Summary: Phase separation is a process where cells condense related biomolecules into independent compartments for orderly and efficient biological reactions, playing a significant role in cellular processes such as signal transduction control, gene expression regulation, and stress response. In recent years, many phase separation events have been discovered in the immune response process.

Summary: NLRP6 plays a crucial role in host defense by inducing inflammasome activation and interferon production through LLPS upon interaction with dsRNA. The poly-lysine sequence (K350-354) of NLRP6 is essential for multivalent interactions and inflammasome activation. Deficiency or mutation of Nlrp6 leads to impaired immune responses.

Summary: Immunocompromised populations are at high risk of life-threatening infections, and strategies to protect these patients are urgently needed. This study explores the use of trained immunity, which enhances the immune response to subsequent infections, as a promising approach. The researchers demonstrate that the MyD88-dependent signaling pathway plays a critical role in TLR-mediated trained immunity, providing valuable insights into the mechanisms underlying this process.

Summary: Osteoarthritis (OA) is a common condition affecting horses, humans, and companion animals with diverse etiologies. It is not a single disease, but rather a disease process initiated by different events, including acute trauma, irregular or repetitive overload of articular structures, and spontaneous development with aging. OA is increasingly considered a multifactorial disease in which the innate immune system plays a key role in regulating and perpetuating low-grade inflammation, resulting in sustained cartilage injury and destruction.

Summary: Ablation of RGS12 in hematopoietic cells was shown to prevent bone loss in a periodontitis model, inhibit osteoclast formation and activity, and reduce inflammatory cytokine production in periodontal tissue. In vitro experiments also demonstrated that overexpression of RGS12 promoted proinflammatory macrophage reprogramming and enhanced migration ability, while knockdown of RGS12 inhibited inflammatory cytokine production and macrophage migration in response to stimuli like lipopolysaccharide. The results suggest that targeting RGS12 in macrophages could be a promising therapeutic approach for periodontitis.

Summary: The numbers of DUOX2(+)ACE2(+) small cholangiocytes are inversely associated with disease severity in PBC. They may be the target of pIgR-mediated humoral responses. Protecting these cells and targeting anti-pIgR autoantibodies could be valuable strategies for therapeutic interventions in PBC.

Summary: Liver cirrhosis is associated with impaired phagocytic function of macrophages and an immunosuppressive transcriptome profile. Overexpression of PD-L1 in monocytes correlates with disease severity and increased infection risk. Blocking PD-L1 reverses immune suppression and enhances antimicrobial activity of liver macrophages in cirrhosis.

Summary: This research identifies a novel mechanism by which early-life exposure to Acinetobacter lwoffii (AL) can prevent chronic inflammatory diseases such as asthma. The study found that AL triggers a proinflammatory immune response characterized by elevated levels of IL-6 and subsequent IL-10 production in CD4(+) T-cells. This immune response, along with changes in the gastrointestinal microbiome, contribute to the protection against asthma.

Summary: Afatinib can suppress CD8(+)T lymphocyte proliferation by targeting CAD, and its transient immunomodulatory effect may enhance therapeutic efficacy when combined with anti-PD1 immunotherapy. Furthermore, the timing window for combined therapy with afatinib and immunotherapy may prevent the dampening of ICB efficacy by EGFR-TKIs.

Summary: HLJ1 knockout mice show reduced organ injury and mortality in lipopolysaccharide-induced endotoxic shock. HLJ1 deletion affects IFN-gamma-related gene signatures in distinct immune cell clusters. HLJ1 deficiency leads to reduced IL-12 levels, dampened IFN-gamma production in natural killer cells, and improved survival rate. HLJ1 converts misfolded IL-12p35 homodimers to monomers, maintaining bioactive IL-12p70 heterodimerization and secretion.

Summary: Primary biliary cholangitis (PBC) is a liver autoimmune disease characterized by immune-mediated destruction of small bile ducts. In this study, the early events in a murine model of autoimmune cholangitis were examined. It was found that apoptosis of biliary epithelial cells (BECs) and alterations in gut microbiota, particularly an increase in gram-positive Firmicutes, occurred in the early stage. Furthermore, BECs expressed molecules and cytokines associated with inflammation and TLR2 signaling was involved in BEC apoptosis. These findings provide a new mechanism for the breakdown of tolerance in autoimmune cholangitis.
