Summary: Diarylethene-based photoswitches are widely used in chemistry, materials science, and biotechnology due to their unique physical and chemical properties. In this study, a diarylethene-based photoswitchable compound was separated and characterized using high-performance liquid chromatography, and the isomers were purified for individual study. Supercritical fluid chromatography was explored as an alternative separation method, offering faster analysis times and lower solvent consumption compared to high-performance liquid chromatography. This technique could be upscaled for future purification of diarylethene isomeric compounds, providing a more environmentally-friendly approach.

Summary: To apply metabo-lipidomics in ophthalmological and systemic diseases, it is necessary to standardize the preanalytical and analytical steps in human tears. This study compared different methods for each step of the workflow and found that methanol/water extraction provided the best results for metabolomics, while Folch extraction was superior for lipidomics. The use of microcapillary glass tube for sampling was superior to Schirmer strip for metabolomics. A volume of 5 mu L provided satisfactory metabo-lipidomic coverage. These findings will contribute to improving the metabo-lipidomic workflow in human tears for the identification of new biomarkers.

By employing off-line prep-HPLC, the soluble fractions were reliably separated and enriched, leading to the identification of heavy aromatic compounds not detected by GC/MS. Summary: In this study, modern reversed-phase high performance liquid chromatography (HPLC) and stacking-micellar electrokinetic chromatography (MEKC) methods were developed and compared for the determination of alkenylbenzenes in food flavouring ingredients. The HPLC method showed better analytical performance and faster total run time compared to the stacking-MEKC method. The developed analytical methodology was successfully applied to actual food samples, which is crucial for future studies on determining alkenylbenzenes in food.

Summary: In this study, polymerization mixture was optimized to synthesize poly(acrylamide-co-N,N'-methylenebisacrylamide) monolithic stationary phases for HILIC of intact proteins. The monolithic structure exhibited a permeable and polar stationary phase suitable for HILIC. The monolith showed superior separation performance and lower ion interaction compared to silica-based particulate amide phase, enabling successful hyphenation with high-resolution mass spectrometry in HILIC of intact proteins.

Summary: We have developed a new protocol for determining D/L amino acids using a chiral phosphinate derivatization. (RP)-L-Menthyl phenylphosphinate successfully labeled eighteen pairs of amino acids except for Cys, and the chirality of amino acids was discriminated by 31P NMR. Seventeen pairs of amino acids were separated in 45 minutes using a C18 column, with resolution values ranging from 2.01 to 10.76. The lowest limit of detection was 10 pM in parallel reaction monitoring, highlighting the potential of chiral phosphine oxides in future chiral metabolomics.

Summary: In this study, the compositional characterization of lignocellulosic biomass from three Acacia species was performed using various analytical techniques. The effects of white-rot fungi treatments on the biomass composition and enzymatic saccharification were investigated. The results showed that the pretreatments with white-rot fungi and mild alkali can enhance saccharification yield, providing valuable insights for utilizing unused biomass resources and reducing wildfire risk.

Summary: The field of epitranscriptomics, specifically the understanding of RNA methylation, is rapidly expanding and becoming increasingly complex and challenging. Methyltransferases, demethylases, and RNA-binding proteins play crucial roles in adding, removing, and recognizing methyl groups on RNA. Plant methyltransferases have been identified and linked to stress and development processes, but the understanding of these enzymes, particularly those located in organelles, is still in its early stages. Comparative and evolutionary analyses have revealed the existence of multiple yet uncharacterized methyltransferases, and this review discusses the functions of nuclear and organellar RNA methyltransferases in plant development and stress responses, as well as the discoveries made across different kingdoms.

Summary: This study aimed to investigate the antioxidant activity of four genotypes of tamarillo. The Mealhada genotype showed the highest content of phenolic compounds, flavonoids, and anthocyanins, as well as the highest antioxidant activity. Fourier-transform infrared spectroscopy revealed the presence of phenols, triterpenoids, and polysaccharides in the aqueous ethanolic extracts of the fruits.

Summary: This study aimed to improve the protocols for P. ponderosa regeneration through in vitro organogenesis by analyzing the influence of different explants, culture media, cytokinins, auxins, and light treatments on shoot multiplication and rooting phases. The results showed that whole zygotic embryos and 44 mu M 6-benzyladenine had the best survival rate for explants. Blue LED light treatment promoted shoot elongation, while fluorescent light treatment improved acclimatization percentage.

Summary: This study successfully achieved genetic transformation of embryogenic callus in Solanum betaceum using an optimized faster protocol. The success rate was increased by employing cold-shock treatment, coconut water, and polyvinylpyrrolidone. The efficiency of the process was evaluated using GUS assay and PCR-based techniques, and a 100% efficiency rate was confirmed. The findings of this study provide a useful tool for functional gene analysis and biotechnology research.
