Interleukin 7 Plays a Role in T Lymphocyte Apoptosis Inhibition Driven by Mesenchymal Stem Cell without Favoring Proliferation and Cytokines Secretion

Since 2004, when a case report describing the use of human mesenchymal stem cells (hMSCs) infusion as a therapy for GVHD after bone marrow transplantation, a new perspective in MSC function emerged. Since then hMSCs immunomodulatory potential became the target of several studies. Although great progress has been made in our understanding of hMSCs, their effect on T cell remains obscure. Our study has confirmed the already described effect of hMSCs on lymphocytes proliferation and survival. We also show that the impairment of lymphocyte proliferation and apoptosis is contact-independent and occurs in a prostaglandin-independent manner. A potential correlation between IL-7 and hMSCs effect is suggested, as we observed an increase in IL-7 receptors (CD127) on lymphocyte membrane in MSC presence. Additionally, blocking IL-7 in hMSCs-lymphocytes co-cultures increased lymphocytes apoptosis and we also have demonstrated that hMSCs are able to produce this interleukin. Moreover, we found that during Th1/Th17 differentiation in vitro, hMSCs presence leads to Th1/Th17 cells with reduced capacity of INF-y and IL-17 secretion respectively, regardless of having several pro-inflammatory cytokines in culture. We did not confirm an increment of Treg in these cultures, but a reduced percentage of INF-y/IL-17 secreting cells was observed, suggesting that the ratio between anti and pro-inflammatory cells changed. This changed ratio is very important to GvHD therapy and links hMSCs to an anti-inflammatory role. Taken together, our findings provide important preliminary results on the lymphocyte pathway modulated by MSCs and may contribute for developing novel treatments and therapeutic targets for GvHD and others autoimmune diseases.

Need for hemodialysis in patients undergoing hematopoietic stem cell transplantation: risk factors and survival in a retrospective cohort

Summary: This study analyzed the survival and risk factors associated with hemodialysis in hematopoietic stem cell transplantation patients. The results showed that hemodialysis was significantly associated with male gender, age, grade II-IV acute GVHD, various drug exposures, microangiopathy, septic shock, and steroid exposure. The HD group had a higher mortality rate.

Optimization of Multimodal Nanoparticles Internalization Process in Mesenchymal Stem Cells for Cell Therapy Studies

Summary: This study aims to optimize the labeling conditions for human bone marrow mesenchymal stem cells (hBM-MSC) using multifunctional nanoparticles (MFNP) in order to monitor these cells in vivo. The results show that the use of a magnetic field and higher concentration of MFNP during cell labeling can significantly increase the iron load and improve the detection of cells by near-infrared fluorescence (NIRF) technique without compromising cellular viability.

Preclinical Evaluation of the Tumorigenic and Immunomodulatory Properties of Human Bone Marrow Mesenchymal Stromal Cell Populations with Clonal Trisomy 5

Summary: Cytogenetic aberrations, such as trisomy 5, may occur in human mesenchymal stromal cells (MSC) during ex vivo expansion. It is still unclear how these aberrations affect the neoplastic transformation threshold or medicinal properties of MSC products. In this study, MSC cultures with clonal trisomy 5 showed reduced cell proliferation, increased senescence-associated beta-galactosidase activity, and poor tumor initiating capability. However, subcutaneous injection of these MSC populations did not generate tumors in immunodeficient mice within a six-month follow-up period. Importantly, MSC populations with clonal trisomy 5 maintained their immunomodulatory properties similar to euploid MSC upon interferon gamma (IFN gamma) licensing. These findings suggest that bone marrow MSC products with clonal trisomy 5 may still retain their therapeutic potential and support the definition of release criteria for autologous MSC products in critical clinical scenarios.

Summary: This study analyzed the karyotype test and myeloid panel with next-generation sequencing findings in patients with myelofibrosis, and compared transplant characteristics in patients referred for bone marrow transplantation. The results showed that patients with high-risk molecular mutations were more suitable for bone marrow transplant. Molecular analysis is an important tool for guiding therapy in myelofibrosis.

Bioluminescence Imaging and ICP-MS Associated with SPION as a Tool for Hematopoietic Stem and Progenitor Cells Homing and Engraftment Evaluation

Summary: Bone marrow transplantation is a treatment for various diseases, and this study proposes a new method to evaluate the engraftment and behavior of hematopoietic stem cells. The method involves the use of bioluminescence imaging and inductively coupled plasma mass spectrometry associated with superparamagnetic iron oxide nanoparticles. The study found that the administration of Fluorouracil enriched the population of hematopoietic stem cells in the bone marrow, and labeling the cells with nanoparticles provided information on their internalization status. The study also utilized blood count and bioluminescence imaging to monitor the reconstitution of hematopoiesis and evaluate the transplantation effectiveness.

Immune thrombocytopenia associated with lymph node tuberculosis: a case report

Summary: Extrapulmonary tuberculosis is rare with immune thrombocytopenia (ITP). A case of ITP associated with pulmonary and lymph node tuberculosis in a 29-year-old male patient was reported. After tuberculosis treatment, the platelet count effectively normalized, suggesting an association between tuberculosis and ITP.

Summary: This study analyzed the impact of anti-T-cell globulin (ATG) in HLA-matched and unrelated donor peripheral blood stem cell transplantation (PBSC HCT). The results showed that ATG can reduce the occurrence of chronic graft-versus-host disease (GVHD) and improve the quality of life of patients, but it may increase the relapse rate. Therefore, ATG should be considered as a standard treatment in this population.
