Summary: In this study, strong electrochemiluminescence (ECL) emission was achieved using a G-quadruplex selective iridium (III) complex as an efficient ECL signal probe. By introducing the iridium (III) complex through its specific interaction with G-quadruplex DNA modified on the surface of negatively charged gold nanoparticles ((- )AuNPs), an increased ECL signal was induced, which allowed for quantitative detection of cTnI in a range of 5.0 fg/mL100 ng/mL with a detection limit of 1.67 fg/mL. The proposed immunosensor showed great potential in the early diagnosis of acute myocardial infarction (AMI) by successfully detecting cTnI in human serums from healthy individuals and AMI patients.

Summary: A phosphorescent probe Ir-PDP containing an Ir(III) complex was designed and synthesized to evaluate the level of G4 in the cytoplasm. Compared with other nucleic acid secondary structures, Ir-PDP exhibited a higher phosphorescence lifetime after interacting with G4. Two-photon phosphorescence lifetime imaging was used to detect the binding events of the probe in the cytoplasm. The addition of G4 binder PDS significantly regulated cytoplasmic phosphorescence lifetime. This project explored a new sensing pathway to observe the binding manners of probes in the cytoplasm through the phosphorescence lifetime of probes.

Summary: Development of simple, low-cost and highly sensitive electrochemiluminescence (ECL) sensors for various targets is urgent but remains challenging. Here, an iridium (III) complex (Ir-ECL) was synthesized and applied for improved target sensing. Ir-ECL exhibited ECL emission wavelength at 590 nm and showed superior intercalation ability into G-quadruplex DNA. Using these properties, Ir-ECL was applied to enzyme-free, label-free, sensitive and homogeneous ECL analysis of pesticide acetamiprid (Ace) based on aptamer-target recognition-driven hybridization chain reaction (HCR), resulting in improved Ace ECL sensing.

Summary: In this study, a rapid and label-free interference reduction nucleic acid amplification strategy (IR-NAAS) was developed for diagnosing COVID-19 RNAs sequences. It utilized luminescent iridium(III) probes, time-resolved emission spectroscopy (TRES), and multi-branch rolling circle amplification (mbRCA) to achieve high sensitivity and specificity. The method was successfully applied to detect COVID-19 RNA sequences in various environmental and biological samples, and also showed potential for detecting other RNA viruses.

Summary: Auramine o (AO) is a synthetic dye used in paper and textile industries. Although it has been banned as a food additive in many countries, it is still illegally used in certain food products and pharmaceuticals. Monitoring the presence of AO in food products is important to minimize health risks. This study developed a simple, sensitive, and selective label-free detection method based on G-quadruplex for AO. The method showed high specificity for AO and was successfully applied to detect AO in pasta, bulgur, and curry powder.

Summary: We synthesized a cobalt(II) complex of a curcumin-cysteine conjugate (Co-CC) as a fluorescent probe for detecting G-quadruplex DNA sequences. The probe showed strong fluorescence emission upon adding guanine-rich DNA sequences, with high selectivity and low detection limits. It also exhibited improved cytotoxicity towards A549 lung cancer cells and mild toxicity towards Vero cells compared to pristine curcumin. Real-world sample analysis showed promising results, and this study provides insights for improving bioavailability through metal-organic ligand complexation.

Summary: The study demonstrates that high levels of c-Myc protein in late-stage CRC can be effectively targeted by dihydroartemisinin treatment, resulting in reduced cell viability and altered metabolic phenotypes. Dihydroartemisinin shows stronger growth-inhibitory effects in late-stage CRC compared to early-stage CRC, indicating its potential as a therapeutic option for late-stage CRC. Additionally, the treatment reduces c-Myc expression, leading to downregulation of key metabolic enzymes and suppression of tumor cell proliferation.

Summary: This study reports the use of dehydroeburicoic acid (DEA) as a dual inhibitor of the Keap1-Nrf2 protein-protein interaction (PPI) and GSK3 beta, providing potential therapeutic strategies for alcoholic liver disease (ALD). DEA disrupts the Keap1-Nrf2 PPI and restores Nrf2 activity, leading to the activation of antioxidant genes. It also alleviates ethanol-induced mitochondrial dysfunction and exhibits minimal toxicity in the ALD cell model.
