Immunomodulatory Effects of Streptococcus suis Capsule Type on Human Dendritic Cell Responses, Phagocytosis and Intracellular Survival

Streptococcus suis is a major porcine pathogen of significant commercial importance worldwide and an emerging zoonotic pathogen of humans. Given the important sentinel role of mucosal dendritic cells and their importance in induction of T cell responses we investigated the effect of different S. suis serotype strains and an isogenic capsule mutant of serotype 2 on the maturation, activation and expression of IL-10, IL-12p70 and TNF-alpha in human monocyte-derived dendritic cells. Additionally, we compared phagocytosis levels and bacterial survival after internalization. The capsule of serotype 2, the most common serotype associated with infection in humans and pigs, was highly anti-phagocytic and modulated the IL-10/IL-12 and IL-10/TNF-alpha cytokine production in favor of a more anti-inflammatory profile compared to other serotypes. This may have consequences for the induction of effective immunity to S. suis serotype 2 in humans. A shielding effect of the capsule on innate Toll-like receptor signaling was also demonstrated. Furthermore, we showed that 24 h after phagocytosis, significant numbers of viable intracellular S. suis were still present intracellularly. This may contribute to the dissemination of S. suis in the body.

Summary: In this study, it is demonstrated that a metal stent is not notably heated up by IRE pulses under typical clinical conditions. By applying Maxwell's equations and simplifying the electric field generated by two needles in tissue, the heat generation and transport in IRE procedures can be accurately modeled. This explanation contributes to the observed low temperature rim of viable tissue around the stent in tissue model experiments.

Summary: Pancreatic ductal adenocarcinoma (PDAC) is an aggressive cancer with a poor prognosis. While immunotherapy has revolutionized the treatment of other advanced cancers, its effectiveness in PDAC is limited due to the immunosuppressive and hard-to-reach tumor microenvironment. However, various immunotherapies have shown potential in initiating immune responses, suggesting an immune potentiating effect. Successful PDAC treatment may rely on the synergistic effects of combination immunotherapies.

Summary: This study evaluated the impact of primary tumor sidedness of CRC, RAS and BRAF mutations, and MSI status on complications, survival outcomes, and local tumor progression following surgery and thermal ablation in patients with CRLM. The findings suggest the importance of wider margins for reducing LTP rates in patients with RAS mutated CRLM, particularly for thermal ablation, and highlight the potential contribution of upfront knowledge of molecular biomarkers to improve oncological outcomes.

Changes in the oral status and periodontal pathogens in a Sardinian rural community from pre-industrial to modern time

The research found that historical females had a higher incidence of caries, periodontal pathogens, and bacterial load than historical males; and modern individuals showed a notable increase in Red complex bacteria associated with periodontitis and other chronic diseases compared to historical individuals.

However, while C-11-sorafenib PET did not predict intratumoral sorafenib concentrations, a decrease in tumor perfusion after therapy showed an association with clinical benefit, suggesting further investigation is needed.

Summary: Viral respiratory infections often lead to bacterial co-infections, increasing disease severity and mortality, particularly during the SARS-CoV-2 pandemic. Streptococcus pneumoniae is a major lethal bacterial pathogen, but difficult to detect in SARS-CoV-2 patients. Rapid and sensitive diagnostic methods can aid targeted therapy.
