The Scavenger Receptor MARCO Modulates TLR-Induced Responses in Dendritic Cells

The scavenger receptor MARCO mediates macrophage recognition and clearance of pathogens and their polyanionic ligands. However, recent studies demonstrate MARCO expression and function in dendritic cells, suggesting MARCO might serve to bridge innate and adaptive immunity. To gain additional insight into the role of MARCO in dendritic cell activation and function, we profiled transcriptomes of mouse splenic dendritic cells obtained from MARCO deficient mice and their wild type counterparts under resting and activating conditions. In silico analysis uncovered major alterations in gene expression in MARCO deficient dendritic cells resulting in dramatic alterations in key dendritic cell-specific pathways and functions. Specifically, changes in CD209, FCGR4 and Complement factors can have major consequences on DC-mediated innate responses. Notably, these perturbations were magnified following activation with the TLR-4 agonist lipopolysaccharide. To validate our in silico data, we challenged DC's with various agonists that recognize all mouse TLRs and assessed expression of a set of immune and inflammatory marker genes. This approach identified a differential contribution of MARCO to TLR activation and validated a major role for MARCO in mounting an inflammatory response. Together, our data demonstrate that MARCO differentially affects TLR-induced DC activation and suggest targeting of MARCO could lead to different outcomes that depend on the inflammatory context encountered by DC.

Defining the Molecular Hallmarks of T-Cell Memory

Summary: The pool of memory CD8 T cells consists of highly specialized subpopulations of cells with both shared and distinct functions. Current research on T-cell memory focuses on how these subpopulations arise, how the cells are maintained over the host's lifespan, and how they protect the host against reinfection. However, the use of surface markers to define and study these memory T-cell subsets is proving to be inadequate, and there is a need for an updated definition using the epigenetic state of cells to better characterize the functional capacity of CD8 memory T-cell subsets.

Summary: By adding α GC to a tumor cell vaccine, the therapeutic effect against prostate cancer can be enhanced. In a therapeutic model, mice treated with the combination vaccine showed prolonged survival and tumor inhibition. Mechanistic studies revealed that GM-CSF and iNKT cells in the vaccine played a crucial role in anti-tumor immune responses.

New epigenetic regulators of T cell exhaustion

Summary: In this article, Belk et al. conducted an in vitro CRISPR screen to identify genes that regulate CD8(+) T cell exhaustion. They discovered several genes related to epigenetic modification and demonstrated that eliminating Arid1a enables CD8(+) T cells to retain their proliferative and cytotoxic function in vivo, resulting in improved anti-tumor activity.

Summary: Improvements in tumor immunotherapies rely on understanding the T cell response to tumors. By studying tumor-draining lymph nodes (TDLNs), it was discovered that activated CD8+ T cells in TDLNs shared characteristics with stem-like cells in tumors. These TDLN cells were revealed to be precursors of tumor-resident stem-like CD8+ T cells. Murine tumor models showed that tumor-specific CD8+ T cells were activated in TDLNs and later differentiated into effector cells in the tumor with co-stimulation.

Longitudinal Analysis of the Phenotype, Transcriptional Profile, and Anatomic Location of Memory CD8 T Cell Subsets after Acute Viral Infection

Summary: A better understanding of long-lived memory CD8 T cell differentiation is necessary due to increased demand for novel vaccination strategies. Using a mouse model of acute LCMV infection, we conducted an in-depth, longitudinal analysis of memory CD8 T cell subsets, examining their phenotype, transcriptional programming, and anatomical location in the spleen. We characterized a comprehensive memory phenotype associated with higher CD28 expression and found distinct localization programs within the spleen for memory subsets. These findings suggest that memory formation and survival may be influenced by location, emphasizing the importance of considering memory phenotypes and their trafficking for robust memory responses following vaccination.
