We report herein the G-quadruplex-selective property of a luminescent cyclometallated iridium(III) complex for the detection of adenosine-5'-triphosphate (ATP) in aqueous solution. The ATP-binding aptamer was employed as the ATP recognition unit, while the iridium(III) complex was used to monitor the formation of the G-quadruplex structure induced by ATP. The sensitivity and fold enhancement of the assay were higher than those of the previously reported assay using the organic dye crystal violet as a fluorescent probe. This label-free luminescent switch-on assay exhibits high sensitivity and selectivity towards ATP with a limit of detection of 2.5 mu M.

Use of NAD tagSeq II to identify growth phase-dependent alterations in E. coli RNA NAD+ capping

Hailei Zhang, Huan Zhong, Xufeng Wang, Shoudong Zhang, Xiaojian Shao, Hao Hu, Zhiling Yu, Zongwei Cai, Xuemei Chen, Yiji Xia

Summary: New findings suggest that NAD-RNAs are involved in regulating gene expression in prokaryotes and eukaryotes through noncanonical RNA capping. The development of NAD tagSeq II using copper-free SPAAC provides a more accurate method for labeling NAD-RNAs and analyzing their profiles, revealing genome-wide alterations in E. coli during different growth phases.

Combination of Wogonin and Artesunate Exhibits Synergistic anti-Hepatocellular Carcinoma Effect by Increasing DNA-Damage-Inducible Alpha, Tumor Necrosis Factor α and Tumor Necrosis Factor Receptor-Associated Factor 3-mediated Apoptosis

Minting Chen, Hsin Ling Wu, Tsz Sin Wong, Baisen Chen, Rui-Hong Gong, Hoi Leong Xavier Wong, Haitao Xiao, Zhaoxiang Bian, Hiu Yee Kwan

Summary: The combination treatment of wogonin and artesunate shows synergistic anti-HCC effect by increasing apoptosis and reducing cell viability. The treatment significantly increases the expression of GADD45A, TNF alpha, and TRAF3, leading to enhanced apoptosis and reduced HCC cell viability. The study supports further development of this combination therapy for HCC management.

Summary: The study identified the master regulator TLR4 in CRC growth under high-fat diet, showing that palmitic acid and TLR4 play key roles in promoting cancer growth. Furthermore, palmitic acid regulates TLR4 expression in a PU.1-dependent manner, and TLR4 controls cancer metabolism to enhance CRC growth under HFD conditions.

Dihydroartemisinin is potential therapeutics for treating late-stage CRC by targeting the elevated c-Myc level

Xianjing Hu, Sarwat Fatima, Minting Chen, Tao Huang, Yuen Wa Chen, Ruihong Gong, Hoi Leong Xavier Wong, Rongmin Yu, Liyan Song, Hiu Yee Kwan, Zhaoxiang Bian

Summary: The study demonstrates that high levels of c-Myc protein in late-stage CRC can be effectively targeted by dihydroartemisinin treatment, resulting in reduced cell viability and altered metabolic phenotypes. Dihydroartemisinin shows stronger growth-inhibitory effects in late-stage CRC compared to early-stage CRC, indicating its potential as a therapeutic option for late-stage CRC. Additionally, the treatment reduces c-Myc expression, leading to downregulation of key metabolic enzymes and suppression of tumor cell proliferation.

Dehydroeburicoic Acid, a Dual Inhibitor against Oxidative Stress in Alcoholic Liver Disease

Shasha Cheng, Yi Kuang, Guodong Li, Jia Wu, Chung-Nga Ko, Wanhe Wang, Dik-Lung Ma, Min Ye, Chung-Hang Leung

Summary: This study reports the use of dehydroeburicoic acid (DEA) as a dual inhibitor of the Keap1-Nrf2 protein-protein interaction (PPI) and GSK3 beta, providing potential therapeutic strategies for alcoholic liver disease (ALD). DEA disrupts the Keap1-Nrf2 PPI and restores Nrf2 activity, leading to the activation of antioxidant genes. It also alleviates ethanol-induced mitochondrial dysfunction and exhibits minimal toxicity in the ALD cell model.
