Summary: Syne2b/nesprin-2 is a giant protein important for tethering the nucleus to the cytoskeleton and maintaining cellular architecture. Disruption of Syne2b function delays epiboly and impairs cytoskeletal organization, leading to disintegration of the epithelial blastoderm during gastrulation.

Summary: CTSK plays a critical role in maintaining the ECM homeostasis and actin cytoskeleton-mediated contractile properties in the TM outflow pathway, affecting the regulation of intraocular pressure. Loss of CTSK activity results in elevated IOP, while constitutive expression of CTSK has an anti-fibrotic effect.

Summary: The actin cytoskeleton contributes to various cellular processes and actin-depolymerizing factor (ADF) is an important regulator of actin filament dynamics. This study investigates the role of HbADF4 in regulating actin dynamics in latex flow in rubber tree. The downregulation of HbADF4 resulted in an increase in the F/G-actin ratio in latex, while upregulation caused a decrease in the ratio. HbADF4 is also associated with stress and hormone responses in rubber tree and plays a role in cytokinesis and cell elongation.

Summary: Filamin A (FLNA) plays a crucial role in cell motility and vascular development by stabilizing actin networks and interacting with transcription factors and cell signaling molecules. Mutations in FLNA can lead to cardiovascular malformations in humans, highlighting its importance in cardiovascular health. The interaction of FLNA with transcription factors and cell signaling molecules dictates its function in vascular cells.

Summary: In this study, we found that the host factor DUSP5 was upregulated after dengue virus infection and demonstrated its role in inhibiting viral replication. DUSP5 restricts viral entry into host cells by negatively regulating the ERK-MLCK-Myosin IIB signaling axis and suppressing F-actin rearrangement. Furthermore, DUSP5 exhibits broad-spectrum antiviral effects against dengue and Zika viruses.

Summary: The study revealed that NDRG1 plays a crucial role in actin reorganization and tumor invasion in colorectal cancer by affecting CDC42 activation. Loss of NDRG1 disrupts the binding between RhoGDI alpha and CDC42, leading to the activation of CDC42 and downstream cascades that promote CRC invasion, and enhances tumor dissemination in vivo.

Summary: The histone demethylase JMJD1C is associated with human platelet counts and plays a role in megakaryopoiesis. The study used various models and samples to investigate the expression and function of JMJD1C. The results suggest that JMJD1C may regulate megakaryopoiesis by influencing the actin network.

Summary: This study reveals the essential role of the protein Daam2 in the regulation of oligodendrocyte cytoskeleton and myelin formation. Daam2 regulates the levels of Gelsolin to modulate the cytoskeleton during oligodendrocyte differentiation, and its deficiency leads to myelin decompaction and motor coordination deficits. These findings highlight the critical role of cytoskeletal dynamics in myelination and provide new potential avenues for the treatment of white matter diseases.

Summary: Actin cytoskeletal reorganization is crucial for regulating smooth muscle contraction, with Abi1 playing a key role in this process through acetylation at K416. The interaction between Abi1 and N-WASP enhances actin polymerization, ultimately promoting smooth muscle contraction. Additionally, the localization of p300 in the cytoplasm of smooth muscle cells suggests a novel mechanism for the acetylation-dependent regulation of the actin cytoskeleton.
