Nomograms for colorectal cancer: A systematic review

AIM: To assist in the selection of suitable nomograms for obtaining desired predictions in daily clinical practice. METHODS: We conducted electronic searches for journal articles on colorectal cancer (CRC)-associated nomograms using the search terms colon/rectal/colorectal/nomogram. Of 174 articles initially found, we retrieved 28 studies in which a nomogram for CRC was developed. RESULTS: We discuss the currently available CRC-associated nomograms, including those that predict the oncological prognosis, the short-term outcome of treatments, such as surgery or neoadjuvant chemoradiotherapy, and the future development of CRC. Developing nomograms always presents a dilemma. On the one hand, the desire to cover as wide a patient range as possible tends to produce nomograms that are too complex and yet have C-indexes that are not sufficiently high. Conversely, confining the target patients might impair the clinical applicability of constructed nomograms. CONCLUSION: The information provided in this review should be of use in selecting a nomogram suitable for obtaining desired predictions in daily clinical practice.

Summary: Primary care physicians are expected to prioritize roles in prevention, diagnosis, and palliative care across the cancer care continuum, although some physicians have lower expectations for their involvement in cancer diagnosis and treatment. Expectations for clinic-based general practitioners' involvement in cancer treatment and survivorship care were not as high as anticipated, indicating potential gaps that need to be addressed.

Summary: In this study, risk factors for the pouch not reaching the anus after restorative proctocolectomy were analyzed. Male sex and laparoscopic approach were identified as independent risk factors for non-reaching. Furthermore, a distance between the terminal of the superior mesenteric artery ileal branch and the anus >11 cm was found to be a risk factor for non-reaching. Preoperative CT measurements may be useful in predicting this outcome.

Clinical significance of the KRAS G13D mutation in anastomotic recurrence of colorectal cancer

Summary: This study aimed to investigate the association between the KRAS G13D mutation and anastomotic recurrence (AR) in colorectal cancer (CRC) patients. Results showed that the prevalence of KRAS G13D mutation was significantly higher in the AR group compared to the non-anastomotic local recurrence (NALR) group (AR vs. NALR, 33.3 vs. 4.8%; P=0.047). Furthermore, in the AR group, all patients with KRAS G13D mutation who underwent resection had subsequent recurrence within 2 years after resection, and their overall survival rate was lower (3-year survival rate: Positive vs. negative, 68.6 vs. 90.9%; P=0.02).

Development of fluorogenic substrates for colorectal tumor-related neuropeptidases for activity-based diagnosis

Summary: In this study, a fluorogenic substrate probe was developed to analyze enzymatic activity changes in tissue and plasma samples. The probe was particularly useful for studying enzyme activities in a single-molecule enzyme assay platform. We detected the activity of neurolysin in plasma samples and found higher enzyme activity in the blood samples of colorectal tumor patients. The results suggest that single-molecule neurolysin activity could be a promising blood biomarker for colorectal cancer diagnosis.

PD-L1 expression on tumor cells may be associated with the process of epithelial mesenchymal transition, and quantifying cell type-specific PD-L1 expression using multicolor flow cytometry may provide valuable information for the immunotherapy of solid tumors.
