For two decades Vogelstein's model has been the paradigm for describing the sequence of molecular changes within protein-coding genes that would lead to overt colorectal cancer (CRC). This model is now too simplistic in the light of recent studies, which have shown that our genome is pervasively transcribed in RNAs other than mRNAs, denominated non-coding RNAs (ncRNAs). The discovery that mutations in genes encoding these RNAs [i.e., microRNAs (miRNAs), long non-coding RNAs, and circular RNAs] are causally involved in cancer phenotypes has profoundly modified our vision of tumour molecular genetics and pathobiology. By exploiting a wide range of different mechanisms, ncRNAs control fundamental cellular processes, such as proliferation, differentiation, migration, angiogenesis and apoptosis: these data have also confirmed their role as oncogenes or tumor suppressors in cancer development and progression. The existence of a sophisticated RNA-based regulatory system, which dictates the correct functioning of protein-coding networks, has relevant biological and biomedical consequences. Different miRNAs involved in neoplastic and degenerative diseases exhibit potential predictive and prognostic properties. Furthermore, the key roles of ncRNAs make them very attractive targets for innovative therapeutic approaches. Several recent reports have shown that ncRNAs can be secreted by cells into the extracellular environment (i.e., blood and other body fluids): this suggests the existence of extracellular signalling mechanisms, which may be exploited by cells in physiology and pathology. In this review, we will summarize the most relevant issues on the involvement of cellular and extracellular ncRNAs in disease. We will then specifically describe their involvement in CRC pathobiology and their translational applications to CRC diagnosis, prognosis and therapy.

FUS driven circCNOT6L biogenesis in mouse and human spermatozoa supports zygote development

Teresa Chioccarelli, Geppino Falco, Donato Cappetta, Antonella De Angelis, Luca Roberto, Martina Addeo, Marco Ragusa, Davide Barbagallo, Liberato Berrino, Michele Purrello, Concetta Ambrosino, Gilda Cobellis, Riccardo Pierantoni, Rosanna Chianese, Francesco Manfrevola

Summary: The formation of circular RNAs (circRNAs) involves a backsplicing reaction facilitated by inverted repeats and RNA-binding proteins, with FUS playing a key role in this process. Research on male mice showed that sperm cells have the ability to circularize mRNAs, with FUS acting as a modulator of backsplicing under CB1 stimulation. A physical interaction between FUS and CNOT6L, as well as cooperation among FUS, RNA Polymerase II, and Quaking, was observed in sperm cells.

Competing endogenous RNA network mediated by circ_3205 in SARS-CoV-2 infected cells

Davide Barbagallo, Concetta Ilenia Palermo, Cristina Barbagallo, Rosalia Battaglia, Angela Caponnetto, Vittoria Spina, Marco Ragusa, Cinzia Di Pietro, Guido Scalia, Michele Purrello

Summary: This study found that SARS-CoV-2 can synthesize a circular RNA called circ_3205 upon host cell infection. It interacts with miRNA and contributes to the upregulation of KCNMB4 and PRKCE mRNA, which are associated with blood coagulation and immune response.

Summary: In the tumor microenvironment, cancer cells activate Cancer Associated Fibroblasts (CAFs) to increase their secretion of soluble factors, which contribute to tumor cell proliferation, invasion, and dissemination. STAT3 and IL-6, as key factors, work together to maintain high levels of IL-6 secretion and STAT3 activation. The study demonstrated the essential role of STAT3 in the pro-tumorigenic functions of murine breast cancer CAFs, and identified a STAT3 signature enriched for genes encoding secreted proteins.

Genetics and RNA Regulation of Uveal Melanoma

Cristina Barbagallo, Michele Stella, Giuseppe Broggi, Andrea Russo, Rosario Caltabiano, Marco Ragusa

Summary: Uveal melanoma (UM) is a rare eye cancer with a high mortality rate due to metastases. UM and cutaneous melanoma (CM) have different molecular alterations and biological behavior. This review discusses the genetic and epigenetic landscapes of UM, with potential implications for biomarker identification and therapeutic targets.

Er:Y2O3 and Nd:Y2O3 Nanoparticles: Synthesis, Pegylation, Characterization and Study of Their Luminescence Properties

Regina Maria Chiechio, Rosalia Battaglia, Angela Caponnetto, Ester Butera, Giorgia Franzo, Riccardo Reitano, Michele Purrello, Marco Ragusa, Davide Barbagallo, Cristina Barbagallo, Cinzia Di Pietro, Valerie Marchi, Maria Jose Lo Faro, Annalinda Contino, Giuseppe Maccarrone, Paolo Musumeci

Summary: Lanthanide-doped yttrium oxide nanoparticles with selective upconversion properties have significant value in nanomedicine for sensing and diagnostics. Different syntheses of Er:Y2O3 and Nd:Y2O3 nanoparticles were optimized to obtain small particles of regular shape and good crystallinity. The nanoparticles were well dispersed with dimensions of tens of nanometers, and exhibited good emission and upconversion. Pegylation functionalization allowed for biocompatibility, water solubility, and dispersion of the nanoparticles, enabling them to serve as photoactive materials for theragnostics and biosensing.

An Uncharacterised lncRNA Coded by the ASAP1 Locus Is Downregulated in Serum of Type 2 Diabetes Mellitus Patients

Cristina Barbagallo, Michele Stella, Stefania Di Mauro, Alessandra Scamporrino, Agnese Filippello, Francesca Scionti, Maria Teresa Di Martino, Michele Purrello, Marco Ragusa, Francesco Purrello, Salvatore Piro

Summary: Diabetes mellitus (DM) is a complex disease characterized by high blood glucose levels. Type 2 Diabetes (T2D) is the most common form, affecting middle-aged or elderly individuals and its incidence is increasing among children and adults. This study analyzed the transcriptome of serum samples from T2D patients and identified a long non-coding RNA (lncRNA) as a potential diagnostic biomarker. The combination of this lncRNA with clinical parameters showed better diagnostic performance than the glycated haemoglobin test. Using this biomarker in clinical practice would improve the diagnosis and management of T2D patients.

Anatomical distribution of cancer stem cells between enhancing nodule and FLAIR hyperintensity in supratentorial glioblastoma: time to recalibrate the surgical target?

Roberto Altieri, Giuseppe Broggi, Francesco Certo, Daniela Pacella, Giacomo Cammarata, Massimiliano Maione, Marco Garozzo, Davide Barbagallo, Michele Purrello, Rosario Caltabiano, Gaetano Magro, Giuseppe Barbagallo

Summary: This study investigates the expression and distribution of cancer stem cells in glioblastoma. The results show no significant difference in the expression of cancer stem cells between the enhancing nodule and FLAIR hyperintensity zones. This suggests the need to recalibrate the surgical target and consider the use of 5-ALA to improve surgical outcomes.
