A High Throughput Genotyping Approach Reveals Distinctive Autosomal Genetic Signatures for European and Near Eastern Wild Boar

The lack of a Near Eastern genetic signature in modern European porcine breeds indicates that, although domestic pigs from the Fertile Crescent entered Europe during the Neolithic, they were completely replaced by their European counterparts in a short window of time. Whilst the absence of such genetic signature has been convincingly demonstrated at the mitochondrial level, variation at the autosomal genomes of European and Near Eastern Sus scrofa has not been compared yet. Herewith, we have explored the genetic relationships among 43 wild boar from Europe (N = 21), Near East (N = 19) and Korea (N = 3), and 40 Iberian (N = 16), Canarian (N = 4) and Mangalitza (N = 20) pigs by using a high throughput SNP genotyping platform. After data filtering, 37,167 autosomal SNPs were used to perform population genetics analyses. A multidimensional scaling plot based on genome-wide identity-by-state pairwise distances inferred with PLINK showed that Near Eastern and European wild boar populations are genetically differentiated. Maximum likelihood trees built with TreeMix supported this conclusion i.e. an early population split between Near Eastern and European Sus scrofa was observed. Moreover, analysis of the data with Structure evidenced that the sampled Iberian, Canarian and Mangalitza pigs did not carry any autosomal signature compatible with a Near Eastern ancestry, a finding that agrees well with previous mitochondrial studies.

Characterizing the Mitochondrial Diversity of Arbi Goats from Tunisia

Supplementation of Estradiol Into Semen Extender Improved Goat Sperm Cryopreservation

Summary: The study found that supplementation of 3 and 5 mu M concentrations of estradiol can improve sperm viability and membrane integrity in goat semen, with the 3 mu M concentration also increasing the highest rate of progressive motility. However, a higher concentration of estradiol (5 mu M) reduced progressive motility. Additionally, there is a considerable interaction between EDTA and estradiol for sperm motility, with EDTA reducing the supportive effects of estradiol on sperm cryopreservation parameters.

An Electrochemical Protocol for CRISPR-Mediated Gene-Editing of Sheep Embryonic Fibroblast Cells

Summary: In this study, a protocol for electrotransfection of sheep embryonic fibroblast cells was developed. Optimal pulsing conditions and the addition of 3% dimethyl sulfoxide (DMSO) resulted in high transfection efficiency (>98%) and cell survival rate (>97%). CRISPR/Cas9-mediated gene editing of BMP15 and GDF9 genes in sheep EF cells was successfully achieved using the electroporation method.

Summary: The n-6/n-3 PUFA ratio in pig skeletal muscle affects the transcriptome expression related to muscle growth and immune pathways, as well as adipogenesis and immune response. Predicted miRNA-to-mRNA regulatory networks are involved in lipid metabolism, obesity, muscle growth, and protein degradation. The n-6/n-3 PUFA ratio differences revealed genes, miRNAs, and enriched pathways involved in lipid metabolism, cell proliferation, and inflammation.

CRISPR/Cas9-mediated targeted knock-in of large constructs using nocodazole and RNase HII

Copy Number Variation on ABCC2-DNMBP Loci Affects the Diversity and Composition of the Fecal Microbiota in Pigs

Summary: This study found that structural variants in the pig genome are associated with the diversity and composition of the pig gut microbiota. By analyzing whole-genome sequencing data, a CNV named DUP was found to be associated with richness and Shannon alpha-diversity of the fecal bacterial diversity. This association was validated using real-time quantitative PCR and confirmed in an independent crossbreed population. These results highlight the importance of considering host-genome structural variants as potential modulators of microbial ecosystems and identify ABCC2-DNMBP CNV as a host-genetic factor for the modulation of the pig fecal microbiota.
