Nine clerodane diterpenes, solidagoic acids C-I (1-7), cleroda-3,13(14)-dien-16,15:18,19-diolide (8) and cleroda-3,13(14)-dien-15,16:18,19-diolide (9) were isolated and characterised from the ethanol-ethyl acetate (1:1) extract of Solidago virgaurea. The structures were determined by NMR spectroscopic analysis. Several displayed moderate antibacterial activity against Staphylococcus aureus. (C) 2009 Elsevier Ltd. All rights reserved.

Summary: The combination of HPLC-based activity profiling and advanced UHPLC-HRMS/MS metabolite profiling allows for the identification of highly active compounds in crude extracts, which can be beneficial to the drug discovery process. This strategy was successfully applied to a subset of extract library screening, leading to the identification and isolation of potent natural products inhibiting oncogenic signaling in melanoma.

Summary: In this study, we found that regular consumption of capsaicin can change the structure of the gut microbial community and increase the abundance of certain short-chain fatty acids, particularly butanoic acid. These changes may be responsible for the health benefits associated with capsaicin consumption.

Flavonoids from Ericameria nauseosa inhibiting PI3K/AKT pathway in human melanoma cells

Summary: The PI3K/AKT and MAPK/ERK pathways are frequently mutated in metastatic melanoma. In this study, a plant extract from Ericameria nauseosa was found to inhibit the oncogenic activity of AKT in human melanoma cells. Further analysis led to the identification of several active compounds, including flavones, which were more potent than flavanones. Two new natural products, diterpenes, were also discovered in this extract.

A Dimerosesquiterpene and Sesquiterpene Lactones from Artemisia argyi Inhibiting Oncogenic PI3K/AKT Signaling in Melanoma Cells

Summary: A screening of over 2500 plant extracts in melanoma cells revealed that the ethyl acetate extract from Artemisia argyi displayed significant inhibition of the PI3K/AKT pathway. Through activity profiling, 21 active compounds were isolated, including one novel dimerosequiterpenoid, one new disesquiterpenoid, three new guaianolides, 12 known sesquiterpenoids, and four known flavonoids.

Summary: This study investigated the roots of Operculina hamiltonii, a traditional medicinal plant in Brazil. Three new acyl sugars or resin glycosides were identified and their structures elucidated. The combination of these compounds with other drugs showed enhanced cytotoxic effects on breast cancer cells.

Summary: A new naturally occurring compound, Linderagatins C-F (1-4), with the structures of diaryltetrahydrofuran-type 7,9'-dinorlignans, was discovered in the roots of Lindera aggregata. The neuroprotective effects of compounds 2 and 3 on erastin-induced ferroptosis in HT-22 cells were found to be significant.
