Novel Aryl Hydrocarbon Receptor Agonist Suppresses Migration and Invasion of Breast Cancer Cells

Background Despite the remarkable progress to fight against breast cancer, metastasis remains the dominant cause of treatment failure and recurrence. Therefore, control of invasiveness potential of breast cancer cells is crucial. Accumulating evidences suggest Aryl hydrocarbon receptor (Ahr), a helix-loop-helix transcription factor, as a promising target to control migration and invasion in breast cancer cells. Thus, an Ahr-based exploration was performed to identify a new Ahr agonist with inhibitory potentials on cancer cell motility. Methods For prediction of potential interactions between Ahr and candidate molecules, bioinformatics analysis was carried out. The interaction of the selected ligand with Ahr and its effects on migration and invasion were examined in vitro using the MDA-MB-231 and T47D cell lines. The silencing RNAs were transfected into cells by electroporation. Expressions of microRNAs (miRNAs) and coding genes were quantified by real-time PCR, and the protein levels were detected by western blot. Results The in silico and in vitro results identified Flavipin as a novel Ahr agonist. It induces formation of Ahr/Ahr nuclear translocator (Arnt) heterodimer to promote the expression of cytochrome P450 family 1 subfamily A member 1 (Cyp1a1). Migration and invasion of MDA-MB-231 and T47D cells were inhibited with Flavipin treatment in an Ahr-dependent fashion. Interestingly, Flavipin suppressed the pro-metastatic factor SRY-related HMG-box4 (Sox4) by inducing miR-212/132 cluster. Moreover, Flavipin inhibited growth and adhesion of both cell lines by suppressing gene expressions of B-cell lymphoma 2 (BcI2) and integrina4 (ITGA4). Conclusion Taken together, the results introduce Flavipin as a novel Ahr agonist, and provide first evidences on its inhibitory effects on cancer cell motility, suggesting Flavipin as a candidate to control cell invasiveness in breast cancer patients.

Summary: This study proposes a new alkali pretreatment method assisted by humic acid to improve the accessibility of enzymes to cellulose. The pretreatment resulted in the recovery of lignin with distinctive properties, and the delignified biomass was used as a solid support for enzyme immobilization. The interaction mechanism between the pretreated bagasse and enzyme was analyzed through molecular docking and UV spectrophotometric analysis.

Atropine Is a Suppressor of Epithelial-Mesenchymal Transition (EMT) That Reduces Stemness in Drug-Resistant Breast Cancer Cells

Summary: Atropine, extracted from belladonna plant, is an anticholinergic drug used to treat cholinergic drug overdose or mushroom poisoning. In silico docking analysis suggests that atropine has strong virtual binding to two major signaling molecules involved in EMT regulation. Both gene and protein expression results indicate that atropine effectively reduces EMT and colony formation in breast tumor cells. Thus, atropine could be a potential suppressor of EMT in drug-resistant breast tumor cells and be used in combination with other chemotherapeutic drugs.

Enterococcus faecium from chicken feces improves chicken immune response and alleviates Salmonella infections: a pilot study

Summary: In this study, beneficial probiotics, specifically Enterococcus faecium, were isolated from broiler chicken carcasses and found to improve chicken health against Salmonella infections. Chickens fed with E. faecium showed improved body weight, reduced mortality, and lower levels of S. enterica. The presence of E. faecium also led to increased levels of short-chain fatty acids and reduced levels of pro-inflammatory markers. These findings highlight the potential of E. faecium as a preventive measure against bacterial infections in broiler chickens.

Probiotic-Fermented Camel Milk Attenuates Neurodegenerative Symptoms via SOX5/miR-218 Axis Orchestration in Mouse Models

Summary: This study investigated the ameliorative effect of Bacillus amyloliquefaciens fermented in camel milk (BEY) on an autoimmune-mediated neurodegenerative model. The results showed that BEY reduced the expression of inflammatory cytokines, increased the expression of neuroprotective markers, and improved the levels of short-chain fatty acids. These findings suggest that BEY could be a promising clinical approach for the treatment of neurodegenerative diseases and promote the use of probiotic food as medicine.

Epigenetic changes have been found to be crucial in the etiology of obesity. Although the use of epigenetic treatments has been discussed in cancer, challenges remain for type 2 diabetes mellitus.
