Apolipoprotein J/Clusterin in Human Erythrocytes Is Involved in the Molecular Process of Defected Material Disposal during Vesiculation

Background: We have showed that secretory Apolipoprotein J/Clusterin (sCLU) is down-regulated in senescent, stressed or diseased red blood cells (RBCs). It was hypothesized that sCLU loss relates to RBCs vesiculation, a mechanism that removes erythrocyte membrane patches containing defective or potentially harmful components. Methodology/Principal Findings: To investigate this issue we employed a combination of biochemical and microscopical approaches in freshly prepared RBCs or RBCs stored under standard blood bank conditions, an in vitro model system of cellular aging. We found that sCLU is effectively exocytosed in vivo during membrane vesiculation of freshly prepared RBCs. In support, the RBCs' sCLU content was progressively reduced during RBCs ex vivo maturation and senescence under cold storage due to its selective exocytosis in membrane vesicles. A range of typical vesicular components, also involved in RBCs senescence, like Band 3, CD59, hemoglobin and carbonylated membrane proteins were found to physically interact with sCLU. Conclusions/Significance: The maturation of RBCs is associated with a progressive loss of sCLU. We propose that sCLU is functionally involved in the disposal of oxidized/defected material through RBCs vesiculation. This process most probably takes place through sCLU interaction with RBCs membrane proteins that are implicit vesicular components. Therefore, sCLU represents a pro-survival factor acting for the postponement of the untimely clearance of RBCs.

Beta thalassemia minor is a beneficial determinant of red blood cell storage lesion

Summary: Blood donor genetics and lifestyle have an impact on the quality of stored red blood cells (RBCs). This study examined the storability of RBCs from beta thalassemia donors and found that they exhibited better levels of storage hemolysis and resistance to osmotic, oxidative, and mechanical stress compared to controls. Metabolomics analysis revealed changes in purine and arginine pathways in beta thalassemia RBCs at baseline and during storage. The findings suggest that beta thalassemia mutations affect the metabolism and susceptibility to hemolysis of stored RBCs.

Summary: This study compared the neutralizing antibody levels after vaccination with BNT162b2 mRNA vaccine and ChAdOx1 (AZD1222) viral vector vaccine at different time points. The results showed that BNT162b2 induced stronger neutralizing antibody responses compared to AZD1222, and both vaccines showed a gradual decline in antibody titers over time. Additionally, the rate of antibody elimination was higher in those vaccinated with AZD1222 compared to BNT162b2. Age, gender, body mass index, or comorbidities did not significantly impact antibody levels over time.

Evaluation of the Small Heat Shock Protein Family Members HSPB2 and HSPB3 in Bladder Cancer Prognosis and Progression

Summary: This study investigated the expression and role of HSPB2 and HSPB3 genes in bladder cancer patients and bladder cancer cells. The results showed increased expression levels of HSPB2 and HSPB3 genes and proteins in high-grade bladder cancer cells. In clinical samples, the expression levels of HSPB2 and HSPB3 genes were significantly downregulated in bladder tumors but showed an upregulation trend in muscle-invasive and high-grade tumors. These findings suggest that HSPB2 and HSPB3 have a pro-survival role in advanced bladder cancer and can be targeted as novel molecular markers for treatment optimization.

Summary: The study aimed to evaluate the diagnostic role of rotational thromboelastometry (ROTEM) in periprosthetic joint infections (PJIs). The results showed that several ROTEM parameters differed in patients with PJI, indicating a higher coagulation potential associated with the infection. Among these parameters, EXTEM maximum clot firmness (MCF) had the highest diagnostic accuracy for PJI.
