This paper provides a synopsis of the main papers on diagnosis, imaging, treatment, prognostication and treatment outcomes in patients with oral and oropharyngeal squamous cell carcinoma (OSCC) and head and neck SCC (HNSCC) published in 2008 in Oral Oncology - an international interdisciplinary journal which publishes high quality original research, clinical trials and review articles, and all other scientific articles relating to the aetiopathogenesis, epidemiology, prevention, clinical features, diagnosis, treatment and management of patients with neoplasms in the head and neck, and orofacial disease in patients with malignant disease. Summary: Oral potentially malignant disorders (OPMDs) are associated with an increased risk of oral cancer, and an expert group updated the nomenclature and classification of OPMDs, adding oral lichenoid lesions and oral manifestations of 'chronic graft-versus-host disease' to the list. It is recommended to establish a global research consortium to study the natural history of OPMDs based on the proposed classification.

Malignant transformation of oral proliferative verrucous leukoplakia: A systematic review and meta-analysis

Summary: This study investigated the malignant transformation of oral proliferative verrucous leukoplakia, finding that females and gingiva were more commonly affected. The pooled proportion of malignant transformation was 43.87%, with conventional squamous cell carcinoma being the most frequent histopathological subtype. No conclusive results were found between malignant transformation and sex, age, tobacco, or alcohol consumption.

Differentially methylated genes in proliferative verrucous leukoplakia reveal potential malignant biomarkers for oral squamous cell carcinoma

Potential Non-Invasive Biomarkers for Early Diagnosis of Oral Squamous Cell Carcinoma

Prediction of the risk of cancer and the grade of dysplasia in leukoplakia lesions using deep learning

On the Oral Microbiome of Oral Potentially Malignant and Malignant Disorders: Dysbiosis, Loss of Diversity, and Pathogens Enrichment

Summary: This study aims to compare and describe the composition and diversity of the oral microbiome in patients with homogeneous leucoplakia, proliferative verrucous leukoplakia, oral squamous cell carcinoma, and OSCC preceded by PVL. The number of bacterial species was lower in cancer patients, with Fusobacteriota being the dominant group. PVL and PVL-OSCC patients had a higher abundance of Campilobacterota and lower Proteobacteria compared to other groups. This is the first study comparing oral microbiome alterations in these groups, and further research is needed.

Oral cancers preceded by proliferative verrucous leukoplakia exhibit distinctive molecular features

Summary: This study investigated the pathophysiologic differences between proliferative verrucous leukoplakia-associated oral squamous cell carcinoma (PVL-OSCC) and non-PVL-OSCC through transcriptomic and DNA methylation analyses. The results showed that PVL-OSCC patients had lower expression of cancer-related genes and hypermethylation of the promoter region of many genes, suggesting that DNA methylation could be a regulatory mechanism.

Effect of clinical and histologic features on time to malignancy in 224 cases of oral leukoplakia treated by surgery

Comprehensive clinical, genome and transcriptomic analysis of primary ghost cell odontogenic carcinoma

Summary: This study presents a comprehensive clinical, genomic, and transcriptomic analysis of primary ghost cell odontogenic carcinoma (GCOC). The results provide the first comprehensive molecular atlas for primary GCOC, including previously identified CTNNB1 mutation and novel alterations in MAP3K, EP300, and the 22q11.21 region. The transcriptome analysis reveals significant involvement of cytokine-cytokine receptor interaction and the PI3K-Akt signaling pathway. More GCOC cases should be compared to validate these findings for accurate clinical guidance.

Transoral robotic surgery in oropharyngeal squamous cell carcinoma: A comparative study between da Vinci Single-Port and da Vinci Xi systems

Summary: This study compares the outcomes of transoral resections using the da Vinci SP and da Vinci Xi systems for oropharyngeal squamous cell carcinoma (OPSCC). The results indicate that the da Vinci SP system has advantages in terms of reduced docking and console times, as well as improved rates of wide negative resection margins. However, there were no significant differences in complication rates or postoperative functional outcomes between the two systems.
