Summary: This study analyzed the impact of fluid balance on the liberation of invasive ventilation in COVID-19 patients with ARDS. The findings showed that a higher cumulative fluid balance was associated with a lower probability of successful ventilation liberation, indicating that restricted fluid management may be beneficial in these patients.

Summary: QKI-5 is significantly decreased in metastatic lung adenocarcinoma, and its overexpression inhibits TGF-beta-induced EMT and invasion. KLF6 positively regulates QKI-5 at the transcriptional level, which is associated with TGF beta R1. Targeting the KLF6/QKI-5/TGF beta R1 axis may be a promising strategy for treating LUAD.

Summary: This study found that higher levels of Syn-1 in patients with septic shock are associated with more ARDS, increased cumulative fluid balance, and fewer VFD. However, there was no correlation with sequential organ failure assessment score or APACHE II score.

Summary: This study assessed the function and mechanism of RNA binding motif protein 15 (RBM15) silencing in lung cancer development. The results showed that RBM15 silencing promoted ferroptosis in lung cancer cells through the TGF-beta/Smad2 pathway, thereby inhibiting lung cancer cell growth and metastasis.

Summary: AK2 regulates tumor cell metastasis in lung adenocarcinoma, with its high expression associated with poor patient survival; AK2 is closely related to the migration and invasion ability of lung adenocarcinoma cells; AK2 may regulate epithelial-mesenchymal transition through Smad-related signaling pathways.

Summary: Lung cancer (LC) is the leading cause of cancer incidence and mortality worldwide. LC is associated with genetic mutations, environmental factors (smoking), or pathological conditions, and has a poor prognosis and limited pharmacological options. TGF-beta is a molecule that regulates biological processes in the lungs and its alteration is linked to LC development and metastasis. Studies have shown that TGF-beta overexpression could serve as a predictive marker for LC prognosis, and inhibition of TGF-beta can prevent metastasis and improve patient survival. Targeting TGF-beta may be a valid strategy to fight against LC and provide novel and effective treatment options for this aggressive cancer.

Summary: The study revealed the critical role of PTBP3 in LUAD, with its high expression associated with poor prognosis. PTBP3 mediates TGF-beta-induced EMT and metastasis by activating the Smad pathway for transcription. This research provides a new potential therapeutic target for the treatment of LUAD.

Summary: The interrater reliability of the 2015 PALICC criteria for diagnosing moderate to severe PARDS was substantial, with diagnostic disagreements commonly caused by differences in chest radiograph interpretations. Patients with cardiac disease or chronic respiratory failure were more vulnerable to diagnostic disagreements.

Summary: ALKBH5 plays a key role in regulating TGF-beta/SMAD signaling and controlling TGF-beta-induced EMT in NSCLCs. ALKBH5 expression is significantly reduced in metastatic NSCLC and its overexpression inhibits TGF-beta-induced EMT and invasion of NSCLC cells. ALKBH5 affects the expression and mRNA stability of TGF beta R2, SMAD3, and SMAD6 by erasing m(6)A modification in NSCLC cells, and interacts with m(6)A-binding proteins to further regulate TGF-beta signaling.

Summary: Cortactin (CTTN) is an actin-binding and cytoskeletal protein that plays a crucial role in regulating cellular processes, particularly in vascular endothelial cells. It undergoes post-translational modifications and interacts with other proteins to alter cytoskeletal structures and signaling mechanisms. Its function is important for maintaining barrier integrity and inhibiting vascular permeability in vascular endothelial cells. It also affects the pathophysiology of various lung disorders.

Summary: Acute kidney injury has been shown to impact lung metabolism and lead to lung injury, with alterations in mitochondrial function and mtDAMPs correlating with these metabolic changes. Mitochondrial function and mtDAMPs are potential therapeutic targets in preventing lung injury due to AKI.

Summary: TGF-beta plays essential roles in early embryonic development, organogenesis, immune regulation, tissue repair, and adult homeostasis. Its involvement in fibrosis and cancer is complex and context-dependent, showing both inhibitory and promoting effects. Therapeutics targeting TGF-beta signaling hold promise in treating fibrosis and cancer, although the development is hindered by potential systemic cytotoxicity.
