Erythropoietin Enhances Nerve Repair in Anti-Ganglioside Antibody-Mediated Models of Immune Neuropathy

Guillain-Barre syndrome (GBS) is a monophasic immune neuropathic disorder in which a significant proportion of patients have incomplete recovery. The patients with incomplete recovery almost always have some degree of failure of axon regeneration and target reinnervation. Anti-ganglioside antibodies (Abs) are the most commonly recognized autoimmune markers in all forms of GBS and specific Abs are associated with the slow/poor recovery. We recently demonstrated that specific anti-ganglioside Abs inhibit axonal regeneration and nerve repair in preclinical models by activation of small GTPase RhoA and its downstream effectors. The objective of this study was to determine whether erythropoietin (EPO), a pleiotropic cytokine with neuroprotective and neurotrophic properties, enhances nerve regeneration in preclinical cell culture and animal models of autoimmune neuropathy/nerve repair generated with monoclonal and patient derived Abs. Primary neuronal cultures and a standardized sciatic crush nerve model were used to assess the efficacy of EPO in reversing inhibitory effects of anti-ganglioside Abs on nerve repair. We found that EPO completely reversed the inhibitory effects of anti-ganglioside Abs on axon regeneration in cell culture models and significantly improved nerve regeneration/repair in an animal model. Moreover, EPO-induced proregenerative effects in nerve cells are through EPO receptors and Janus kinase 2/Signal transducer and activator of transcription 5 pathway and not via early direct modulation of small GTPase RhoA. These preclinical studies indicate that EPO is a viable candidate drug to develop further for neuroprotection and enhancing nerve repair in patients with GBS.

Virtual mouse brain histology from multi-contrast MRI via deep learning

Summary: This study demonstrates the use of deep convolutional neural networks to estimate histological staining intensity directly from MRI signals, providing three-dimensional images that closely mimic target histology and enhanced sensitivity and specificity compared to conventional MRI markers. Additionally, the relative contribution of each MRI contrast within the networks can be used to optimize multi-contrast MRI acquisition, serving as a resource for validating novel MRI techniques.

Inhomogeneous magnetization transfer MRI of white matter structures in the hypomyelinated shiverer mouse brain

Summary: In this study, we investigated the ability of ihMT MRI to detect white matter structures in the hypomyelinated shiverer mouse brain. Our results showed that ihMT can provide clear visualization of the hypomyelinated corpus callosum, with higher myelin contrast compared to conventional MT. These findings suggest the potential use of ihMT as a marker for early myelination or myelin repair.

Summary: This study analyzed the role of infections in Guillain-Barre syndrome (GBS) and their association with clinical features and disease severity. The distribution of infections varied across geographical regions, but the association between infection and clinical phenotype differed. Coinfections were common, highlighting the importance of broad serologic testing. Infections were also found to have prognostic value for GBS outcome.

CIDP: Analysis of Immunomarkers During COVID-19 mRNA-Vaccination and IVIg-Immunomodulation: An Exploratory Study

Summary: This exploratory study analyzed the impact of COVID-19 mRNA vaccine on disease activity and IVIg-immunomodulation in CIDP patients. The results showed that except for a significantly lower expression of CD32b on naive B cells after vaccination, there were no significant alterations in immunomarkers for CIDP or IVIg-mediated immunomodulation. This study suggests that COVID-19 mRNA vaccine does not affect immune activity in CIDP and does not alter the immunomodulatory effects of IVIg.

Using mesoscopic tract-tracing data to guide the estimation of fiber orientation distributions in the mouse brain from diffusion MRI

Summary: Diffusion MRI (dMRI) tractography is the only non-invasive tool for mapping macroscopic structural connectivity. This study demonstrated that a deep learning network trained on mesoscopic tract-tracing data can improve the estimation of fiber orientation distributions (FODs) in mouse brain dMRI data, leading to enhanced specificity in tractography results without compromising sensitivity.

Towards reliable reconstruction of the mouse brain corticothalamic connectivity using diffusion MRI

Summary: In this study, we aimed to optimize the imaging and computational pipeline to achieve the best possible spatial overlaps between dMRI tractography and tracer-based axonal projection maps in the mouse brain corticothalamic network. We developed a dMRI-based atlas of the mouse forebrain and reconstructed detailed corticothalamic structural connectivity matrices using different imaging and tractography parameters. Our results suggest that these parameters significantly affect tractography outcomes and our atlas can be used to investigate macroscopic structural connectivity in the mouse brain.

Summary: Peripheral neuropathy is a common side effect of cancer treatment with paclitaxel, but the mechanisms of paclitaxel transport into neurons are not well understood. This study identified OATP1A1 and OATP1B2 as the primary neuronal transporters for paclitaxel and suggested that inhibiting these transporters may prevent paclitaxel-induced neuropathy.

Repurposing MS immunotherapies for CIDP and other autoimmune neuropathies: unfulfilled promise or efficient strategy?

Summary: Despite advances in the treatment of CIDP and other AN, many patients remain unsatisfied with available treatments. Repurposing DMTs from other autoimmune conditions, particularly MS and NMOSD, is a promising strategy for establishing new treatment options for AN. B cell-directed and complement-targeting strategies, such as anti-CD20/anti-CD19 agents, Bruton's tyrosine kinase inhibitors, and anti-C5 agents, show promise as therapeutic approaches for AN.
