Summary: A scalable continuous flow process has been developed for the efficient and safe electrophilic alpha-aminohydroxylation of enolizable ketones using alpha-chloronitroso derivatives. The process showcases the economic and efficient nature of alpha-chloronitroso derivatives in transferring electrophilic synthons of hydroxylamine, allowing for the preparation of valuable molecules with high atom economy. The process features sequential modules, in-line extractions, and in-line analytical procedures to improve process control and safety, making it easily transposable to pilot scale and ideal for compound library screening in medicinal chemistry.

Summary: To combat bacteria and biofilm infections, this study developed bioactive chitin/Mn3O4 composite hydrogels with outstanding photothermal antibacterial and antibiofilm activities. These hydrogels showed favorable biocompatibility and antioxidant properties, and demonstrated excellent wound healing performance in a mouse model with S. aureus biofilm-infected wounds. This research broadens the scope for chitin hydrogels with antibacterial properties and offers a promising alternative for bacterial-associated wound infection therapy.

Summary: Detection and quantification of bioanalytes using NIR probes have advantages of negligible light scattering, minimal interference from background matrices, and deep sample penetration. This study presents a supramolecular dye-polymer complex as a promising NIR emitting turn-on probe for protamine sensing. The complex shows fluorescence enhancement due to binding onto the polymer surface, and further enhancement in the presence of protamine. This selective turn-on response is attributed to a structural change in the polymer upon interaction with protamine.

Summary: Genome assembly is a computational technique used to create a comprehensive and precise representation of the entire genome by piecing together DNA fragments generated by sequencing technologies. Generating a high-quality human reference genome is crucial for understanding human biology and downstream genomic variation analysis. This paper benchmarks various strategies for human genome assembly using different sequencing technologies and tools, and compares their performances in terms of continuity, accuracy, completeness, variant calling, and phasing. It recommends the use of PacBio HiFi long-reads for low base errors, Oxford Nanopore long-reads for continuous contigs (with further polishing for quality improvement), and Hi-C for chromosome-level scaffolding. Combining multiple technologies can further enhance the quality and completeness of genome assembly, and for diploid assembly, hifiasm is recommended as the best tool for human diploid genome assembly using PacBio HiFi and Hi-C data. Advances in human diploid assemblers using PacBio HiFi reads and other technologies are expected to enable high-quality, chromosome-level, and haplotype-resolved human genome assemblies in the future.
