Summary: This study identified pathogenic variants in the POU1F1 gene that are associated with combined pituitary hormone deficiency and other neurologic phenotypes. The findings highlight the importance of genetic testing in countries with high rates of consanguineous marriage, and contribute to a more rational approach to the clinical management of hypopituitarism.

Summary: Traumatic brain injury (TBI)-related hypopituitarism has been found to be more common than previously thought, affecting pituitary hormones such as growth hormone, gonadotropins, thyroid hormones, etc. The mechanisms of pituitary damage in TBI patients involve primary and secondary injuries, with long-standing growth hormone deficiency leading to neurocognitive and behavioral deficits. Further research is needed to better understand and characterize this clinical syndrome.

Summary: Recovery from AGHD after initial TSS is more likely in younger female patients with higher peak GH levels assessed by preoperative PPT. Patients aged <= 62.2 years and with a peak GH level of >= 0.74 μg/L based on preoperative PPT are likely to recover from AGHD.

Summary: Adult patients with growth hormone deficiency (GHD) exhibit individual sensitivity to recombinant human growth hormone (rhGH) replacement therapy, which is influenced by factors such as age, sex, body mass index, baseline GH status, and quality of life. Individualized dose-titration strategies have been shown to improve efficacy and reduce adverse events in the treatment of GHD. Efforts to develop predictive mathematical models to distinguish good and poor responders to rhGH therapy have not yet been prospectively tested in a large cohort of adult GHD individuals.

Summary: This study aimed to estimate the national incidence of congenital combined pituitary hormone deficiency (cCPHD) diagnosed before the age of 18, and in subgroups. Data from Danish registries revealed an estimated national incidence of cCPHD of 10.34 per 100,000 births, with an annual incidence rate of 5.74 per million. The incidence was highest in the 1-17 years age group, highlighting the importance of investigating multiple pituitary hormones throughout childhood and adolescence in children with only one hormone deficiency.

Summary: Congenital hypopituitarism (CH) is a disorder characterized by deficiency in one or more pituitary hormones. The incidence is 1 in 4,000 to 1 in 10,000, with the majority of cases being sporadic. CH pathogenesis involves mutations in various genes such as HESX1, PROP1, and POU1F1. Clinical manifestations range from severe midline developmental disorders to isolated hormone deficiencies.

Summary: This review provides an overview of newly reported causative genes for combined pituitary hormone deficiency (CPHD), as well as discussing novel variants of known CPHD-related genes. Clinical and basic research may help in inferring the causative genes of CPHD, and future analysis may reveal new genes related to pituitary development.

Summary: The value of the TRH test in diagnosing CH in patients with pituitary disease was evaluated. Patients diagnosed with CH had lower T4(corr) and TSH increase compared to patients without CH. In some patients with low T4(corr), the CH diagnosis was rejected, and a spontaneous increase in T4(corr) was observed during follow-up. CH patients reported more symptoms of hypothyroidism. The TRH test did not provide relevant clinical information in patients with normal T4 or patients awaiting pituitary surgery.

Summary: This study evaluated the clinical safety and efficacy of mesenchymal stem cells (MSCs) in sepsis and septic shock patients. The results suggest that MSCs treatment may have a positive impact on the survival rates of sepsis during the early phase, but further randomized controlled studies with a large group of patients are needed to confirm these findings.

Summary: Anthrax is an important zoonotic disease caused by Bacillus anthracis. It primarily infects herbivores but can also infect humans. The disease can be contracted from infected animals, contaminated animal products, or exposure to spores in the environment. Diagnosis requires bacterial culture or molecular testing due to the similarity of early symptoms with other diseases.
