Summary: This study demonstrated that mTOR inhibitors can help preserve renal function in liver transplantation recipients, especially in patients with renal impairment, without serious adverse events. Regardless of the timing of initiation, a switch to mTOR inhibitors showed significant improvement in kidney function.

Summary: This study investigated the safety and adaptability of combination options of targeted agents with immune checkpoint inhibitors in patients with advanced or metastatic renal cell carcinoma. The results showed that lenvatinib plus everolimus had worse safety compared to other treatments, while everolimus was generally the safest agent. This study has important implications for treatment options and trial design optimization in advanced or metastatic RCC.

Summary: This study reveals a mechanism for controlling lysosome biogenesis involving calcium release from the endoplasmic reticulum, independent of mechanistic target of rapamycin complex 1. The mechanism includes inositol polyphosphate-5-phosphatase A-mediated triphosphate hydrolysis, calcineurin, and the nuclear translocation and elevated transcriptional activity of TFEB/TFE3.

Summary: Neurofibromatosis type 1 is a common disorder that requires early diagnosis for proper guidance on preventive measures. Mutations in the NF1 gene lead to loss of function of the neurofibromin protein, which affects the regulation of the RAS signaling pathway. Treatment options include drugs and radiofrequency therapy.

Summary: Withdrawal of steroids or switching to everolimus instead of cyclosporine are two effective strategies to reduce treatment-related side effects and improve long-term outcomes of kidney transplantation. Both regimens provide similar 5-year patient survival and death-censored graft survival, but the everolimus group showed higher (51)CrEDTA clearance at 1 and 5 years post-transplantation.

Summary: This study aimed to investigate the efficacy of sequential nivolumab treatment for metastatic renal cancer. The results showed that the overall survival was significantly longer in the 2017-2020 group compared to the 2014-2016 group. Nivolumab treatment as second-line therapy and prompt switching to nivolumab were identified as important factors for improved survival. These findings suggest the importance of sequential nivolumab treatment in patients who have received first-line molecular targeted therapy.

Summary: This study investigated the efficacy and safety of sirolimus in children with TSC-associated RAML. It was found that sirolimus could promote the disappearance of TSC-related RAML, and the disappearance rate was correlated with the maximum diameter at baseline, with smaller tumors having a higher disappearance rate.

Summary: This study evaluated the role and expression of MEK5 signaling in ccRCC and determined its relevance to the mTOR signaling pathway. The results showed that MEK5 expression was significantly increased in ccRCC and activated the mTOR pathway, regulating cell proliferation, migration, and clone formation in ccRCC. Targeted inhibition of MEK5 may represent a promising new strategy for treating ccRCC.

Summary: Recent studies have shown that translation inhibitors can facilitate the nuclear translocation of transcription factors EB (TFEB) and E3 (TFE3), leading to increased expression of downstream genes involved in autophagosome and lysosome biogenesis and function. However, translation inhibition also impairs degradative autolysosome formation due to lysosomal dysfunction. This highlights a novel biological role of translation inhibition in autophagy regulation.

Summary: This study aimed to detect the expression of midkine in psoriasis and investigate its potential role in the disease. The results showed that levels of midkine were significantly increased in both lesions and serum of patients with psoriasis, and a positive correlation was found between midkine and disease severity. It was also found that midkine promoted cell proliferation and angiogenesis through the Notch2/HES1/JAK2-STAT5A pathway. These findings suggest that midkine may be a potential therapeutic target for psoriasis treatment.
