Prostacyclin receptor (PTGIR) in the porcine endometrium: Regulation of expression and role in luminal epithelial and stromal cells

The prostacyclin (PGI(2)) signaling pathway plays an important role during early pregnancy in rodents and ruminants. Abundant concentrations of PGI(2) were also found in the endometrium and uterine lumen of gilts during the period of implantation. The present study was designed to examine (1) the expression of PGI(2) receptor (PTGIR) messenger RNA (mRNA) and protein in the endometrium of cyclic and early-pregnant gilts; (2) possible regulation of endometrial PTGIR gene expression by conceptus products, estradiol and cytokines; (3) the effect of iloprost (a PGI(2) analogue) on cAMP formation and the expression of fibroblast growth factor-2 (FGF-2) and vascular endothelial growth factor (VEGF; isoform 164) mRNA in luminal epithelial (LE) and stromal (ST) cells. Increased PTGIR mRNA expression in the endometrium was detected on Days 11 to 12 and 18 to 20 of pregnancy compared with respective days of the estrous cycle (P < 0.05). Moreover, greater PTGIR protein level was observed in pregnant than in cyclic gilts on Days 11 to 12 after estrus (P < 0.05). Gilts with unilateral pregnancy revealed abundant PTGIR expression in the endometrium collected from the gravid uterine horn of Day-11 pregnant gilts compared with the respective horn of cyclic animals (P < 0.01). Both LE and ST cells of the endometrium possess PTGIR protein. Moreover, IL1 beta, IFN gamma, and conceptus-exposed medium, but not estradiol, stimulated PTGIR mRNA expression in LE and ST cells in vitro. Activation of PTGIR by incubation of LE and ST cells with iloprost resulted in greater CAMP generation (P < 0.01). Moreover, iloprost increased FGF-2 and VEGF(164) mRNA expression in ST (P < 0.05), but not LE cells. In conclusion, this study revealed increased expression of PTGIR in the porcine endometrium during the pen-implantation period and reported a possible regulation of PTGIR abundance by conceptus-derived factors. Moreover, besides its important role in vascular system, PGI(2) may promote the expression of proangiogenic genes in the uterine stroma. (C) 2015 Elsevier Inc. All rights reserved.

The maturation of spermatozoa in the female reproductive tract is crucial for fertilization. Follicular fluid (FF) and its extracellular vesicles (EVs) play a significant role in supporting sperm functions, with specific effects based on species and source of FF EVs. This study demonstrates that bovine FF EVs can support sperm function and contribute to a favorable periconceptional microenvironment.

Summary: Psoriasis vulgaris (PsV) and psoriatic arthritis (PsA) are inflammatory diseases with unresolved pathophysiological aspects. In this study, the miRNA contents and surface proteome of blood serum extracellular vesicles (EVs) were compared between PsV and PsA patients and healthy controls. It was found that there were differentially enriched EV-bound miRNAs in both PsV and PsA groups, suggesting that EVs may play a role in the pathophysiological processes of PsV and PsA.

Diverse effects of prostacyclin on angiogenesis-related processes in the porcine endometrium

Summary: In this study, the researchers investigated the regulation of prostacyclin (PGI2) synthesis and PGI2 receptor (PTGIR) expression by cytokines, and evaluated the effect of PGI2 on pro-angiogenic gene expression, cell proliferation and migration, cell cycle distribution, and capillary-like structure formation in porcine endometrial cells. They found that IL1ss, IFNgimel, and/or TNFα increased PGI2 secretion and PTGIR expression. PGI2 enhanced pro-angiogenic gene expression, cell proliferation, and cell cycle progression.

Pyruvate enhances stallion sperm function in high glucose media improving overall metabolic efficiency

This improvement is attributed to the conversion of lactate to pyruvate and increased NAD(+) enhancing glycolysis efficiency.

Differential transcript usage across mammalian oocytes at the germinal vesicle and metaphase II stages

Summary: Ongoing progress in mRNA-Sequencing technologies has contributed to the refinement of assisted reproductive technologies. This study utilized mRNA-Seq datasets of various mammalian species to analyze differential transcript usage (DTU) and identified ten cross-species DTU genes. Gene Ontology (GO) term enrichment analysis revealed the involvement of these genes in RNA and cell-cycle control mechanisms, suggesting their importance during oocyte maturation. Further exploration of these genes' transcript isoforms may lead to the discovery of novel markers for reproductive technologies.

Effects of Nobiletin supplementation on the freezing diluent on porcine sperm cryo-survival and subsequent in vitro embryo development

Summary: This study found that Nobiletin (NOB) can promote the cryosurvival and embryo development of porcine sperm after in vitro fertilization (IVF). The addition of NOB improves sperm motility, acrosome integrity, viability, and mitochondrial membrane potential, reduces lipid peroxidation, and increases cleavage and blastocyst formation rates. Furthermore, NOB treatment upregulates the expression of genes related to pluripotency and cell differentiation in the resulting embryos.

Fluorochloridone induces mitochondrial dysfunction and apoptosis in primary goat Sertoli cells

Summary: This study investigated the cytotoxicity and toxicological mechanisms of fluorochloridone (FLC) on cultured goat Sertoli cells. The results showed that FLC exposure induced oxidative stress, promoted apoptosis and autophagy, inhibited lysosomal biogenesis, and blocked autophagic flux in goat Sertoli cells. FLC also caused mitochondrial dysfunction and disturbed mitophagy, leading to cell apoptosis. The antioxidant N-acetyl-L-cysteine (NAC) reduced FLC-induced ROS accumulation and reversed the disorder of autophagy levels.
