We have developed a rat brain organotypic culture model, in which tissue slices contain cortex-subventricular zone-striatum regions, to model neuroblast activity in response to in vitro ischemia. Neuroblast activation has been described in terms of two main parameters, proliferation and migration from the subventricular zone into the injured cortex. We observed distinct phases of neuroblast activation as is known to occur after in vivo ischemia. Thus, immediately after oxygen/glucose deprivation (6-24 hours), neuroblasts reduce their proliferative and migratory activity, whereas, at longer time points after the insult (2 to 5 days), they start to proliferate and migrate into the damaged cortex. Antagonism of ionotropic receptors for extracellular ATP during and after the insult unmasks an early activation of neuroblasts in the subventricular zone, which responded with a rapid and intense migration of neuroblasts into the damaged cortex (within 24 hours). The process is further enhanced by elevating the production of the chemoattractant SDf-1 alpha and may also be boosted by blocking the activation of microglia. This organotypic model which we have developed is an excellent in vitro system to study neurogenesis after ischemia and other neurodegenerative diseases. Its application has revealed a SOS response to oxygen/glucose deprivation, which is inhibited by unfavorable conditions due to the ischemic environment. Finally, experimental quantifications have allowed us to elaborate a mathematical model to describe neuroblast activation and to develop a computer simulation which should have promising applications for the screening of drug candidates for novel therapies of ischemia-related pathologies.

Nonresonant bilinear forms for partially dissipative hyperbolic systems violating the Shizuta-Kawashima condition

Roberta Bianchini, Roberto Natalini

Summary: In the context of hyperbolic systems of balance laws, the Shizuta-Kawashima coupling condition guarantees dissipativity of all variables, even in partially dissipative systems. This condition is crucial for the global existence of classical solutions. However, it is common to find physically based models that violate this condition. In this paper, we study two examples of partially dissipative hyperbolic systems that violate the Shizuta-Kawashima condition, and prove that as long as the source term is nonresonant in the non-dissipative directions, singularities formation is prevented and smooth solutions exist globally in time.

A forecasting model for the porosity variation during the carbonation process

Gabriella Bretti, Maurizio Ceseri, Roberto Natalini, Maria Carla Ciacchella, Maria Laura Santarelli, Giulia Tiracorrendo

Summary: This paper introduces a mathematical model to describe the intermediate chemical reactions in the carbonation process of concrete. Through simulation algorithms, researchers are able to accurately describe the effects of the interaction between concrete and CO2 on the porosity of the material, which is in substantial accordance with experimental results. Additionally, the dissolution of calcium carbonate under an acid environment is considered, and a trend inversion in the evolution of porosity is observed for long exposure times, significantly affecting the durability of the concrete.

Spatial organization of neuron-astrocyte interactions in the somatosensory cortex

Andres M. Baraibar, Lindsey Belisle, Giovanni Marsicano, Carlos Matute, Susana Mato, Alfonso Araque, Paulo Kofuji

Summary: Microcircuits in the neocortex are functionally organized along layers and columns, and astrocytes play a role in the functional organization and bidirectional interaction between neurons. Activation of astrocytes by endocannabinoids leads to an increase in Ca2+ levels, ATP/adenosine release, and temporary synaptic depression between layer 5 pyramidal neurons at relatively distant synapses. This form of heteroneuronal synaptic depression is specific to cortical columns and layers, depending on layer-specific neuron-to-astrocyte signaling.

Microglia and meningeal macrophages depletion delays the onset of experimental autoimmune encephalomyelitis

Alejandro Montilla, Alazne Zabala, Marco Er-Lukowiak, Bjoern Rissiek, Tim Magnus, Noelia Rodriguez-Iglesias, Amanda Sierra, Carlos Matute, Maria Domercq

Summary: In multiple sclerosis and EAE, both resident microglia and infiltrating macrophages contribute to demyelination and remyelination, but their specific roles are unknown. By depleting microglia using CSF-1R inhibitor and distinguishing blood-derived macrophages from microglia, it was found that microglia and meningeal macrophages have a specific role in antigen presentation and T cell reactivation at initial stages of EAE.

Summary: Oxidized cholesterol metabolite 27-hydroxycholesterol (27-OH) is a potential link between hypercholesterolemia and neurodegenerative diseases as it can cross the blood-brain barrier. This study found that high levels of 27-OH can impact oligodendrocyte function and contribute to the disconnection of neural networks in neurodegenerative diseases.

THE MEAN-FIELD LIMIT FOR HYBRID MODELS OF COLLECTIVE MOTIONS WITH CHEMOTAXIS

Roberto Natalini, Thierry Paul

Summary: In this paper, a general class of hybrid mathematical models is proposed to study the collective motions of cells under chemical stimuli. The models combine discrete entities represented by ODEs for cells and a continuous signal described by a diffusive equation for chemoattractant. The mean-field limit of this model is proved in the Wasserstein distance to a system comprised of a Vlasov-type equation coupled with the chemoattractant equation. The research is not based on empirical measures but instead focuses on the marginals of individual densities, and the limit system is shown to have explicit bounds with existence and uniqueness.

Summary: This study reveals that hepatic S-adenosylmethionine (SAMe) acts as a metabolic sensor of nutrition and fine-tunes the fasting response by regulating various cellular processes in mice. It shows that SAMe modulates PEMT activity, ER-mitochondria contacts, lipid oxidation, ATP production in the liver, and FGF21-mediated lipolysis and thermogenesis in adipose tissues. Additionally, the study highlights the role of glucagon-induced SAMe synthesis in preventing excessive lipid oxidation and mitochondrial ATP synthesis, thereby protecting against ER stress and liver injury.

Alterations of Oligodendrocyte and Myelin Energy Metabolism in Multiple Sclerosis

Eneritz Lopez-Muguruza, Carlos Matute

Summary: Multiple sclerosis (MS) is an autoimmune disease characterized by demyelination and neurodegeneration. Metabolic changes, including mitochondrial dysfunction and alterations in glucose and lipid metabolism, contribute significantly to the pathogenesis of MS. Understanding these mechanisms is crucial for developing targeted therapies to preserve myelin and promote neurological recovery in individuals with MS.

Summary: In this paper, a mathematical model is proposed and analyzed to describe the sulphation process of marble in a specific material. The model considers rugosity and damaging effects. However, there are technical difficulties that hinder the theoretical advancement. Physically reasonable modifications are introduced to establish the existence of a suitable solution. Numerical simulations are presented and discussed for further research.
