Summary: This study identified A20 as a molecular target that can attenuate the fibrotic response induced by TGF beta 2-TLR4 signaling. Overexpression of A20 rescued the fibrotic response in TM cells when treated with TGF beta 2, LPS, or cFN-EDA.

Summary: Intraocular pressure is determined by the resistance of aqueous humor outflow, which is influenced by the mechanical properties of trabecular meshwork, Schlemm's canal inner wall, and juxtacanalicular connective tissue. Understanding the biomechanical interactions of aqueous humor with outflow tissues is crucial for regulating outflow resistance and fluid dynamics in the eye.

Summary: In this study, the researchers investigated the segmental outflow patterns in the trabecular meshwork of living mice and their changes over time and with age. They found that these patterns are not static but redistribute over time, with an age-related decline in the rate of redistribution.

Summary: This study investigates the dynamic fluid-structure interaction of the aqueous humor with the trabecular meshwork, juxtacanalicular tissue, and Schlemm's canal. The hyperviscoelastic biomechanical properties of the aqueous outflow tissues are revealed. Understanding the biomechanics of these tissues is crucial for studying the effects of drugs on intraocular pressure.

Summary: This study investigates the differences in outflow facility between high-flow and low-flow segments of the conventional outflow pathway in porcine eyes. It finds that there is an uneven distribution of local outflow facility across different parts of the trabecular meshwork.

Summary: The glycocalyx layer covering the conventional aqueous outflow pathway in the eye plays an important role in regulating outflow resistance and intraocular pressure. Research indicates that the charged glycocalyx layer has a minor biomechanical effect on the hydrodynamics of aqueous humor when intraocular pressure is elevated.

Summary: This study found that non-psychotropic cannabidiol (CBD) has a positive effect on regulating intraocular pressure. It enhances aqueous humor outflow and modifies signaling pathways in trabecular meshwork (TM) cells.

Summary: This study used X-ray computed tomography and scanning electron microscopy to reconstruct 3D microstructural finite element models of the healthy and glaucoma outflow tissues. The results showed that the primary site of outflow resistance in glaucoma eyes is the trabecular meshwork, while in healthy eyes, it is the juxtacanalicular connective tissue and the inner wall of Schlemm's canal. The glaucoma eyes had thicker trabecular meshwork and juxtacanalicular connective tissue and smaller trabecular space compared to healthy eyes.

Summary: The selective ROCK inhibitor Y39983 induces actin remodeling, decreases cell motility and transepithelial resistance in human TM cells. Proteomic analysis identified 20 significantly altered proteins after Y39983 treatment, involving pathways such as p53, integrin signaling, and cytoskeletal regulation by Rho GTPase. Downregulation of TSP1 by Y39983 and its antagonists led to reduced cell migration and increased outflow facility.

Summary: Elevated intraocular pressure (IOP) in primary open-angle glaucoma (POAG) is a major risk factor for irreversible vision loss. Over-expression of ID1 and ID3 can suppress TGF beta 2-induced ocular hypertension and decreased aqueous humor (AH) outflow facility. These proteins may be potential candidates for future IOP-lowering treatments in POAG.

Summary: The researchers developed an in vitro JCT model using a polycaprolactone nanofibrous scaffold, which could recapitulate the functional JCT morphology and study the flow of aqueous humor. The model exhibited HTM-like characteristics and allowed for investigation of the relationship between aqueous humor outflow and HTM degeneration as well as drug screening.

Summary: Intraocular pressure (IOP) fluctuates due to ocular pulse, which induces shear stress in Schlemm's canal (SC). The ocular pulse may modulate outflow facility by stimulating NO production. Studies showed the immediate increase in outflow facility in mice in response to the ocular pulse, with NO playing a significant role in this regulation.

Summary: The trabecular meshwork is a highly mechanosensitive tissue in the eye that regulates intraocular pressure. This study identified the Piezo1 channel as a key transducer of mechanical stimuli in TM cells, shedding light on the mechanisms of intraocular pressure regulation.
