We have previously demonstrated that both parasite genetic variability and host genetic background were important in determining the differential tissue distribution of the Col1.7G2 and JG T. cruzi monoclonal strains after artificial infections in mice. We observed that the JG strain was most prevalent in hearts of mouse lineages with the MHC haplotype H-2(d) (BALB/c and DBA2), while Col1.7G2 was predominant in hearts from C57BL/6 mice, which have the H-2(b) haplotype. To assess whether the MHC gene region indeed influenced tissue tropism of T. cruzi, we used the same two parasite strains to infect C57BL/6 (H-2(b)) and C57BLKS/J (H-2(d)) mice; the latter strain results from the introgression of DBA2 MHC region into the C57BL/6 background. We also performed ex vivo infections of cardiac explants from four congenic mice lineages with the H-2(b) and H-2(d) haplotypes arranged in two different genetic backgrounds: C57BLKS/J (H-2(d)) versus C57BL/6 (H-2(b)) and BALB/c (H-2(d)) versus BALB/B10-H2(b) (H-2(b)). In agreement with our former observations, Col1.7G2 was predominant in hearts from C57BL/6 mice (H-2(b)), but we observed a clear predominance of the JG strain in hearts from C57BLKS/J animals (H-2(d)). In the ex vivo experiments Col1.7G2 also prevailed in explants from H-2(b) animals while no predominance of any of the strains was observed in H-2(d) mice explants, regardless of the genetic background. These observations clearly demonstrate that the MHC region influences the differential tissue distribution pattern of infecting T. cruzi strains, which by its turn may be in a human infection the determinant for the clinical forms of the Chagas disease.

Summary: Zika virus (ZIKV) causes Congenital Zika Syndrome (CZS) in individuals exposed during pregnancy. Studies show that ZIKV infection upregulates miR-124 expression, leading to a decrease in TFRC, a targeted gene of this miRNA. This study investigates whether genetic variants affecting the expression of these genes could increase the risk of CZS development.

New synthetic nano-immunotherapy (OncoTherad?) for non-muscle invasive bladder cancer: Its synthesis, characterization and anticancer property

W. J. Favaro, J. C. C. Alonso, B. R. de Souza, I. B. Reis, J. M. Goncalves, A. C. Deckmann, G. Oliveira, Q. C. Dias, N. Duran

Summary: BCG-based intravesical immunotherapy is the gold standard treatment for high-risk non-muscle invasive bladder cancer. However, some high-risk patients experience relapse and do not respond to BCG. Our research group developed OncoTherad (R) nano-immunotherapy, which showed positive outcomes and mechanisms of action in an animal model with NMIBC. OncoTherad (R) nano-immunotherapy was more effective than BCG treatment, reducing the malignancy rate by 70% through modulation of TLR4-mediated interferon and RANK/RANKL signaling pathways.

Summary: The sediments collected from different locations in the Mariu Archipelago, Rio Negro, Brazil were analyzed. Quartz was found to be the predominant mineral in the sediments, along with a trace quantity of kaolinite. The sediments were also found to contain rare-earth elements, metals, and semi-metals. The characteristics and lifetimes of the quartz's thermoluminescence peaks were determined, and the samples were dated using the conventional SAR protocol.

The Microbiota-Dependent Worsening Effects of Melatonin on Gut Inflammation

Jefferson Luiz da Silva, Lia Vezenfard Barbosa, Camila Figueiredo Pinzan, Viviani Nardini, Irislene Simoes Brigo, Cassia Aparecida Sebastiao, Jefferson Elias-Oliveira, Vania Brazao, Jose Clovis do Prado Junior, Daniela Carlos, Cristina Ribeiro de Barros Cardoso

Summary: Dysbiosis and disturbances in gut homeostasis can lead to dysregulated responses in inflammatory bowel diseases. Melatonin has shown potential as a therapeutic option due to its interactions with immune responses and gut microbiota. In this study, MLT treatment increased inflammation in acute colitis and delayed recovery during remission.

Toxicological Effects of Copaiba Oil (Copaifera spp.) and Its Active Components

Camila Castanho Cardinelli, Josiane Elizabeth Almeida E. Silva, Rayssa Ribeiro, Valdir F. F. Veiga-Junior, Elisabete Pereira dos Santos, Zaida Maria Faria de Freitas

Summary: Vegetable oils are important resources in the Amazon, including oleoresins with interesting characteristics and potential pharmacological properties. Copaiba oils, which come from the trunks of Copaifera (Fabaceae) spp. trees, are composed of terpenes from the sesquiterpene and diterpene classes, with varying amounts depending on factors such as soil type. This review examines the toxicological studies of copaiba oils, both in vitro and in vivo, as well as the cytotoxic characteristics of the sesquiterpenes and diterpenes in these oils, using in silico, in vitro, and in vivo models.

Generalization of deep learning models for natural gas indication in 2D seismic data

Luis Fernando Marin Sepulveda, Marcelo Gattass, Aristofanes Correa Silva, Roberto Quevedo, Diogo Michelon, Carlos Siedschlag, Roberto Ribeiro

Summary: Methods based on Machine Learning and Deep Learning are popular for interpreting large volumes of data across various areas and tasks. This work proposes a method to improve the generalization of a Deep Learning model for indicating natural gas in 2D seismic data by recommending training data and hyperparameter operations. The experiments show an increase in the correct indication of natural gas by 8% to 37% and a fluctuation in the increase of false positives by -2% to 13%, resulting in an improvement in the generalization of the Deep Learning model of up to 11% according to the F1 score metric or up to 10% according to the IoU metric.

Talita Caroline de Oliveira Valentino, Carlos Eduardo Paiva, Marco Antonio de Oliveira, David Hui, Eduardo Bruera, Miguel Juliao, Bianca Sakamoto Ribeiro Paiva

Summary: This study aimed to evaluate the preferred place-of-death (PPoD) among patients with advanced cancer over time, and the concordance between preferred and actual place-of-death. The study found that home was the most common PPoD for patients, while palliative care units (PCUs) and hospitals were preferred for patients with severe symptoms. The overall agreement between last preference and actual place-of-death was low.

Seismic data classification for natural gas detection using training dataset recommendation and deep learning

Luis Fernando Marin Sepulveda, Marcelo Gattass, Aristofanes Correa Silva, Roberto Quevedo, Diogo Michelon, Carlos Siedschlag, Roberto Ribeiro

Summary: This article introduces a domain adaptation method based on comparing seismic features, which is used to detect natural gas in 2D seismic data. The proposed method does not require modifying the training data or DL model architecture, but focuses on analyzing the training data to recognize patterns for comparing and clustering similar seismic data. Compared to the DL model trained with all available seismic data, the proposed method achieves significant improvements in precision, recall, F1 score, and IoU.
