Summary: This study reveals the relationship between Piezo1-mediated Ca2+ influx and cellular behavior using innovative biosensors and imaging analysis. The results demonstrate that Piezo1 induces extracellular Ca2+ influx, which requires the support of caveolin, cholesterol, and the cytoskeleton. Increased cytoplasmic Ca2+ levels enhance the activity of PKA, ERK, Rac1, and ROCK, promoting cancer cell survival and migration. Furthermore, Piezo1-mediated Ca2+ influx upregulates membrane ruffling, a characteristic feature of cancer cell metastasis.

Summary: Mutations in CLN3 lead to photoreceptor cell loss in CLN3 disease, affecting the phagocytosis of photoreceptor outer segments by retinal pigment epithelium (RPE) cells. Patient-derived cells show that CLN3 is essential for RPE cell structure and phagocytosis, suggesting potential for gene therapy targeting RPE cells in treating CLN3 disease.

Summary: Using ultrasounds, the extraction time for membrane-assisted solvent extraction (MASE) in isolating synthetic cathinones from urine has been shortened to 30 minutes. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used for the separation and determination of eleven synthetic cathinones. The developed method showed efficient extraction and pre-concentration of synthetic cathinones with LogP values higher than 2.0.

Summary: Compared to globular proteins, the folding and insertion of beta-barrel membrane proteins are slow but rapid, occurring within seconds upon arrival at the membrane interface. This combination of infrequent yet fast folding events resolves the apparent contradiction between slow ensemble kinetics and typical biomolecular folding timescales.

The method showed good sensitivity and detection limits, and can be applied to the sensitive detection of biogenic amines in animal-derived foodstuff.

Summary: Scientific research on heart rate variability (HRV) biofeedback faces methodological challenges, including lack of consistent training quality assessment and control conditions. In this study, a novel sham HRV-biofeedback training was proposed as a credible control condition. The use of a training fidelity criterion based on the Yield Efficiency of Training Index (YETI) significantly improved the analysis of training effects and sample homogeneity. This methodological approach has the potential to benefit other behavioral training procedures involving psychophysiological mechanisms.
