Summary: Cathepsin L (CTSL) has been found to play a critical role in chemosensitivity and tumor progression. This study aimed to investigate the potential roles and molecular mechanisms of CTSL in chemoresistance in neuroblastoma (NB). The researchers assessed the correlation between clinical characteristics, survival, and CTSL expression, and found that CTSL served as a prognostic marker for poor clinical outcome in NB patients. Furthermore, they found that CTSL promoted chemoresistance in NB cells through up-regulating multi-drug resistance proteins, inhibiting autophagy level and cell apoptosis. The study also identified a positive correlation between CTSL expression and Serglycin (SRGN) expression, and showed that CTSL could mediate chemoresistance by up-regulating SRGN expression in NB cells.

Summary: Neuroblastoma is a common solid tumor in pediatric patients that arises from sympathetic nervous system cells. Current treatments have limited success, leading to high mortality and relapse rates. Marine cyanobacteria, which contain bioactive metabolites, have shown promise as alternative treatments. This review focuses on the anticancer effects of cyanobacterial peptides against neuroblastoma. Marine peptides offer advantages such as small size, easy manufacturing, ability to cross cell membranes, minimal drug interactions, little impact on the blood-brain barrier, and targeting capabilities. The review discusses the cytotoxic effects of cyanobacterial peptides, including apoptosis, caspase activation, cell cycle arrest, sodium channel blocking, autophagy, and anti-metastasis properties.

Summary: In this study, the feasibility and mechanism of cordycepin combined with doxorubicin for liver cancer treatment were investigated. Both in vitro and in vivo studies demonstrated that the combination effectively inhibited cell proliferation and induced apoptosis in hepatocellular carcinoma. Network pharmacology analysis revealed the potential targets and pathways involved in the anticancer mechanism. Additionally, the combination treatment regulated the expression of genes related to cell migration, thus inhibiting cell migration.

Summary: This study investigated the combined effects and apoptotic mechanisms of hyperthermia and cordycepin on human leukemia U937 cells. The results showed that combined treatment with hyperthermia and cordycepin significantly augmented apoptosis by upregulating Bax and suppressing Bcl-2, Bid, and activated caspase expression. The MAPK pathway and mitochondrial dysfunction were also involved in the induction of apoptosis. Furthermore, NAC could rescue the cells by restoring ROS levels and caspase-3 activity. Cordycepin has the potential to be used as a hyperthermia sensitizer to treat leukemia.

Summary: Recent studies have shown that radiofrequency (RF) can reduce DNA damage induced by chemical or physical agents, similar to the adaptive response observed in radiobiology. This effect has been reported in vitro and in vivo, and possible cellular mechanisms activated by RF pre-exposure include perturbation of the cell redox status, DNA repair mechanisms, and stress response machinery. This study found that autophagy contributes to the RF-induced adaptive response in human neuroblastoma cells.
