Proteins of the IQGAP family display complicated and often contradictory activities in tumorigenesis. IQGAP1 has well documented oncogenic potential and IQGAP2 has putative tumor-suppressive function. IQGAP3 is the latest addition to this family and its role in cancer development remains to be defined. Here we demonstrate IQGAP3 expression is markedly increased in lung cancer tissues at both mRNA and protein levels. Overexpression of IQGAP3 promoted tumor cell growth, and migration and invasion, whereas knockdown of IQGAP3 exhibited opposite effects. Moreover, suppression of IQGAP3 in a lung cancer cell line caused a reduction in the tumorigenicity of these cells in lung tissue after intravenous injection. Furthermore, we showed that IQGAP3 is able to interact with ERK1 and enhance its phosphorylation following treatment with EGF. These data suggest that IQGAP3 may contribute to the pathogenesis of lung cancer by modulating EGFR-ERK signaling.

The deubiquitinase OTUD1 enhances iron transport and potentiates host antitumor immunity

Summary: This study identifies OTDU1 as a crucial regulator in iron transportation in cancer, promoting the immune response against tumors. OTUD1 depletion limits tumor-reactive T-cell accumulation and exacerbates cancer progression. The data highlights the significance of OTUD1-IREB2-TFRC signaling axis in host antitumor immunity.

Summary: The study in Japanese patients found that the body roundness index was positively linearly associated with an elevated risk of incident type 2 diabetes, especially in women. In men, the relationship was J-shaped. BRI proved to be an effective predictor of type 2 diabetes, with higher discriminatory power compared to BMI and WC in both sexes.

Persistent increase of carbohydrate antigen 19-9 with an unknown reason: A seven-year follow-up case

Wei Zhao, Lei Duan, Long Fang, Jinghua Li, Shirui Li, Lina Wang, Jingying Zhang, Wan Zhang, Yongtong Cao

Summary: This study reported a case of a patient with gradually increased CA19-9 concentrations over 4 years, but without any clinical manifestations. Through multiple system detection and other tests, it was found that the increase in CA19-9 concentration in the patient's blood sample was due to the presence of heterophilic antibodies. It is recommended to evaluate heterophilic antibodies in cases of elevated CA19-9 levels without associated clinical manifestations to prevent false positive results.

EDTA plasma in glass and polyethylene terephthalate tubes is suitable for measurement of B-type natriuretic peptide on the Mindray CL-6000i

Wei Zhao, Gang Feng, Jun Wan, Xing Wang, Jiping Feng, Wanwan Zhang, Yalan Yu, Rui Pu, Yong Du, Yongtong Cao

Summary: This study aimed to investigate differences in B-type natriuretic peptide (BNP) concentration and stability in different collection tubes, and to assess BNP accuracy. The results showed that BNP concentrations in EDTA/glass tubes were 31.4% higher than those in heparin/glass tubes, and 3.04% lower than those in EDTA/PET tubes. BNP stability significantly decreased in the heparin/glass tube, while it remained stable in EDTA/glass and EDTA/PET tubes at room temperature for 4 hours. The recovery rates of BNP in heparin/glass, EDTA/glass, and EDTA/PET tubes were 77.46%, 86.04%, and 88.23%, respectively. In conclusion, plasma in EDTA/glass and EDTA/PET tubes is suitable for BNP measurement on the Mindray CL-6000i.
