Functional expression of the oligopeptide transporter PepT1 from the sea bass (Dicentrarchus labrax)

Complementary RNA, derived from the intestine of the sea bass Dicentrarchus labrax and putatively coding for a pH-dependent oligopeptide transporter PepT1 (SLC15 family), was injected in Xenopus oocytes that were subsequently tested with electrophysiological techniques. Transport-associated currents were observed when various di- or tripeptides were applied at concentrations ranging between 0.1 and 10 mM. No currents were generated by histidine nor by other single amino acids. Sea bass PepT1 also exhibited presteady-state currents in the absence of substrates. Acidic pH slowed down the relaxation time constant of these currents and shifted both Q/V and tau/V relationships toward more positive voltages. Michaelis-Menten analysis of the transport currents showed an increase in apparent substrate affinity at acidic pH, which was very similar to that exhibited by the related transporter from zebrafish (Danio rerio), but in contrast, did not demonstrate a significant effect of pH on the maximal transport current.

Functional characterization of Atlantic salmon (Salmo salar L.) PepT2 transporters

Summary: Two slc15a2-type genes were identified in the Atlantic salmon genome, encoding PepT2-type peptide transporters. While slc15a2a transcripts are widely distributed in fish tissues, abundant in the brain and gills, slc15a2b transcripts are mainly expressed in the kidney and distal gastrointestinal tract. Both PepT2a and PepT2b function as high-affinity transporters.

Betaine-The dark knight of the brain

Summary: The role and function of betaine in the brain have been little studied and reviewed, despite its well-documented effects in the liver and kidney. This review analyzes the beneficial effects of betaine in neurodegeneration, excitatory and inhibitory imbalance, and oxidative stress in the central nervous system. However, there are still many unknown aspects regarding its transport and the actions beyond the known protein, betaine-GABA transporter 1. This article highlights the lack of detailed molecular and cellular information and the importance of studying the interaction of betaine with CNS components for its therapeutic use.

Bile acid interactions with neurotransmitter transporters

Summary: Bile acids (BAs) synthesized from cholesterol in the liver play a key role in emulsifying fats for absorption. Recent evidence suggests that BAs can modulate the activity of neuronal receptors and transporters, including the dopamine transporter (DAT), involved in gut-brain signaling. This study investigated the effects of BAs on three transporters and found that obeticholic acid (OCA), a semi-synthetic BA, generated an inward current in DAT, GABA transporter 1 (GAT1), and glycine transporter 1 (GlyT1b), proportional to the current generated by their respective substrates. The findings support a molecular model suggesting the ability of BAs to occlude the transporter, avoiding small depolarizations and enhancing neurotransmitter action.

This review aims to compare GAT1 and BGT-1, leveraging the knowledge of GAT1 to shed light on the unanswered questions regarding BGT-1.

Summary: The transport capability of charged dipeptides in teleosts is related to the position of the charged amino acid. In Atlantic salmon, the transport currents of PepT1a and PepT1b are similar, while in zebrafish, PepT1a is highly dependent on the position of Asp and PepT1b has low transport currents.

How Different Dietary Methionine Sources Could Modulate the Hepatic Metabolism in Rainbow Trout?

Summary: In this study, a feeding trial was conducted with rainbow trout to evaluate the bioavailability and efficacy of five different forms of methionine. The results showed that different forms of methionine had no effects on growth performance but led to changes in the whole-body composition of the fish. Additionally, specific forms of methionine affected hepatic methionine metabolism through different molecular pathways.
