Phytochemical studies on themethanolic extract of Drypetes gossweileri afforded N-beta-D-glucopyranosyl-p-hydroxyphenylacetamide (1), p-hydroxyphenylacetic acid (2), p-hydroxyphenyl-acetonitrile (3), phydroxyacetophenone (4), 3,4,5-trimethoxyphenol (5), dolichandroside A (6), and beta-amyrone (7). Compounds 1-7 were identified with the aid of extensive NMR and MS spectroscopic studies. Compound 1 was a new natural product and was isolated for the first time from plant containing N-glucose moiety incorporated in its structure. Compounds 1-7 exhibited moderate to the weak source anti-alpha-glucosidase inhibitory activity. Compound 7 exhibited moderate anti-acetylcholinesterase (AChE) activity while the rest of the isolates were weakly active in this bioassay. Compounds 1-7 also showed moderate antifungal activity. Summary: Researchers used a cell-free expression workflow to screen and optimize constructs containing computationally designed miniprotein inhibitors of SARS-CoV-2. They found that a homotrimeric version of the ACE2 mimic AHB2 (TRI2-2) achieved the broadest efficacy. The TRI2-2 miniprotein neutralized Omicron, Delta, and other variants with greater potency than clinically used monoclonal antibodies. It also provided prophylactic and therapeutic protection against SARS-CoV-2 in mice when administered intranasally. The designed miniprotein receptor mimics offer a widely applicable antiviral therapeutic strategy with advantages over antibodies and native receptor traps.

Iridoid and lignan glycosides from Lancea tibetica

Yi Zhu, Junlin Wang, Lu Tang, Yuxin Shang, Weiyi Zhao, Wei Zhang, Ping Hai, Yongbiao Zheng, Wenbo Li, Yanfang Su

Summary: One new iridoid glucoside and two new lignan glycosides, along with three known compounds, were isolated from the whole plants of Lancea tibetica. The structures were determined by spectroscopic methods and X-ray crystallographic analysis. One of the new iridoid glucosides and the known compounds exhibited moderate antioxidant activity.
