Summary: F-18-fluorination is an important and rapidly growing field in organic synthesis that has attracted significant attention from chemists. This manuscript presents a biased perspective on recent advances in the field, with a focus on the author's own chemistry, and discusses challenges for the development of new chemistry with potential impact on imaging. The aim is to provide a useful guide for chemists to develop reliable reaction chemistry suitable for radiofluorination to make a real impact on human health.

Summary: Here, a general method for copper-catalyzed N-arylation of isatoic anhydrides with unsymmetrical iodonium salts at room temperature is reported. The protocol is mild and simple, employing aryl(TMP)iodonium trifluoroacetate as the arylating partner, and provides access to N-arylated isatoic anhydrides with diverse aryl groups in moderate to excellent yields (53-92%). The methodology is also compatible with substituted isatoic anhydrides and can be used for the synthesis of biologically relevant fenamic acid derivatives and N,N'-diarylindazol-3-ones in a one-pot step economical system, as demonstrated here. The scale-up synthesis of flufenamic acid is also illustrated.

Summary: A facile synthesis of triarylsulfonium triflates has been reported, where selective aryl transfer from readily available mesityl iodonium(III) salts to diarylsulfides is achieved in the presence of a copper catalyst. The advantage of this protocol is the efficient and selective S-arylation of diarylsulfides to produce triarylsulfonium triflates without the formation of undesired byproduct salts.

Summary: This study investigated the complexation of two acyclic aminopolycarboxylate chelators with trivalent lanthanides and minor actinides. The chelators, H(4)octapa and H(4)pypa-peg, are suitable for radiometal coordination and f-element isolation. H(4)octapa forms a compact coordination packet, while H(4)pypa-peg is less constrained. The solubility of H(4)pypa-peg is increased by introducing a polyethylene glycol group, without affecting its ability to complex the lanthanides and minor actinides.

Summary: Alkylammonium salts, particularly tetraoctylammonium, have been used to investigate the structure and function of potassium channels. In this study, it was found that TOA(+) binds to the channel cavity with high affinity and causes a shift in the equilibrium of the channel's selectivity filter conformation towards an inactivated-like form. Additionally, the TOA(+)-bound state differs significantly from the collapsed channel state described by X-ray crystallography as the inactivated form of KcsA.

Summary: A new 1,2,4-triazole-pyrrolo[1,2-a]pyrazine hybrid system, 6-acylpyrrolo[1,2-a][1,2,4]triazolo[5,1-c]pyrazine, was synthesized using a catalyst-free method. This method involved sequential exposure of pyrrole-2-carbonitrile-derived substrates to DMF-DMA and acyl hydrazide, resulting in the formation of acylated pyrazine and 1,2,4-triazole rings, enabling the installation of various substituents at specific positions on the core skeleton.

Summary: Alcohol dehydrogenases are enzymes that use cofactors for oxidation or reduction reactions of alcohols or carbonyl compounds. They are utilized in green chemistry and have applications in the production of pharmaceuticals. Recombinant enzymes have solved the challenge of producing purified enzymes in large quantities. Engineered alcohol dehydrogenases have been used in asymmetric synthesis in industry. Various methods have been established for regenerating expensive cofactors to make the enzymatic process more efficient and economically viable.
