Effect of Temperature Downshift on the Transcriptomic Responses of Chinese Hamster Ovary Cells Using Recombinant Human Tissue Plasminogen Activator Production Culture

Recombinant proteins are widely used as biopharmaceuticals, but their production by mammalian cell culture is expensive. Hence, improvement of bioprocess productivity is greatly needed. A temperature downshift (TDS) from 37 degrees C to 28-34 degrees C is an effective strategy to expand the productive life period of cells and increase their productivity (q(p)). Here, TDS in Chinese hamster ovary (CHO) cell cultures, initially grown at 37 degrees C and switched to 30 degrees C during the exponential growth phase, resulted in a 1.6-fold increase in the qp of recombinant human tissue plasminogen activator (rh-tPA). The transcriptomic response using next-generation sequencing (NGS) was assessed to characterize the cellular behavior associated with TDS. A total of 416 (q > 0.8) and 3,472 (q > 0.9) differentially expressed transcripts, with more than a 1.6-fold change at 24 and 48 h post TDS, respectively, were observed in cultures with TDS compared to those at constant 37 degrees C. In agreement with the extended cell survival resulting from TDS, transcripts related to cell growth arrest that controlled cell proliferation without the activation of the DNA damage response, were differentially expressed. Most upregulated genes were related to energy metabolism in mitochondria, mitochondrial biogenesis, central metabolism, and avoidance of apoptotic cell death. The gene coding for rh-tPA was not differentially expressed, but fluctuations were detected in the transcripts encoding proteins involved in the secretory machinery, particularly in glycosylation. Through NGS the dynamic processes caused by TDS were assessed in this biological system.

Summary: This study presents the spread of the SARS-CoV-2 Omicron BA.1 subvariant in Mexico, which quickly replaced the Delta variant and caused the fourth COVID-19 surge in the country. The spread of BA.1 began in early December 2021 and became dominant within three weeks. Unlike previous variants, BA.1 did not exhibit a geographically distinct circulation pattern, but there were regional differences in the speed of replacing the Delta variant.

Is heterogeneity in large-scale bioreactors a real problem in recombinant protein synthesis by Pichia pastoris?

Summary: Culture medium heterogeneity in industrial bioreactors leads to concentration gradients due to the loss of mixing efficiency. This study investigated the cell response to transient perturbations of high methanol concentration combined with hypoxia using a scale-down system. The results showed that stressful culture conditions affected cell metabolism, biomass formation, and cell flocculation. Surprisingly, these conditions actually benefited recombinant protein productivity by increasing the expression levels of genes and eGFP.

Overview of fungal terpene synthases and their regulation

Summary: Terpenes and terpenoids are the largest group of natural products and secondary metabolites produced by living things. They are synthesized by enzymes called terpene synthases, which add functional groups to cyclized structures. Understanding the regulation of their synthesis is important for directing their production with desired pharmacological properties. This review covers the detailed study of the biosynthesis of fungal terpenoids and their regulation by various physiological and environmental factors.

Macroscopic model for generalised Newtonian inertial two-phase flow in porous media

Summary: A closed macroscopic model for quasi-steady, inertial, incompressible, two-phase generalised Newtonian flow in rigid and homogeneous porous media is derived by upscaling the pore-scale equations. The derived model includes macroscopic equations for mass and momentum balance as well as an expression for the macroscopic pressure difference between the two fluid phases. The predictions from the upscaled models are in excellent agreement with direct numerical simulations, confirming the validity of the derived macroscopic models.

Multiple approaches of cellular metabolism define the bacterial ancestry of mitochondria

Summary: We breathe by using oxygen consumed by mitochondria in our cells to extract energy. Mitochondria, which derive from aerobic bacteria, play crucial roles in oxidative phosphorylation and other metabolic pathways in eukaryotic cells. The exact bacterial origin of mitochondria and their relationship to the aerobic metabolism of our cells is still controversial despite the abundance of genomic data. In this study, we employ various approaches to identify the most likely bacteria that are the living relatives of the ancestral bacteria from which mitochondria originated. These bacteria inhabit marine environments and possess a high frequency of aerobic traits as well as genes for the metabolism of essential lipids found in eukaryotic membranes, such as sphingolipids and cardiolipin.

Teaching bioreactors operation and scale-up of bioprocesses: multidisciplinary training in a theoretical-practical course of one week

Summary: Biotechnology is an emerging field for developing countries, and bioreactors are crucial for the development and innovation of bioprocesses. An international training course on bioprocess scaling and bioreactor operation has been held to provide theoretical and practical training for the Latin American scientific community. The course has trained over 1400 people through lectures and 216 people with laboratory skills, and has created a network of contacts in consulting and people mobility.
