Summary: The roles of PCNA as a scaffold protein in DNA replication and repair are well known, but its functions in the cytosol are less understood. ENO1 and 6PGD, two metabolic enzymes, interact with PCNA. Mutation of the PCNA interacting motif APIM in ENO1 resulted in reduced ENO1 protein levels, impaired growth rate, decreased glucose consumption, and reduced AKT activation. The interaction between PCNA and ENO1 has significant effects on metabolism.

The results show that the UP CIM recruits the CCR4-NOT deadenylase complex and activates mRNA degradation cooperatively with the conserved tryptophan residues of UP. Interestingly, the UP CIM remains unphosphorylated and can still promote association with the CCR4-NOT complex and mRNA decay upon activation of the p38-MAPK-activated kinase MK2.

ECD interacts with high-risk HPV E6 proteins and is overexpressed in cervical and head and neck squamous cell carcinoma. Its overexpression predicts shorter survival in patients and ECD knockdown impairs oncogenic behavior in cervical cancer cells.
