Summary: Palmitoylation has been found to be relevant to learning, memory, and disorders such as post-traumatic stress disorder and aging-related cognitive decline. However, the mechanisms underlying palmitoylation in these processes are not well understood. This study investigated the role of palmitoylation in hippocampal synaptic transmission and fear memory formation and identified the involvement of APT1-mediated depalmitoylation in the regulation of glutamatergic synaptic plasticity.

Bidirectional plasticity of GABAergic tonic inhibition in hippocampal somatostatin- and parvalbumin-containing interneurons

Summary: In this study, we investigated the effect of NMDA-induced plasticity on tonic inhibition in somatostatin- and parvalbumin-containing interneurons. Our results showed that NMDA application enhanced tonic conductance in somatostatin-containing interneurons, while reducing tonic inhibition in both fast-spiking and non-fast-spiking parvalbumin-positive cells. These changes were associated with alterations in the content of α5 and δ-subunit-containing GABA(A) receptors.

Summary: GCs act through hippocampal mineralocorticoid and glucocorticoid receptors to impact gene regulatory programs related to neuronal plasticity, ciliogenesis, and behavioral adaptation. The receptors MR and GR regulate genes involved in synaptic/neuro-plasticity, cell morphology and development, behavior, and neuropsychiatric disorders. These findings suggest a crucial role of GCs in maintaining brain health and adapting to changing environments.

Gestational 1-nitropyrene exposure causes anxiety-like behavior partially by altering hippocampal epigenetic reprogramming of synaptic plasticity in male adult offspring

Summary: The study found that gestational exposure to 1-NP caused anxiety-like behavior in male adult offspring and impaired dendritic arborization, dendritic length, and dendritic spine density in the ventral hippocampus. Gephyrin-related genes, Nrg1 and Erbb4, were also decreased in 1-NP-exposed fetuses, along with decreased TET activity and α-KG content in the fetal forebrain. Supplementation with α-KG ameliorated the effects of 1-NP, improving synaptic plasticity and anxiety-like behavior in male adult offspring.

Hippocampal expression of the cannabinoid receptor type 1 in canine epilepsy

Summary: Canine drug-resistant epilepsy is a common problem in veterinary neurology. Treatment with cannabinoids shows promising results, and understanding the distribution of cannabinoid receptor type 1 (CB1R) in the epileptic brain can contribute to better disease understanding and application of cannabinoids in dogs with epilepsy. This study found decreased CB1R expression in the CA1 region of the hippocampus in dogs with idiopathic epilepsy, while dogs with structural epilepsy showed increased CB1R signal intensity. The distinct disease-associated CB1R expression should be considered in the development of new treatments for dogs with epilepsy.

Recurring Cholinergic Inputs Induce Local Hippocampal Plasticity through Feedforward Disinhibition

Summary: The CA1 pyramidal neurons in the hippocampus interact with various interneurons in complex local circuits. Recent studies have shown that activating a7 nACh receptors expressed in OLMa2 interneurons can induce long-term potentiation (LTP) in SC-CA1 synapses. According to a biophysically based model, activation of a7 nAChRs increases OLM GABAergic activity, leading to the inhibition of fast-spiking interneurons that provide feedforward inhibition to CA1 pyramidal neurons. This disinhibition, coupled with synchronized SC stimulation, can result in potentiation of excitatory synapses in CA1 pyramidal neurons.

Summary: N-methyl-D-aspartate receptors (NMDARs) are receptors that mediate calcium-permeable excitatory neurotransmission. Genetic variants in the GRIN2A gene are associated with NMDAR diseases, and individuals with null GRIN2A variants experience transient seizure susceptibility during infancy. Studies on mice show increased circuit excitability and delayed electrophysiological maturation of inhibitory interneurons in CA1 in relation to Grin2a gene dosage.
