Structural Characterization of a Gcn5-Related N-Acetyltransferase from Staphylococcus aureus

The Gcn5-related N-acetyltransferases (GNATs) are ubiquitously expressed in nature and perform a diverse range of cellular functions through the acetylation of small molecules and protein substrates. Using activated acetyl coenzyme A as a common acetyl donor, GNATs catalyse the transfer of an acetyl group to acceptor molecules including aminoglycoside antibiotics, glucosamine-6-phosphate, histones, serotonin and spermidine. There is often only very limited sequence conservation between members of the GNAT superfamily, in part, reflecting their capacity to bind a diverse array of substrates. In contrast, the secondary and tertiary structures are highly conserved, but then at the quaternary level there is further diversity, with GNATs shown to exist in monomeric, dimeric, or tetrameric states. Here we describe the X-ray crystallographic structure of a GNAT enzyme from Staphyloccocus aureus with only low sequence identity to previously solved GNAT proteins. It contains many of the classical GNAT motifs, but lacks other hallmarks of the GNAT fold including the classic beta-bulge splayed at the beta-sheet interface. The protein is likely to be a dimer in solution based on analysis of the asymmetric unit within the crystal structure, homology with related GNAT family members, and size exclusion chromatography. The study provides the first high resolution structure of this enzyme, providing a strong platform for substrate and cofactor modelling, and structural/functional comparisons within this diverse enzyme superfamily.

Importin ?/?-dependent nuclear transport of human parvovirus B19 nonstructural protein 1 is essential for viral replication

Summary: Human parvovirus B19 (B19V) is a major pathogen that primarily infects human progenitor cells in bone marrow. The non-structural protein NS1 plays a crucial role in viral replication and host gene expression. This study identified a specific sequence of amino acids as the nuclear localization signal responsible for NS1 nuclear import, which was found to be dependent on importin alpha/beta and energy. Ivermectin, an antiparasitic drug that interferes with importin alpha/beta-dependent nuclear import, inhibited NS1 nuclear accumulation and viral replication.

Summary: Acot enzymes play important roles in cell signaling, lipid metabolism, inflammation, and ion channel opening. Acot7 can spontaneously form amyloid fibrils under physiological conditions. The aggregation mechanism of Acot7 involves conformational rearrangement and the formation of enhanced beta-sheet structures in the initial phase, followed by the formation of amyloid fibrils.

Lesions and viral loads in racing pigeons naturally coinfected with pigeon circovirus and columbid alphaherpesvirus 1 in Australia

Summary: Coinfection of CoHV1 and PiCV in racing rock pigeons caused oral and upper respiratory tract lesions, encephalitis, and fatal systemic disease. Both viruses were found in high viral load in liver, oropharynx, and bursa of Fabricius. The lesions induced by CoHV1 may have been exacerbated by concomitant PiCV infection.

The first genomic insight into Chlamydia psittaci sequence type (ST)24 from a healthy captive psittacine host in Australia demonstrates evolutionary proximity with strains from psittacine, human, and equine hosts

Summary: This article reports the complete genome sequence of Chlamydia psittaci strain BF_amazon_parrot13, which was detected in the feces of a healthy captive parrot in Australia. Analysis revealed that BF_amazon_parrot13 belongs to the ST24 lineage and has an ompA genotype A. Genomic comparison showed that BF_amazon_parrot13 clusters with seven C. psittaci strains from equines in Australia. This study is important for understanding the health risks and transmission routes of chlamydia among humans and animals.

Explainable learning analytics to identify disengaged students early in semester: an intervention supporting widening participation

Summary: The aim of this study was to use explainable learning analytics to identify and support commencing undergraduate students at an Australian regional university. Non-submission of an early assessment item accurately identified disengaged students, and those students who had successful dialogue with the Outreach Team were less likely to receive a failing grade. Analysis of intersectionality revealed that student progress rate decreased with additional equity factors. A holistic conversation with the Outreach Team increased retention for equity students the following semester, indicating that explainable learning analytics can be used to support equity students.

Liver virome of a Little Corella (Cacatua sanguinea) reveals coinfection with a novel parvovirus and two beak and feather disease viruses

Summary: This study identified a novel psittaciform chaphamaparvovirus 3 (PsChV-3) in an Australian free-ranging little corella, as well as two complete novel beak and feather disease (BFDV) genomes. Phylogenetic analysis showed that the sequenced novel BFDV genomes clustered with other BFDVs isolated from Australian cockatoos. This study contributes to the understanding of chaphamaparvoviruses and BFDV in Australian parrots, highlighting the need for ongoing monitoring and molecular studies.

Characterisation of a novel aviadenovirus associated with disease in tawny frogmouths (Podargus strigoides)

The identified complete genome of this aviadenovirus, named tawny frogmouth aviadenovirus A (TwAviAdV-A), exhibits common genomic organization with a notable monophyletic subclade. The virus is hepatotrophic and the infected frogmouths commonly exhibit regurgitation.

Embedding equity: online tutor support to provide effective feedforward on assessments

Summary: There is a high demand for sustainable and innovative programs that support students in transitioning to university and improve their progress rates and retention. An institution-wide model for an online Embedded Tutor Program was established to meet this demand. The program provided support to first-year undergraduate students and proved to be effective post-pandemic. Tutors with unit-specific knowledge were assigned to 98 first-year courses, where they met with students via Zoom to provide feedback on written assessments. The program had positive outcomes, including higher assessment marks and cumulative unit marks, as well as improved confidence, overall learning, and satisfaction among students. Students with multiple equity factors benefited the most from this support.

A novel pathogenic avipoxvirus infecting oriental turtle dove (Streptopelia orientalis) in China shows a high genomic and evolutionary proximity with the pigeon avipoxviruses isolated globally

Summary: This study presents the first whole genome sequence of a novel pathogenic avipoxvirus (turtle dovepox virus, TDPV) isolated from a wild oriental turtle dove. Phylogenetic analysis showed that TDPV is closely related to avipoxviruses isolated from pigeon in South Africa and India. These findings contribute to our understanding of the genetic evolution of avipoxviruses and enrich the genomic information of the genus Avipoxvirus.
