Mangiferin ameliorates 6-hydroxydopamine-induced cytotoxicity and oxidative stress in ketamine model of schizophrenia

Background: Accumulating evidence indicates that mangiferin (MGF), a natural xanthone, by virtue of its antioxidant and anti-inflammatory properties is neuroprotective. Here we sought to verify the cytoprotective role of MGF on cultured rat primary mesencephalic cells exposed to 6-hydroxydopamine (6-OHDA) in vitro, and the MGFs anti-inflammatory potential in mouse model of ketamine-induced schizophrenia in vivo. Methods: 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT)-assay was performed to measure cell viability in mesencephalic cell cultures exposed to neurotoxin (6-OHDA, 40 mu M). Schizophrenia was induced in mice by ketamine (50 mg/kg, ip, twice a day, for 7 days). The treatment effects of MGF (50 mg/kg,po, for 7 days) were verified on locomotor behavioral changes in open-field test, and on the oxidant stress-related increase in lipid-peroxidation (malondialdehyde) and interleukin-6 (IL-6) levels in brain tissues. Results: MGF (10-100 mu M) produced no per se effect on cell viability as measured by MTT assay, but significantly prevented the 6-OHDA-induced cell death in a concentration-dependent manner. Acridine orange/ethidium bromide (AO/EtBr) staining confirmed the absence of 6-OHDA-induced morphological changes characteristic of apoptosis/necrosis. In open-field test, ketamine-induced impaired locomotor activity and behavioral changes such as grooming and stereotyped but not rearing were effectively ameliorated by MGF pretreatment. Also, ketamine-associated increase in brain tissue levels of IL-6 and MDA were significantly lowered in MGF-pretreated mice. Conclusion: Mangiferin has a neurocytoprotective role related, at least in part, to an antioxidant and anti-inflammatory mechanism, which could be explored for more effective therapies of schizophrenia and other neurodegenerative diseases.

This study focused on the bioactivity of the flowers of Senna cearensis, a native species in northeast Brazil. These findings contribute to the understanding of the bioactive compounds in Senna cearensis and its potential medicinal uses.

Summary: This study isolated and characterized a thermostable trypsin inhibitor from chia seeds, which showed antibacterial activity against Staphylococcus aureus sensitive and resistant to methicillin. The trypsin inhibitor, named ShTI, displayed competitive inhibition and 1:1 stoichiometry ratio for the ShTI:trypsin complex. ShTI exhibited antibacterial activity alone and in combination with oxacillin, and its antibacterial action involved overproduction of reactive oxygen species and plasma membrane pore formation.

Farmers Views on the Implementation of On-Farm Emergency Slaughter for the Management of Acutely Injured Cattle in Ireland

Summary: Four options exist for the management of acutely injured cattle in Ireland: treatment, on-farm emergency slaughter (OFES), casualty slaughter (CS), or euthanasia. A survey of farmers revealed that most of them had a positive view of OFES, but there were concerns about the availability and cost of this option. Increasing availability and reducing cost of OFES could help decrease the number of cattle going for CS.

Anti-inflammatory kaurane diterpenoids of Erythroxylum bezerrae

Summary: Five new kaurane diterpenes (bezerraditerpenes A-E) and six known ones were isolated from the stems of Erythroxylum bezerrae. The structures of these compounds were determined by various spectroscopic methods. The anti-inflammatory potential of these diterpenes was evaluated on murine macrophage-like cells RAW 264.7, showing significant activity for compounds 1, 2, and 8 without cytotoxicity.

Activity of amlodipine against Staphylococcus aureus: association with oxacillin and mechanism of action

Summary: The aim of this study was to evaluate the in vitro antimicrobial activity of amlodipine against Staphylococcus aureus strains. The antimicrobial activity of amlodipine was evaluated using the broth microdilution method, and its interaction with oxacillin was evaluated using a checkerboard assay. The possible mechanism of action was evaluated using flow cytometry and molecular docking techniques. Results showed that amlodipine exhibited antibacterial activity against S. aureus and had synergistic effects with approximately 58% of the strains used. Amlodipine also showed good activity against forming and mature biofilms. The possible mechanism of action may be attributed to its ability to induce cell death.

Antibacterial activity of paroxetine against Staphylococcus aureus and possible mechanisms of action

Summary: This study evaluated the antibacterial activity of paroxetine alone and in combination with oxacillin against methicillin-sensitive and -resistant Staphylococcus aureus isolates. The results showed that paroxetine had bactericidal activity at a minimum inhibitory concentration (MIC) of 64 μg/ml, and exhibited mostly additive interactions when combined with oxacillin. Mechanism studies revealed the action of paroxetine on genetic material and membrane, as well as morphological changes in microbial cells and influence on virulence factors.

Sertraline has in vitro activity against both mature and forming biofilms of different Candida species

Summary: Candida spp. infections are a serious health problem, and drug repurposing, such as using sertraline, has shown potential in treating Candida spp. biofilm infections. The present study evaluated the in vitro antibiofilm activity of sertraline and found that it effectively reduced the viability of forming and matured biofilms of all Candida species tested. Sertraline acted at low and safe concentrations, and its preventive mechanism seemed to be related to binding with ALS3. These findings suggest that sertraline could be a promising drug for treating Candida biofilm infections, but further research is needed.

Synthesis, characterization and molecular docking study of pyrazolines synthesized from chalcones: Antioxidant and acetylcholinesterase activities

Summary: Pyrazolines are heterocyclic compounds with nitrogen-containing rings known for their pharmacological potential. This study synthesized thirteen pyrazoline derivatives with total yields of 56 - 90% and characterized them using various spectroscopic techniques. The compounds were evaluated for antioxidant activity and acetylcholinesterase inhibition, showing varying levels of activity. Molecular docking studies revealed two compounds with excellent binding affinity and pose.

Summary: The objective of this study was to explore the associations between mortality in cSLE patients and their characteristics, as well as to evaluate the risk factors and determine the main causes of death. The results showed that the mortality rate in cSLE patients in Brazil was low, but neuropsychiatric lupus and chronic kidney disease were found to be significant risk factors for mortality.

Summary: This study tested a drug delivery system based on chitosan microparticles loaded with geranium and lemongrass essential oils to inhibit the mixed biofilms of Candida albicans and Streptococcus mutans. The results showed that this system had good inhibitory effects on planktonic activity and reduced the metabolic activity of the biofilms.
