Summary: Adoptive transfer of CAR-T cells can effectively treat refractory hematologic malignancies, but the expansion and persistence of T cells in vivo are limited. In this study, the researchers found that limiting the differentiation of CAR-T cells during ex vivo manufacturing can enhance their proliferation and antitumor efficacy. By providing an agonistic NOTCH1 signal during CD4(+) T-cell activation and CAR-T production, the researchers induced a stem cell memory phenotype and increased antigen responsiveness, leading to improved tumor clearance. Additionally, the activation of NOTCH1 signaling enhanced the expansion and effectiveness of cotransferred CD8(+) CAR-T cells. These findings suggest that activating NOTCH1 signaling during genetic modification ex vivo can enhance the function of tumor-targeting CAR-T cells.

Summary: This study investigated the effects of cART on biomarkers of platelet activation in HIV-infected individuals. The findings suggest that sCD40L may be a potential biomarker of successful cART, while PDGF-BB levels may indicate persistent low-level antigenemia. The study highlighted the importance of monitoring biomarkers of platelet activation in HIV treatment.

Summary: This study investigates the expression of CREM alpha in SLE CD4(+) T cells and explores the mechanisms for its regulation. The findings suggest that decreased SUV39H1 and increased Set1 levels contribute to the overproduction of CREM alpha in SLE CD4(+) T cells, leading to altered secretion of IL-2 and IL-17A and ultimately promoting SLE progression.

Summary: This study investigated the relationship between coagulation and changes in lipid parameters in newly-diagnosed patients with systemic lupus erythematosus (SLE) in the presence of pulmonary arterial hypertension (PAH). It found that age and D-dimer were independent predictors of PAH in SLE, and that D-dimer mediated 25.61% of the effect of low LDL on the presence of PAH in SLE.

Summary: HIV-infected immune non-responders (INR) exhibit higher proportions of CD4 T cell subsets expressing markers of exhaustion and senescence, as well as elevated plasma levels of IL-6, IP10, and sCD14. INR have higher levels of IP10 and lower levels of TGF-beta, with decreased cycling proportions of CD4 T regulatory cells. Proportions of T cells expressing TIGIT, PD-1, and CD57 positively correlate with IL-6 levels, while TGF-beta levels negatively correlate with proportions of TIGIT+ and PD-1+ T cell subsets.
