Summary: This study characterized the global epidemiology of different types of metastatic prostate cancer and assessed the prevalence and prognostic impact of homologous recombination repair gene alterations in advanced disease setting. The results indicated that there is limited real-world evidence on the prevalence of prostate cancer and HRRm beyond mCRPC, with BRCA2 being one of the most common genes associated with HRRm in mCRPC.

Summary: The purpose of this study was to determine the mortality rates of prostate cancer and other causes in low and favorable intermediate-risk active surveillance patients. The study found that there was no significant difference in the 5-year mortality rates between low-risk and intermediate-risk patients, but within the intermediate-risk group, the subgroup with PSA 10-20 ng/ml had significantly higher cancer mortality rates.

Summary: The study aims to evaluate the effectiveness of physical exercise in improving quality of life in patients with prostate cancer through a systematic review and meta-analysis. Results show that aerobic training regimens have a protective effect on the quality of life of patients with prostate cancer compared to other types of exercise.

Summary: This article systematically reviewed and meta-analyzed the association between nitrate, nitrite, and N-nitroso compounds (NOCs) in food and water and the risk of gastrointestinal cancers. The results showed that high intake of these compounds was significantly associated with overall GI cancer risk as well as nitrite and N-nitrosodimethylamine (NDMA). Dietary nitrite intake was linked to increased gastric cancer (GC) and esophageal cancer (EC) risks, while dietary NDMA intake was associated with increased colorectal cancer (CRC) risk.

Summary: This study analyzed and compared the efficacy of combination systemic therapies in metastatic hormone-sensitive prostate cancer and metastatic castration-resistant prostate cancer with or without visceral metastasis. It was found that adding androgen receptor signaling inhibitor to standard of care improved overall survival in patients with visceral metastasis. In metastatic hormone-sensitive prostate cancer patients, darolutamide + docetaxel + androgen deprivation therapy had the highest likelihood of improved overall survival.

Summary: This study systematically assessed the early detection rate of biochemical prostate cancer recurrence using choline, fluciclovine, and PSMA. The results showed that the detection rates of PSMA-radiotracers were greater than those of choline-radiotracers and fluciclovine-radiotracers at the patient level, especially with F-18-labeled PSMA.

Summary: Causes of death during PCa survivorship vary by patient and tumor characteristics. Men with early-stage prostate cancer are 4-fold more likely to die of other causes, whereas those with advanced prostate cancer are at increased risk for several causes not related to prostate cancer in comparison with the general population. These findings can help guide physicians taking care of men with a diagnosis of prostate cancer.

Summary: Based on the systematic review, there was no significant difference in infection rate, fever, sepsis, or readmission rate after transperineal ultrasound-guided prostate biopsy between cases utilizing antibiotic prophylaxis and those not utilizing antibiotic prophylaxis.

Summary: The PI-RADS v2 score is independently associated with the risk of biochemical recurrence (BCR) after definitive treatment for prostate cancer, with higher scores indicating a higher risk. This suggests that MRI findings using the PI-RADS v2 classification have prognostic significance in addition to their diagnostic role in prostate cancer.

Summary: The study found that men with prostate cancer have higher other-cause mortality in lower risk disease compared to higher risk disease, even after controlling for lifestyle characteristics and comorbidities. Further research is needed to identify factors contributing to this difference.

Summary: This study found that men with prostate cancer who received androgen deprivation therapy have a higher risk of developing dementia and/or Alzheimer's disease compared to those who did not receive the therapy, especially when the therapy lasted for more than 12 months.

Summary: This systematic review and network meta-analysis assessed the efficacy of different penile rehabilitation strategies post radical prostatectomy. The findings suggest that daily or nightly 100 mg sildenafil is the best strategy to improve erectile function recovery, while pelvic floor muscle training also shows effectiveness in increasing erectile function recovery rate.

Summary: This study evaluated the accuracy of MRI-inclusive nomograms and traditional clinical nomograms in predicting extraprostatic extension (EPE) in prostate cancer (PCa), and found that both had moderate diagnostic performance. The study provides baseline estimates for future studies, which are useful for evaluating preoperative risk stratification in PCa patients.

Summary: This population-based study examines the trends and variation in the use of primary radiotherapy for non-metastatic prostate cancer in the Netherlands from 2008 to 2019. The results show that EBRT use remained stable in low-risk patients, while increasing in intermediate and high-risk patients. The study also finds variation in treatment decisions based on patient and disease characteristics.

Summary: The study found that negative followup biopsy is significantly associated with a decreased risk of reclassification and upgrading in prostate cancer patients, particularly among those in Gleason Grade prognostic group 1. Incorporating negative followup biopsy results into current protocols can help tailor personalized active surveillance and improve decision-making accuracy.
