Early detection and estimation of infection burden by real-time PCR in rats experimentally infected with Clonorchis sinensis

A real-time PCR assay targeting the cytochrome c oxidase gene of Clonorchis sinensis was developed and its diagnostic value was investigated in experimentally infected rats. The PCR assay amplified as little as 0.1 pg of C. sinensis genomic DNA and a single egg from experimentally spiked negative feces. The assay could detect C. sinensis DNA in feces 2 weeks post-infection (PI), whereas the Kato-Katz method and formalin-ether concentration techniques could detect the eggs in the feces from 3 weeks PI. Inverse correlations between eggs per gram of feces (EPG) and Ct values were observed 4, 6, and 8 weeks PI. Worm burden was positively correlated with EPG but negatively correlated with Ct values in different groups at 8 weeks PI. Fecal real-time PCR provides a potential tool for the diagnosis of clonorchiosis before eggs can be detected by conventional methods, and may be used to estimate the worm burden in the host.

Summary: Saliva can be accurately used for early diagnosis of SARS-CoV-2 in clinical and community settings. The sensitivity and specificity of saliva samples were 80.3% and 99.4%, respectively. Higher detection rates were observed in saliva samples when collected within 1 to 5 days of symptom onset.

Summary: In Bangladeshi Mtb isolates, mutations in the gyr genes at codons A90V and D94G were found to be the main drivers of resistance to fluoroquinolones. A90V mutations were associated with lower levels of resistance, while D94G mutations were associated with high levels of resistance to all fluoroquinolones.

Summary: In this study, the diagnostic performances of different test methods for tuberculosis diagnosis using BAL fluid specimens were evaluated. Summary: Accurate and appropriate diagnosis of extrapulmonary tuberculosis (EPTB) remains challenging. This study investigated the diagnostic performance of different methods for EPTB diagnosis and found that the Xpert MTB/RIF assay showed the highest sensitivity. Considering its short turnaround time and promising findings, the Xpert MTB/RIF assay should be integrated into national TB guidelines as a routine diagnostic test.

Programmatic Implications for Schistosomiasis Elimination Based on Community-Based Survey in the Blue Nile, North Kordofan, and Sennar States, Sudan

Summary: Despite repeated mass drug administration interventions, the prevalence of schistosomiasis remains high in certain areas due to reinfection. A community-based survey in Sudan revealed that individuals without latrines in their households or with open defecation had a higher risk of schistosomiasis infection. Installing improved latrines and eliminating open defecation should be emphasized in schistosomiasis elimination projects in high-transmission areas.

Citation Activity of Journal of Korean Medical Science in 2011-2020: Reflection on the Most and Least Cited Items

Summary: This article analyzes the citation patterns of articles published in the Journal of Korean Medical Science (JKMS) from 2011 to 2020. The results show that most articles are cited within 5 years of publication, with a noncitation rate of 1.4%. The study suggests that inviting review articles in clinical sciences, research reports on hot medical topics, and nationwide database analyses may attract more author interest and citations.
