Clinical Significance of Annexin A2 Expression in Breast Cancer Patients

Lee D. Gibbs, Kelsey Mansheim, Sayantan Maji, Rajesh Nandy, Cheryl M. Lewis, Jamboor K. Vishwanatha, Pankaj Chaudhary

Summary: Annexin A2 (AnxA2) is overexpressed in tumor tissues and serum samples of breast cancer patients, especially in the TNBC subtype, where its expression is associated with poor prognosis. The phosphorylation of AnxA2 at tyrosine 23 appears to be a critical step for its secretion from TNBC cells and could serve as a potential diagnostic biomarker in TNBC patients.

Molecular Insights on the Possible Role of Annexin A2 in COVID-19 Pathogenesis and Post-Infection Complications

Prakash Patil, Praveenkumar Shetty, Nithin Kuriakose, Pavan Gollapalli, Sukanya Shetty, Roopa Bhandary, Jamboor K. Vishwanatha, Sudeep D. Ghate

Summary: SARS-CoV-2 has caused infections and deaths worldwide, with millions affected and no clinically proven antivirals available. Efforts to develop effective drugs are being delayed by mutations and lack of understanding of viral mechanisms. AnxA2 may play a potential role in SARS-CoV-2 pathogenesis and post-infection complications.

Novel Use of Hypoxia-Inducible Polymerizable Protein to Augment Chemotherapy for Pancreatic Cancer

Andrew Gdowski, Hamed Hayatshahi, Rafal Fudala, Rohan Joshi, Jin Liu, Jamboor K. Vishwanatha, Rohan Jeyarajah, Paul Guzik, Amalendu P. Ranjan

Summary: Pancreatic ductal adenocarcinoma (PDAC) is an aggressive malignancy with poor clinical outcomes. The dense stroma surrounding the tumor poses a challenge for drug delivery. A novel approach discussed in this study is the use of a self-polymerizing complex of gemcitabine (GEM) and hemoglobin S (HbS) to disrupt the tumor stroma and improve treatment efficacy. In an in vivo mouse model, the self-polymerization of HbS + GEM was more effective than individual agents at reducing tumor size. These findings suggest a potential clinical benefit of delivering the GEM and HbS complex via direct injection for patients with locally advanced disease.

Leading Predictors of COVID-19-Related Poor Mental Health in Adult Asian Indians: An Application of Extreme Gradient Boosting and Shapley Additive Explanations

Mohammad Ikram, Nazneen Fatima Shaikh, Jamboor K. Vishwanatha, Usha Sambamoorthi

Summary: A study investigated the predictors of poor mental health during the COVID-19 pandemic among Asian Indians in the United States. Using machine learning techniques, the study identified sleep disturbance, age, general health, income, wearing a mask, and self-reported discrimination as the leading predictors of poor mental health. The study suggests a paradoxical relationship between income and poor mental health, and emphasizes the importance of addressing sleep disturbance and perceived discrimination to reduce the risk of poor mental health in Asian Indians.

Combination of Small Extracellular Vesicle-Derived Annexin A2 Protein and mRNA as a Potential Predictive Biomarker for Chemotherapy Responsiveness in Aggressive Triple-Negative Breast Cancer

Priyanka P. P. Desai, Kalyani Narra, Johanna D. D. James, Harlan P. P. Jones, Amit K. K. Tripathi, Jamboor K. K. Vishwanatha

Summary: This study demonstrates that Annexin A2 (AnxA2) protein and mRNA highly associate with small extracellular vesicles (sEVs) in the later stages of triple-negative breast cancer (TNBC), which indicates a better treatment response. Therefore, sEV-derived AnxA2 protein and mRNA are introduced as a combined biomarker for neo-adjuvant chemotherapy response in TNBC.

Cabazitaxel-Loaded Nanoparticles Reduce the Invasiveness in Metastatic Prostate Cancer Cells: Beyond the Classical Taxane Function

Jana B. B. Lampe, Priyanka P. P. Desai, Amit K. K. Tripathi, Nirupama A. A. Sabnis, Zhe Chen, Amalendu P. P. Ranjan, Jamboor K. K. Vishwanatha

Summary: We developed cabazitaxel-loaded nanoparticles using PLGA and introduced a bone-targeting moiety on the surface. These nanoparticles showed the ability to inhibit EMT, invasion and migration in prostate cancer cells. Treatment with targeted cabazitaxel-loaded nanoparticles led to decreased expression of EMT marker, Vimentin, and increased expression of E-cadherin, resulting in suppressed cancer progression. Furthermore, phosphorylated Src and cofilin levels were affected, potentially impacting migration and invasion pathways. The nanoparticles also showed increased expression of p-120 catenin and inhibition in IL-8 expression. Overall, targeted cabazitaxel-loaded nanoparticles hold promise for treating bone-metastatic prostate cancer.

Demographic and Psychosocial Correlates of COVID-19 Vaccination Status among a Statewide Sample in Texas

Justin M. M. Luningham, Idara N. N. Akpan, Tanjila Taskin, Sarah Alkhatib, Jamboor K. K. Vishwanatha, Erika L. L. Thompson

Summary: This study explored the demographic and psychosocial correlates of COVID-19 vaccinations among a statewide sample in Texas. The findings showed that Hispanic/Latinx individuals were more likely to be partially vaccinated compared to non-Hispanic White individuals. Higher education levels and confidence in the FDA's ability to ensure a safe COVID-19 vaccine were strongly associated with a higher likelihood of being fully vaccinated. Challenges brought on by the pandemic and concerns about infection were also associated with a higher likelihood of being partially or fully vaccinated. These findings highlight the need to investigate individual and contextual factors in improving COVID-19 vaccination rates, especially among vulnerable and disadvantaged populations.
