Androgens control growth of prostate epithelial cells and androgen deprivation induces apoptosis, leading to prostate involution. We investigated the effects of surgical stress on prostate involution induced by androgen ablation and determined the underlying mechanisms. Androgen ablation in mice was induced by surgical castration and administration of the anti-androgenic drugs bicalutamide and MDV3100. Surgical stress was induced by sham castration under isoflurane anesthesia. Surgical stress delayed apoptosis and prostate involution induced by anti-androgenic drugs. These effects of stress were prevented by administering the selective beta2-adrenoreceptor antagonist ICI118,551 and were also blocked in BAD 3SA/WT mice expressing phosphorylation-deficient mutant BAD3SA. These results indicate that apoptosis and prostate involution in response to androgen ablation therapy could be delayed by surgical stress via the beta2-adrenoreceptor/BAD signaling pathway. Thus, surgery could interfere with androgen ablation therapy, whereas administration of beta2-adrenoreceptor antagonists may enhance its efficacy.

Summary: This study reviewed the feasibility of transcranial ultrasonography (TCU) as a diagnostic tool for evaluating intracranial pathology in critical care or austere settings. The results showed that TCU has reasonable sensitivity and specificity in detecting intracranial pathology including intracranial hemorrhage and tumors. This suggests that TCU can be clinically valuable in remote areas or where standard imaging is unavailable.

Estimating incidence of type 1 and type 2 diabetes using prevalence data: the SEARCH for Diabetes in Youth study

Summary: This study aimed to examine whether a partial differential equation (PDE) can be used to estimate the incidence of diabetes from prevalence. The results showed that the PDE method can accurately estimate the incidence rates of diabetes, and it has the advantage of cost savings compared to traditional methods.

Quad-shot-immunotherapy: quad-shot radiotherapy with pembrolizumab for advanced/recurrent head and neck cancer

Summary: Effective treatment options for advanced/recurrent head and neck squamous-cell carcinoma are limited. The combination of the immune checkpoint inhibitor pembrolizumab and quad-shot, a hypofractionated palliative radiotherapy regimen, shows promise as a potential treatment strategy. This study aims to evaluate the response rates, survival outcomes, and treatment toxicity of this combined therapy, as well as identify molecular biomarkers and immune-related impacts using correlative multiomics analysis.

Summary: This study examined the relationship between diabetes complications and cognitive dysfunction in young adults with type 1 or type 2 diabetes. The results showed that hypoglycemia and peripheral neuropathy were associated with cognitive deficits in type 1 diabetes, while high blood pressure was associated with cognitive deficits in type 2 diabetes. These findings provide insights for targeting specific factors to mitigate cognitive issues in young people with diabetes.

Summary: This study examined arterial stiffness in individuals with type 1 diabetes and explored the differences between Hispanic, non-Hispanic Black, and non-Hispanic White individuals. The results showed that cardiovascular and socioeconomic factors explain a quarter of the racial and ethnic differences in arterial stiffness, but non-Hispanic Black individuals still experienced higher arterial stiffness.

Summary: Different therapy options are used for NSCLC based on tumor stage and genetic mutations. However, biomarkers to guide treatment selection are limited. A study collected clinical and sequencing data from 524 NSCLC patients to examine the association between mutation profiles and treatment response. Mutation composite scores (MCS) were generated for different treatments, and MCS showed superior predictive power compared to other biomarkers in immune therapy-treated patients.
