Hepatitis B virus hijacks CTHRC1 to evade host immunity and maintain replication

HepatitisBvirus (HBV) infection causes acuteand chronic liver diseases, but is not directly cytopathic. Liver injury results from repeated attempts of the cellular immune response system to control the viral infection. Here, we investigate the roles of cellular factors and signaling pathways involved in the regulation of HBV replication to reveal the mechanism underlying HBV infection and pathogenesis. We show that collagen triple helix repeat containing 1(CTHRC1) expression is elevated in HBV-infected patients and in HBV-transfected cells through epigenetic modification and transcriptional regulation. CTHRC1 facilitates HBV replication in cultured cells and BALB/c mice by activating the PKC alpha/ERK/JNK/c-Jun cascade to repress the IFN/JAK/STAT pathway. HBV-activated CTHRC1 downregulates the activity of type I interferon(IFN), the production of IFN-stimulatedgenes (ISGs), and the phosphorylation of signal transducer and activator of transcription 1/2 (STAT1/2), whereas it upregulates the phosphorylation and ubiquitination of type I IFN receptors (IFNAR alpha/beta). Thus, our results show that HBV uses a novel mechanism to hijack cellular factors and signal cascades in order to evade host antiviral immunity and maintain persistent infection. We also demonstrate that CTHRC1 has a novel role in viral infection.

Helical Long-Period Fiber Grating-Based OAM Interferometer and Its Application to Fiber Sensing

Summary: Orbital-angular-momentum (OAM) beams-based interferometers have attracted significant interest and found various applications in optical precision measurement. In this study, an all-fiber OAM beams-based interferometer was proposed and experimentally demonstrated, utilizing helical long-period fiber gratings (HLPGs) to generate conjugated OAM beams for testing and referencing. The interferograms obtained had polarization independence, robustness, and were particularly suitable for phase-demodulation in the azimuthal angle domain. The proposed interferometer showed high sensitivity and measurement accuracy for temperature and strain, making it suitable for ultra-high precision and sensing measurements.

Uncertainty and risk of pruned distributional ranges induced by climate shifts for alpine species: a case study for 79 Kobresia species in China

Summary: This study used fuzzy set-based classification, Monte Carlo scheme, and climate shift data to determine the risk of reduction in distributional ranges of alpine graminoid species. The results showed that the main factor leading to range shifts was changes in the temperature index. Adaptation measures are needed to cope with future climatic conditions.

Alterations in gut microbiome and metabolomics in chronic hepatitis B infection-associated liver disease and their impact on peripheral immune response

Effect of solid particle type, concentration and size on DC breakdown voltage of short needle-plate air gap: An experiment and simulation study

It is found that the presence of solid particles increases the breakdown voltage of the gap, with the increase being related to the particle concentration, size, and dielectric constant. Additionally, the presence of particles enhances both the ionization process and particle adsorption, leading to an increase in the gap breakdown voltage.

The contribution of wetland plant litter to soil carbon pool: Decomposition rates and priming effects

Summary: Plant litter input is a crucial factor in the turnover of soil/sediment organic carbon (SOC). This study investigated the effects of leaf litter and stem litter input on SOC dynamics using the 13C isotope technique. It was found that both leaf and stem litter input facilitated SOC accumulation, with leaf litter having a higher contribution. However, leaf litter input also promoted SOC mineralization more than stem litter input. Overall, litter input led to a net increase in SOC accumulation but accelerated the loss of native SOC.

Summary: Urban rivers are significant hotspots for greenhouse gas (GHG) emissions. This study investigated the impact of different rainfall events on GHG emissions from Suzhou Creek in Shanghai. It was found that typhoon events significantly increased GHG emissions, with factors such as rainfall dilution, pump station drainage, sediment disturbance, and high gas transfer coefficient playing important roles. The study highlights the importance of understanding the dynamic response of urban river GHG emissions to rainfall and typhoons.

Recognition of the orbital-angular-momentum spectrum for hybrid modes existing in a few-mode fiber via a deep learning method

Summary: In this study, it is demonstrated theoretically and experimentally that the convolutional neural network (CNN) combined with residual blocks and regression methods can accurately and quickly reconstruct the OAM spectrum of hybrid OAM modes, regardless of the consistency of the order indices in both azimuthal and radial directions. The mean errors of recognized parameters for hybrid OAM modes in a 4-mode fiber (4MF) and a 6-mode fiber (6MF) are smaller than 0.003 and 0.008, respectively. This is the first time that all OAM modes in 4MFs or 6MFs can be accurately and quickly recognized using the deep learning method.

Flame retardant and anti-dripping polyethylene terephthalate fabric based on bio-based phytic acid/gelatine coating

Summary: This study used bio-based phytic acid (PA) and gelatine (G) to improve the flame retardancy and anti-dripping properties of polyethylene terephthalate (PET) fabric. The coated PET fabric showed a limiting oxygen index (LOI) value greater than 29.5% and no dripping phenomenon, indicating significant improvement in flame retardancy and anti-dripping properties. The PA/G coating altered the decomposition stability and degradation process of PET fabric, promoting the formation of a stable char. The coating formed an intumescent flame retardant (IFR) system on PET fabric, functioning through the IFR mechanism.

Zika virus non-structural protein 4B interacts with DHCR7 to facilitate viral infection

Summary: In this study, it was found that Zika virus infection upregulated 7-dehydrocholesterol reductase (DHCR7) expression, which in turn facilitated viral infection. The nonstructural protein NS4B of Zika virus interacted with DHCR7 to induce its expression, thereby inhibiting the immune response and facilitating viral evasion. This study expands our understanding of the mechanisms by which viral nonstructural proteins antagonize innate immunity during viral infection via cholesterol metabolic enzymes and intermediates.

Gaussian Process Latent Variable Model-Based Multi-Output Modeling of Incomplete Data

However, missing values in the data often result in information loss and affect the modeling performance. This study proposes a new multi-output modeling method based on GPLVM to exploit useful information in the data and improve modeling accuracy. The method demonstrates superior performance in both simulation and case studies, making it a powerful tool for process automation.

Succinate as a signaling molecule in the mediation of liver diseases

Summary: Succinate, an intermediate of the TCA cycle, is crucial in mitochondrial metabolism, energy production, and is also considered a signaling molecule in metabolism and hepatic diseases. Its downstream signaling pathway through GPR91 leads to various intracellular responses, including succinylation, ROS production, HIF-1 alpha stabilization, and significant impact in cellular metabolism due to its role in the TCA cycle. Understanding the signaling mechanisms of succinate in hepatic fibrosis, metabolic reprogramming, inflammatory or immune responses, and carcinogenesis is of great interest. This review aims to provide the current understanding of succinate in mediating metabolism, inflammatory and immune responses in liver diseases, with the aim of establishing a molecular basis for therapeutic strategies.
