Background Data and Objective: Herpes is a common infectious disease that is caused by human herpesviruses. Several treatments have been proposed, but none of them prevent reactivation of the virus. This article describes the use of photodynamic therapy (PDT) as a treatment for herpes lesions, and reports on four cases. Materials and Methods: PDT was used as an adjuvant therapy for the treatment of herpes labialis in four patients. A special type of 0.01% (m/V) of methylene blue solution was applied to the vesicular stage of herpesviral disease and the lesions were irradiated with laser energy (wavelength 660 nm, energy density 120 J/cm(2), output power of 40 mW, 2 min per point, 4.8 J of energy/point, at four points). After 24 h the patients returned and phototherapy was repeated with the same equipment, this time with 3.8 J/cm(2) and 15 mW, for a total dose of 0.6 J. The same procedure was repeated 72 h and 1 wk later. Results: Treatment with low-level laser therapy can be considered as an option in the treatment of herpes labialis, and decreases the frequency of vesicle recurrence and provides comfort for patients. No significant acute side effects were noted and the lesions healed rapidly. Conclusion: Treatment of herpes labialis with PDT was effective, had no side effects, and when associated with laser phototherapy, accelerated the healing process.

Efficacy of a calcium-phosphate/fluoride varnish and ionomeric sealant on cervical dentin hypersensitivity: A randomized, double-blind, placebo-controlled clinical study

Alana Cristina Machado, Vinicius Maximiano, Mirian Lumi Yoshida, Julia Gomes Freitas, Fausto Medeiros Mendes, Ana Cecilia Correa Aranha, Tais Scaramucci

Summary: This study compared the efficacy of two treatments for dentin hypersensitivity, showing that the ionomeric sealant (IS) was the most effective in the short term and also had better long-term results compared to Ca/PO4(-)/F- varnish. The results indicated that IS can significantly reduce DH symptoms, while the varnish did not show significant differences compared to the placebo.

The importance of combining methods to assess Candida albicans biofilms following photodynamic inactivation

Gabriela Alves da Collina, Fernanda Viana Cabral, Carolina Montovam Monteiro, Gabriela Benedito Machado, Jose Marcelo Lacerda Alves Goncalves, Fernanda Freire, Renato Araujo Prates, Martha Simoes Ribeiro, Christiane Pavani

Summary: This study evaluated the photodynamic inactivation (PDI) effects of an oral formulation (OF) containing 0.005% methylene blue (MB) on Candida albicans biofilm using multiple methodologies. The results showed that PDI mediated by MB-OF significantly reduced the colony forming units (CFU) and virulence of the biofilm, but did not have a significant impact on metabolic activity. The crystal violet (CV) staining data were inconclusive due to interference from the components of the formulation.

Treatment of oral leukoplakia with CO2 laser (10,600 nm): analysis of 37 cases

Wladimir Gushiken de Campos, Camilla Vieira Esteves, Camila de Barros Gallo, Carina Domaneschi, Ana Cecilia Correa Aranha, Celso Augusto Lemos

Summary: This study evaluated the outcomes of CO2 laser treatment for oral leukoplakia and found that the treatment was effective in removing the lesions but did not completely prevent relapse or malignant transformation. Among the 37 patients in the study, 13 experienced recurrence of leukoplakia and 8 had malignant transformation.

The Biochemical Mechanisms of Antimicrobial Photodynamic Therapy1

Caetano P. Sabino, Martha S. Ribeiro, Mark Wainwright, Carolina Dos Anjos, Fabio P. Sellera, Milena Dropa, Nathalia B. Nunes, Guilherme T. P. Brancini, Gilberto U. L. Braga, Victor E. Arana-Chavez, Raul O. Freitas, Nilton Lincopan, Mauricio S. Baptista

Summary: The uncontrolled spread of multidrug-resistant pathogens is a major global health threat. Antimicrobial photodynamic therapy (aPDT) offers a promising approach to treating localized infections without being limited by drug resistance. In this study, various techniques were used to investigate the biochemical mechanisms of aPDT. The results suggest that protein degradation correlates with microbial inactivation kinetics, providing important insights for the development of optimized photosensitizing drugs and combination therapies with antimicrobial chemotherapy.

Could Light-Based Technologies Improve Stem Cell Therapy for Skin Wounds? A Systematic Review and Meta-Analysis of Preclinical Studies

Tania M. Yoshimura, Fernanda Cabral, Fabio P. Sellera, Lorena Pozzo, Martha S. Ribeiro

Summary: This article systematically reviews the impact of combining stem cell and photobiomodulation therapy (PBM) in the repair of skin wounds. The meta-analysis shows that the combination of SCs and PBM has a positive influence on wound closure and strength, although there is heterogeneity and bias in the included studies.

A Novel Strategy Based on Zn(II) Porphyrins and Silver Nanoparticles to Photoinactivate Candida albicans

Bruno L. Raposo, Sueden Souza, Gleyciane S. Santana, Max T. A. Lima, Jose F. Sarmento-Neto, Julio S. Reboucas, Goreti Pereira, Beate S. Santos, Paulo E. Cabral Filho, Martha S. Ribeiro, Adriana Fontes

Summary: Researchers proposed a novel method of using polyvinylpyrro-lidone (PVP) coated silver nanoparticles (AgNPs) in combination with cationic Zn(II) porphyrins (ZnPs) for photodynamic inactivation (PDI) of Candida albicans infections. The AgNPs-ZnPs system achieved complete eradication of Candida albicans at lower porphyrin concentrations, reducing the risk of antifungal drug resistance.

Methylene blue-mediated antimicrobial photodynamic therapy on chicken semen

G. A. Novaes, M. H. Blank, T. M. Yoshimura, M. S. Ribeiro, R. J. G. Pereira

Summary: Artificial insemination in poultry often leads to high levels of bacterial contamination in the semen. This study explored the potential use of antimicrobial photodynamic therapy (PDT) as an alternative to antibiotics in preserving semen quality. The results showed that incubation with methylene blue (MB) did not affect sperm motility, but increasing light dose during the PDT process resulted in greater reductions in sperm parameters. However, the conditions that were less harmful to spermatozoa were not effective in reducing bacterial colonies in chicken semen.

Summary: This study investigated the potential of polymeric hydrogels in the treatment of melanoma. The researchers found that hydrogels containing both a nitric oxide donor and copper oxide nanoparticles significantly reduced cell viability and increased reactive oxygen species levels. They also observed changes in cell nuclei, loss of mitochondrial membrane potential, and lipid peroxidation, leading to cell death.

New Insights in Phenothiazinium-Mediated Photodynamic Inactivation of Candida Auris

Abdenego R. Silva, Fernanda V. Cabral, Camila R. Silva, Daniela F. T. Silva, Anderson Z. Freitas, Adriana Fontes, Martha S. Ribeiro

Summary: This study evaluated the impact of photodynamic inactivation mediated by methylene blue (MB-PDI) or 1,9-dimethyl MB (DMMB-PDI) combined with a red LED against the hospital-acquired pathogen C. auris. DMMB-PDI successfully eradicated C. auris, while MB only killed cells when exposed to the highest light dose. DMMB-PDI promoted higher levels of reactive oxygen species, lipid peroxidation, and mitochondrial membrane potential compared to MB. Additionally, DMMB-PDI was able to inhibit biofilm formation and destroy mature biofilms, with no toxicity observed in fibroblasts. We conclude that DMMB-PDI holds great potential to combat the global threat posed by C. auris.
