Differential identification of Entamoeba spp. based on the analysis of 18S rRNA

Entamoeba histolytica is known to cause intestinal and extra-intestinal disease while the other Entamoeba species are not considered to be pathogenic. However, all Entamoeba spp. should be reported when identified in clinical samples. Entamoeba polecki, Entamoeba coli, and Entamoeba hartmanii can be differentiated morphologically from E. histolytica, but some of their diagnostic morphologic features overlap. E. histolytica, Entamoeba dispar, and Entamoeba moshkovskii are morphologically identical but can be differentiated using molecular tools. We developed a polymerase chain reaction (PCR) procedure followed by DNA sequencing of specific regions of 18S rRNA gene to differentiate the Entamoeba spp. commonly found in human stools. This approach was used to analyze 45 samples from cases evaluated for the presence of Entamoeba spp. by microscopy and a real-time PCR method capable of differential detection of E. histolytica and E. dispar. Our results demonstrated an agreement of approximately 98% (45/44) between the real-time PCR for E. histolytica and E. dispar and the 18S rRNA analysis described here. Five previously negative samples by microscopy revealed the presence of E. dispar, E. hartmanii, or E. coli DNA. In addition, we were able to detect E. hartmanii in a stool sample that had been previously reported as negative for Entamoeba spp. by microscopy. Further microscopic evaluation of this sample revealed the presence of E. hartmanii cysts, which went undetected during the first microscopic evaluation. This PCR followed by DNA sequencing will be useful to refine the diagnostic detection of Entamoeba spp. in stool and other clinical specimens.

Summary: This review highlights the importance of human angiostrongyliasis as a significant foodborne zoonosis caused by Angiostrongylus costaricensis and Angiostrongylus cantonensis. The re-emergence and urban expansion of angiostrongyliasis pose significant concerns for the control of these infections worldwide. The article focuses on various aspects of the disease, with a special emphasis on the illness caused by A. cantonensis.

Summary: This study developed a new sequencing-based method for detecting the coccidian parasite Cyclospora cayetanensis, which causes diarrheal illness outbreaks. The method showed high specificity and was able to accurately detect C. cayetanensis in food and water samples. This sensitive detection method adds to the range of screening strategies for food and water samples.

Targeted next generation sequencing of Cyclospora cayetanensis mitochondrial genomes from seeded fresh produce and other seeded food samples

Summary: This study developed a highly sensitive custom-designed targeted sequencing method capable of obtaining near-complete mitochondrial genome sequences of C. cayetanensis from food samples with low levels of contamination. The success of this method represents a critical milestone towards the potential development of a comprehensive genotyping method for C. cayetanensis.

Functional Feeding Group composition and attributes: evaluation of freshwater ecosystems in Atlantic Forest, Brazil

Summary: This study used benthic macroinvertebrates Functional Feeding Group (FFG) as indicators to assess the effects of stream variables on FFG abundance and richness, and evaluate FFG ratios as a tool to assess ecological conditions. Results showed significant differences in FFG richness among sites with different impairment conditions, and FFGs responded differently to abiotic variables. The FFG ratio analysis was found to be a fast and cost-effective tool for monitoring aquatic ecosystems in the Atlantic Forest biome, but further calibration is needed for this region.
