Summary: This study identified a novel circRNA, circUBAP2, in hepatocellular carcinoma (HCC) through bioinformatics analysis. CircUBAP2 was significantly upregulated in HCC tissues and correlated with aggressive clinical characteristics and poor prognosis. Functionally, circUBAP2 promoted cell proliferation, colony formation, migration, and invasion in vitro, and enhanced tumor growth and pulmonary metastasis in vivo. Mechanistically, circUBAP2 acted as a competing endogenous RNA (ceRNA) for miR-194-3p, thereby regulating the expression of MMP9 and promoting HCC progression. CircUBAP2 could potentially serve as a prognostic biomarker and therapeutic target for HCC patients.

Summary: This study investigates the effect of IDH1 hyperacetylation on the acidic microenvironment in colorectal cancer (CRC). The results show that increased alpha-KG affects the hydroxylation of Ago2 and mediates miR-9-5p targeting NHE1 protein. Knockdown of NHE1 inhibits CRC cell proliferation and migration by restricting intracellular H+ transport. This study reveals a novel mechanism through which IDH1 deacetylation regulates the cellular acidic microenvironment and inhibits CRC metastasis.

Summary: The study found that AOC1 expression significantly increased in CRC tissues, especially in liver metastases, and was associated with worse prognosis. AOC1 had higher expression in tumor organoids than in normal organoids, suggesting it was highly expressed in the tumor epithelium. Functional analysis demonstrated that AOC1 knockdown inhibited the proliferation and migration of CRC cells by inducing EMT. Xenograft tumor formation in nude mice showed that knockdown of AOC1 inhibited the tumor xenografts growth.
