Summary: The role of neurotrophic factors BDNF and GDNF in maintaining brain resistance to hypoxia and promoting recovery of neural networks is being actively studied. This study aimed to evaluate the effects of overexpressing BDNF and GDNF using viral vectors in modeling hypoxia. The results showed increased survival rate of mice under hypoxic conditions, indicating that overexpression of neurotrophic factors may be a potential neuroprotection method.

Summary: This study aimed to investigate the expression of glial-derived neurotrophic factor (GDNF) and its receptors, GFR alpha 1 and GFR alpha 2, ciliary neurotrophic factor (CNTF), and its receptor CNTFR alpha, in both normal and glaucomatous human tissue. The results showed that the expression of GFR alpha 2 was significantly decreased in glaucomatous eyes, while the expression of GFR alpha 1 was decreased in the peripapillary region. The findings suggest that the decreased expression of GDNF receptors in glaucomatous tissue may limit the potential for neuroprotective therapy using GDNF supplementation.

Summary: The neuromuscular junction (NMJ) is a crucial interface between the nervous system and muscles, where the Glial cell line-Derived Neurotrophic Factor (GDNF) plays a key role in synaptic formation, stabilization, and function, as well as in protecting the NMJ during neurodegeneration. GDNF also has unique properties that make it essential in neurodegenerative disorders. Research highlights the importance of GDNF in the NMJ-muscle interaction, and its therapeutic potential in diseases like Alzheimer's, ALS, and SMA. Additionally, 3D neuromuscular co-culture systems are being developed as models to study NMJ formation and involvement in neuromuscular disorders.

Summary: This review focuses on the in vivo regulation of spermatogonial stem cells (SSCs) in adult testes by glial cell line-derived neurotrophic factor (GDNF). Through chemical-genetic approach, the inhibition of GDNF stimulation of SSCs was found to decrease replication and increase differentiation of SSCs, ultimately leading to the loss of all SSCs and resembling the condition of Sertoli cell-only (SCO) syndrome. Abnormally low expression of GDNF, FGF2, and FGF8 was observed in infertile human testes, although some SCO testes still contain SSCs. Restoration of GDNF signaling showed partial rebuilding of depleted SSC pool, suggesting GDNF as a potential therapy for men with SCO syndrome.

Summary: This study investigated the differences in serum levels of brain-derived neurotrophic factor (BDNF) and glial cell line-derived neurotrophic factor (GDNF) and clinical symptoms in first-episode depression patients at different ages. The results showed that serum BDNF and GDNF levels were lower in the study group and subgroups compared to the healthy control groups. Additionally, there were differences in serum BDNF levels among different age groups of patients, suggesting the potential of BDNF and GDNF as biomarkers for depression.

Summary: This study achieved continuous overexpression of GDNF in immune-privileged human adipose tissue-derived mesenchymal stromal cells using a non-viral Sleeping Beauty transposon. Although GDNF was detectable shortly after cell transplantation, the 6-OHDA model used was too mild to evaluate disease improvement. Further research using a more appropriate 6-OHDA model is needed to test the therapeutic potential of GDNF-overexpressing cells.

Summary: Through RNA sequencing and tissue-specific MAPK/ERK inactivation experiments, we found that MAPK/ERK signaling maintains ureteric bud tip cells, potentially playing a regulatory role in collecting duct progenitors. Additionally, we provide new mechanistic insights on how MAPK/ERK signaling regulates progenitor maintenance through its effects on chromatin accessibility and energy metabolism.
