Effect and analysis of phenolic compounds during somatic embryogenesis induction in Feijoa sellowiana Berg

The effect of phenolic compounds on somatic embryogenesis in Feijoa sellowiana was analysed. The results showed that caffeic acid (140-560 mu M) significantly increased somatic embryogenesis induction compared with the control. The presence of phloridzin, even at lower concentrations (11.5 mu M), or caffeic acid or phloroglucinol at concentrations greater than 140.0 and 197.5 mu M, respectively, inhibited somatic embryo development beyond the globular stage. When somatic embryos were transferred to the germination medium, the highest rates of germination (81.9%) were obtained with embryos induced in the presence of phloroglucinol (79.0 mu M). At all concentrations tested, somatic embryos induced in medium containing phloroglucinol germinated at higher rates than those induced in the presence of caffeic acid. Histological and ultrastructural studies showed that somatic embryos were formed in close association with phenolic-rich cells which, in more advanced stages of development, formed a zone isolating the embryo from the maternal tissue. A comparative analysis of total phenolic content indicated that phenolics reached a peak by the third week of culture, independently of the medium used. However, after that period, the amount of phenolic compounds was significantly higher in explants cultured in the presence of phloroglucinol than in those cultured in the control or in caffeic acid-containing medium. Attempts to identify the type of phenolic compounds showed that flavan-3-ols and gallic acid derivatives were mainly produced in phloroglucinol-containing medium, whereas flavanones and dihydroflavonols were also present in medium containing caffeic acid. Flavones were the main phenols detected in the control. The ways in which phenolic compounds may affect somatic embryogenesis are discussed.

RNA methyltransferases in plants: Breakthroughs in function and evolution

Summary: The field of epitranscriptomics, specifically the understanding of RNA methylation, is rapidly expanding and becoming increasingly complex and challenging. Methyltransferases, demethylases, and RNA-binding proteins play crucial roles in adding, removing, and recognizing methyl groups on RNA. Plant methyltransferases have been identified and linked to stress and development processes, but the understanding of these enzymes, particularly those located in organelles, is still in its early stages. Comparative and evolutionary analyses have revealed the existence of multiple yet uncharacterized methyltransferases, and this review discusses the functions of nuclear and organellar RNA methyltransferases in plant development and stress responses, as well as the discoveries made across different kingdoms.

Antioxidant Potential of Tamarillo Fruits-Chemical and Infrared Spectroscopy Analysis

Summary: This study aimed to investigate the antioxidant activity of four genotypes of tamarillo. The Mealhada genotype showed the highest content of phenolic compounds, flavonoids, and anthocyanins, as well as the highest antioxidant activity. Fourier-transform infrared spectroscopy revealed the presence of phenols, triterpenoids, and polysaccharides in the aqueous ethanolic extracts of the fruits.

An Efficient Agrobacterium-Mediated Genetic Transformation Method for Solanum betaceum Cav. Embryogenic Callus

Summary: This study successfully achieved genetic transformation of embryogenic callus in Solanum betaceum using an optimized faster protocol. The success rate was increased by employing cold-shock treatment, coconut water, and polyvinylpyrrolidone. The efficiency of the process was evaluated using GUS assay and PCR-based techniques, and a 100% efficiency rate was confirmed. The findings of this study provide a useful tool for functional gene analysis and biotechnology research.

Ethylene Inhibition Reduces De Novo Shoot Organogenesis and Subsequent Plant Development from Leaf Explants of Solanum betaceum Cav.

Summary: This study investigated the role of ethylene in de novo shoot organogenesis (DNSO) and found that ethylene positively regulates shoot regeneration. Ethylene precursors ACC and ethephon increased the number of buds and shoots regenerated, while inhibiting ethylene biosynthesis or perception reduced shoot regeneration. These findings suggest that altering ethylene levels and perception could enhance shoot regeneration efficiency.
