Summary: Cutaneous leiomyosarcoma (cLMS) is a rare soft tissue sarcoma with unclear genetic drivers. The presence of cLMS in Birt-Hogg-Dube (BHD) syndrome suggests that FLCN loss may be involved in cLMS pathogenesis. This report provides novel genetic evidence of FLCN loss in BHD cLMS, indicating that cLMS is a sporadic but recurring element of the BHD phenotype.

Summary: This study evaluates the recurrence and survival of primary cutaneous leiomyosarcoma (cLMS) patients through analyzing their clinicopathologic and treatment characteristics. The results show that primary tumor resection provides excellent local control for low-grade tumors, while patients with intermediate- to high-grade tumors may experience local recurrence, distant metastasis, and death.

Summary: Cutaneous sarcomas are rare but potentially fatal tumors originating from mesenchymal cells in the skin. Guidelines for management exist for soft-tissue sarcomas, but not for cutaneous sarcomas. This review summarizes the available literature on management of seven different types of cutaneous sarcomas and aims to provide practical recommendations for their recognition, management, and follow-up. The goal is to support clinicians in providing the best evidence-based care for these rare but important tumors.

Summary: There is currently no standard adjuvant treatment protocol for localized uterine leiomyosarcoma (uLMS). This study aims to determine the benefit of adjuvant chemotherapy for uLMS. The results showed that patients who received any form of adjuvant therapy had a slightly longer disease-free survival, but the difference was not significant. Tumor size and mitotic rate were identified as independent prognostic factors for worse outcomes. Therefore, the need for adjuvant therapy for resected uLMS remains in question.

Summary: Cutaneous leiomyosarcoma is a rare malignant neoplasm of muscular origin, representing a small percentage of all cutaneous soft tissue sarcomas. This study analyzed 11 cases of cutaneous leiomyosarcoma over a 20-year period, with most cases occurring in men and primarily located in the head and neck region. Immunohistochemical staining played a crucial role in diagnosis and all cases were treated surgically, with a mean survival of 32.2 months.

This study presents a case of PILMS secondary to glioblastoma, highlighting the importance of aggressive multimodal treatment with surgery and radiotherapy for PILMS. Further research, including systemic treatment options, is needed to address the unanswered questions surrounding PILMS.

Summary: This study investigates the in vivo activity of a novel MAP2K4 inhibitor, PLX8725, against uLMS harboring MAP2K4 gene amplification. The results demonstrate that PLX8725 significantly inhibits tumor growth and prolongs survival in mice. PK studies also show that oral administration of PLX8725 provides extended exposure. Ex vivo experiments show that PLX8725 decreases levels of phosphorylated-ATR, JNK, and p38, and increases expression of apoptotic molecules.

Summary: Cutaneous leiomyosarcomas, malignant mesenchymal tumors originating from smooth muscle, occasionally occur in avian species. This case report describes a 14-year-old White Carneau pigeon presenting with a cervical soft tissue mass, diagnosed as leiomyosarcoma based on histopathology and immunohistochemistry. Postoperative examination revealed metastatic foci in the lungs and liver, emphasizing the aggressive behavior of cutaneous LMS in avian species.
