Antiproliferative Effect of Benzophenones and their Influence on Cathepsin Activity

The antiproliferative activity of two prenylated benzophenones isolated from Rheedia brasiliensis. the tri-prenylated garciniaphenone and the tetraprenylated benzophenone 7-epiclusianone, was investigated against human cancer cell lines. The antiproliferative activity on melanoma (UACC-62), breast (MCF-7), drug-resistant breast (NCI-ADR), lung/non-small cells (NCI460), ovarian (OVCAR 03), prostate (PC03), kidney (786-0), lung (NCI-460) and tongue (CRL-1624 and CRL-1623) cancer cells was determined using spectrophotometric quantification of the cellular protein content. The effect of these benzophenones on the activity of cathepsins B and G was also investigated. Garciniaphenone displayed cytostatic activity in all cell lines, whereas 7-epiclusianone showed a dose-dependent cytotoxic effect. The IC50 values for cell proliferation revealed that 7-epiclusianone is more active than garciniaphenone against most of the cell lines. Furthermore, the antiproliferative effects demonstrated by garciniaphenone and 7-epiclusianone were related to their cathepsin inhibiting properties. In conclusion, 7-epiclusianone is a promising naturally occurring agent which displays multiple inhibitory effects which may be working in concert to inhibit cancer cell proliferation in vitro. The putative pathway by which 7-epiclusianone affects cancer cell development may involve cathepsin inhibition. Copyright (C) 2009 John Wiley & Sons, Ltd.

A Systematic Review of the Potential Effects of Propolis Extracts on Experimentally-induced Diabetes

Summary: This systematic review evaluated the impact of propolis administration in diabetic animals. The study found that propolis administration resulted in a hypoglycemic effect in the treated animals, which may be attributed to the reduction of oxidative stress. Additionally, propolis administration restored endogenous antioxidant defenses and improved the lipid profile of the treated animals. However, the review also identified limitations in the studies, including a lack of randomization, blinding, and proper allocation of animals.

Summary: This study predicts the bioaccessibility and intestinal permeability of major phenolics in peanuts using simulated digestion and Caco-2 cell model. The results show that the phenolics in peanuts undergo changes during digestion, but their antioxidant activity remains unchanged. Furthermore, peanuts are found to contain p-coumaric acid as a major phenolic compound.

Summary: This study aimed to investigate the impact of gastrointestinal digestion/Caco-2 cell transport on the biological activities and toxicity of ethanolic extract of organic propolis from southern Brazil (EEOP1). The results demonstrated that EEOP1 deactivated reactive oxygen species and attenuated the activation of transcription factors, while also reducing the release of pro-inflammatory cytokines. Additionally, the phytochemical profile of EEOP1 changed after digestion, and antimicrobial activity was only observed in the non-digested form.

Summary: The effect of dynamic gastrointestinal digestion/Caco-2 cell transport on the stability and activities of Brazilian red propolis extract (EEBRP) was investigated. Encapsulated EEBRP showed higher bioaccessibility and nonencapsulated EEBRP had higher Caco-2 cell transport. Nonencapsulated EEBRP exhibited higher ability to scavenge reactive oxygen species and decrease inflammation markers after cell transport. Further studies on encapsulation are encouraged for improving bioactivity and expanding food applications.

Cocoa powder and fermented jackfruit seed flour: A comparative cell-based study on their potential antioxidant and anti-inflammatory activities after simulated gastrointestinal digestion

Summary: The in vivo toxicity and in vitro antioxidant activity of fermented jackfruit seed flour (Fjs) and non-alkaline cocoa powder (Nac) were investigated in this study. The results showed that there were no significant differences in the antioxidant activity of Nac-D and Fjs-D after simulated gastrointestinal digestion. Furthermore, both Fjs and Nac exhibited beneficial effects in terms of cellular antioxidant activity and anti-inflammatory assay.

Evaluation of the Quality and Antioxidant Activity of Dehydrated Medicinal Herbs

Summary: This study evaluates the effects of drying methods on the drying rates, characteristics, and antioxidant properties of five herbs. Forced-air oven drying resulted in higher drying rates compared to solar drying. Mint was less affected by drying conditions, while all dried herbs exhibited similar antioxidant properties. Standardization of post-harvest steps is important for maintaining the consistency of herbal products.

Germination and its role in phenolic compound bioaccessibility for black mustard grains: A study using INFOGEST protocol

Summary: Germination has been found to improve the antioxidant properties of mustard by increasing the bio-accessibility of phenolic compounds. In this study, the in vitro digestion process was simulated to evaluate the impact of germination on mustard phenolic bio-accessibility. Liquid chromatography-mass spectrometry analysis identified resveratrol, formononetin, and cryptochlorogenic acid as key phenolic compounds. Results showed that digestion positively affected the antioxidant potential of soluble phenolics, although it appeared to negate the improvement caused by germination. However, germinated digested mustard had significantly higher concentrations of sinapic acid compared to non-germinated.
