4 concentrations were positively correlated with each other in both sample types. Minor sex differences in steroid concentrations were observed, and parity and delivery mode were significant factors associated with certain adrenocortical steroid concentrations.
