Lipoxygenases (LOXs) are key enzymes to regulate the production of hormones and defensive metabolites in plants, animals and algae. In this research, a full length LOX gene has been cloned and expressed from the red alga Pyropia haitanensis (Bangiales, Rhodophyta) gametophyte (PhLOX2). Subsequent phylogenetic analysis showed that such LOX enzymes are separated at the early stage of evolution, establishing an independent branch. The LOX activity was investigated at the optimal pH of 8.0. It appears that PhLOX2 is a multifunctional enzyme featuring both lipoxygenase and hydroperoxidase activities. Additionally, PhLOX2 exhibits remarkable substrate and position flexibility, and it can catalyze an array of chemical reactions involving various polyunsaturated fatty acids, ranging from C18 to C22. As a matter of fact, mono-hydroperoxy, di-hydroperoxy and hydroxyl products have been obtained from such transformations, and eicosapentaenoic acid seem to be the most preferred substrate. It was found that at least triple ethylenic bonds are required for PhLOX2 to function as a LOX, and the resulting hydroxy products should be originated from the PhLOX2 mediated reduction of mono-hydroperoxides, in which the hydrogen abstraction occurs on the carbon atom between the second and third double bond. Most of the di-hydroperoxides observed seem to be missing their mono-position precursors. The substrate and position flexibility, as well as the function versatility of PhLOXs represent the ancient enzymatic pathway for organisms to control intracellular oxylipins.

Mixotrophic culture of bait microalgae for biomass and nutrients accumulation and their synergistic carbon metabolism

Yahui Bo, Ruirui Chu, Danni Sun, Xiangyuan Deng, Chengxu Zhou, Xiaojun Yan, Roger Ruan, Pengfei Cheng

Summary: Microalgae cannot fulfill the bait demand for aquaculture due to limitations in light intensity and other unfavorable conditions. This study selected 6 mixotrophic microalgae and screened for the optimal strains and organic carbon sources. Thalassiosira pseudonana and Chlorella sp. were found to be suitable for mixotrophic culture. When glycerol was added, Thalassiosira pseudonana showed a 1.67-fold increase in maximum cell density compared to the photoautotrophic group, while Chlorella sp. exhibited a 1.69-fold increase with acetic acid. Metabolomic analysis revealed that mixotrophic bait microalgae could promote the accumulation of lipids and amino acids.

Huan Jiang, Jiabao Hu, Huihui Xie, Man Zhang, Chunyang Guo, Youyi Zhang, Yaya Li, Cheng Zhang, Shanliang Xu, Danli Wang, Xiaojun Yan, Yajun Wang, Xubo Wang

Summary: In this study, the morphological and histological characteristics of the pharyngeal sac in Pampus argenteus were examined using histochemical techniques and electron microscopy. The pharyngeal sac showed unique characteristics such as well-developed muscular walls, weakly keratinized epithelium, numerous goblet cells, and needle-like processes on the papillae. RNA-seq analysis revealed that phenotype-associated genes were upregulated, while genes related to nutrient digestion and absorption were downregulated in the pharyngeal sac.

Reducing aluminum is the key nutrient management strategy for ameliorating soil acidification and improving root growth in an acidic citrus orchard

Siwen Zhang, Xiaohui Chen, Zongjun Ji, Xiaojun Yan, Kunpeng Kong, Yuanyang Cai, Qichao Zhu, Muhammad Atif Muneer, Fusuo Zhang, Liangquan Wu

Summary: Aluminum (Al) toxicity is a serious threat to root formation and crop growth due to soil acidification. This study focused on the effect of Al3+ on citrus roots in acidified red soil. The results showed that rapid soil acidification occurred in pomelo orchards, and optimized fertilization and liming treatments effectively reduced Al toxicity in roots and increased yield by improving soil pH and base saturation. Structural equation modeling confirmed the importance of increasing soil pH and base saturation while decreasing exchangeable Al for citrus production in acidic soil.

Xinyu Zhao, Xiaolong Yin, Tianzhu Ma, Weihua Song, Lihua Jiang, Xiaolin Zhang, Bingjian Liu, Yifan Liu, Xiaojun Yan

Summary: This study investigated the effects of different concentrations of chloroquine (CQ) on large yellow croakers and found that higher concentrations of CQ led to more obvious toxic effects, resulting in high mortality rates. The study also observed differential expressions of autophagy/inflammation/apoptosis-related genes in representative tissues.

Greasing the wheels of policy reversal: Discursive engineering and public opinion management during the relaxation of China's family planning policy

Xiaojun Yan, La Li

Summary: In this article, we examine how autocratic regimes, in particular China, use political propaganda to facilitate policy reversals of major significance. Through analyzing extensive propaganda materials, we find that the Chinese Communist Party employs a systematic, precise, multi-dimensional, and multi-layered discursive engineering project to reverse its policies effectively. This coordinated maneuver of state narrative is key in achieving the smooth and successful policy reversal.

Mengqi Zhang, Fei Kong, Bin Ma, Deshui Chen, Zhaoshou Ran, Shuonan Ma, Kai Liao, Jiayi Cao, Lin Zhang, Xiaojun Yan, Jilin Xu

Summary: In this study, the effects of seven light treatments on the growth and physiological parameters of juvenile Sinonovacula constricta were investigated. The results showed that yellow light had the highest growth rates, while violet and blue light had the lowest. Yellow light also promoted feeding activity and enzymatic activity related to digestion. Moreover, yellow light exhibited higher antioxidant biomarker activities and red light had higher expression levels of genes related to antioxidation and stress response. Conversely, white light had the lowest malonaldehyde level. Overall, these findings suggest that yellow light is beneficial for the growth of juvenile S. constricta.

The Heat Shock Transcription Factor PtHSF1 Mediates Triacylglycerol and Fucoxanthin Synthesis by Regulating the Expression of GPAT3 and DXS in Phaeodactylum tricornutum

Jianquan Song, Hejing Zhao, Linxin Zhang, Zheng Li, Jichang Han, Chengxu Zhou, Jilin Xu, Xiaohui Li, Xiaojun Yan

Summary: This study revealed that the heat shock transcription factor PtHSF1 positively regulates the synthesis of triacylglycerol and fucoxanthin in Phaeodactylum tricornutum. Overexpression of PtHSF1 increased the contents of triacylglycerol and fucoxanthin and upregulated key enzyme genes involved in their biosynthesis pathways. Gene silencing of PtHSF1 had the opposite effect. This study provided insights into the regulation mechanisms of PtHSF1-mediated triacylglycerol and fucoxanthin synthesis.
