Summary: Development of a vaccine drug product requires formulation optimization to ensure the vaccine's stability and effectiveness. The type of aluminum adjuvant used in the formulation can impact the stability and immunogenicity of the vaccine. In this study, a pneumococcal polysaccharide-protein conjugate vaccine formulated with amorphous aluminum hydroxyphosphate sulfate adjuvant (AAHS) showed reduced immunogenicity and stability compared to the same vaccine formulated with aluminum phosphate adjuvant (AP). The degradation of the polysaccharide antigen caused by AAHS was identified as the main factor contributing to the instability of the vaccine.

Summary: In a randomized study, we showed that both PCV-10 and PPV-23 had lower immunogenicity antepartum than postpartum in women with HIV on antiretrovirals. This indicates that postpartum boosting of vaccines administered during pregnancy may be needed to achieve optimal response.

High-throughput sequencing in this study revealed changes in TCR diversity, V/J gene segment usage, and V-J pairing after rabies vaccination. Identification of altered high-frequency clonotypes provides new insights into T cell receptor status post-vaccination.

Summary: The polysaccharide capsule of Streptococcus pneumoniae plays a crucial role as an intracellular antioxidant within vascular endothelial cells, with different serotypes showing varying resistance to H2O2-mediated killing. This impacts the efficiency of bacterial translocation and the potential for invasive disease. Capsule-mediated protection against intracellular killing is observed in multiple bacterial species, and the differences in antioxidant capabilities between serotypes explain the varying virulence of Spn strains.

Summary: The immunogenicity of a 5-dose pneumococcal vaccination schedule was investigated in adult allo-HSCT recipients with and without immunosuppressive therapy. The study showed that immunosuppressive therapy resulted in a poor response to the vaccine, particularly for the PPSV23 serotypes. Further research on vaccination strategies with higher-valent T-cell-dependent pneumococcal vaccines is needed.
