Purpose: To evaluate in vivo features of Schlemm's canal (SC) in patients with primary open-angle glaucoma (POAG) with spectral-domain optical coherence tomography (SD-OCT) and to investigate the relationship of SC size with intraocular pressure (IOP) and glaucoma severity. Design: Prospective, comparative study. Participants: Fifty Chinese patients with newly diagnosed POAG who had not undergone surgery and 50 normal Chinese subjects from a population-based, cross-sectional study in Shanghai. Methods: All participants underwent SD-OCT. The diameter and area of SC were examined in the temporal and nasal sections and measured with customized software. Main Outcome Measures: Patient demographics, repeatability and reproducibility assessed with the coefficient of variation (CV) and the intraclass correlation coefficient (ICC), SC parameters and their correlation with IOP, and the mean deviation (MD) of the visual field were analyzed. Results: The percentage of sections in which SC was observable was similar between eyes with POAG and normal eyes, and ranged from 78% to 86%. For intraobserver repeatability, the CV and ICC values were 7.9% and 0.97 for diameter, and 13.8% and 0.83 for area, respectively. For interobserver repeatability, the CV and ICC values were 13.6% and 0.89 for diameter, and 13.4% and 0.80 for area, respectively. Significant differences between the 2 groups were found for the average SC area (11 332 +/- 2015 mu m(2) vs. 13 991 +/- 1357 mu m(2); P<0.001), but not for the SC diameter (40.2 +/- 7.1 mu m vs. 45.2 +/- 4.0 mu m; P = 0.195). In addition, the mean IOP values correlated well only with the SC area (rho = -0.674, P<0.001), not with the SC diameter (rho = -0.103, P = 248). No significant correlations were found between the MD values and the SC parameters. Conclusions: Eyes with POAG have a decreased SC area compared with normal eyes. A correlation between the SC area and the IOP also was observed. However, the degree of glaucoma damage was not consistently associated with the SC area. Spectral-domain OCT could be used for investigating SC changes in patients with glaucoma. Design of a Novel Fab-Like Antibody Fragment with Enhanced Stability and Affinity for Clinical use

Chunyu Wang, Jiaxu Hong, Zhenlin Yang, Xujiao Zhou, Yuhan Yang, Yu Kong, Binfan Chen, Huifang Wu, Bin-Zhi Qian, Dimiter S. Dimitrov, Xingtao Zhou, Yanling Wu, Tianlei Ying

Summary: This study introduced a novel Fab-like antibody fragment, FabCH3, generated via an in silico-based engineering approach, which exhibited improved stability and affinity compared to the parental Fab. High-resolution crystal structure analysis revealed that FabCH3 has more rigid structures, explaining its enhanced performance. The stabilized FabCH3 provides a versatile platform for engineers to develop Fab-like antibody fragments with higher stability and antigen-binding affinity.

All-small-molecule supramolecular hydrogels assembled from guanosine 5′-monophosphate disodium salt and tobramycin for the treatment of bacterial keratitis

Xuejing Cheng, Huiyu Chen, Fang Yang, Jiaxu Hong, Yiyun Cheng, Jingjing Hu

Summary: Bacterial keratitis is a common corneal infection that can lead to blindness. Current clinical treatment with antibiotic eye drops is not effective. This study proposes a supramolecular hydrogel composed of guanosine-5'-monophosphate disodium salt and tobramycin, which shows promising therapeutic efficacy and good biocompatibility for bacterial keratitis.

Clinical Investigation of the Safety and Efficacy of Low-temperature Plasma as an Adjuvant Treatment for Mild to Moderate Fungal Keratitis: A Pilot Study

Jingyi Cheng, Anji Wei, Jiaxu Hong, Jianjiang Xu

Summary: This pilot study found that low-temperature plasma (LTP) could be an effective adjuvant treatment for mild to moderate fungal keratitis. Patients in the LTP group had a higher healing rate, deeper initial ulcer depth, and better visual improvement. However, LTP treatment also had risks, and some patients experienced adverse reactions and treatment failure.

Application of Keratograph and Fourier-Domain Optical Coherence Tomography in Measurements of Tear Meniscus Height

Minjie Chen, Anji Wei, Jianjiang Xu, Xingtao Zhou, Jiaxu Hong

Summary: This study compares the interoperator repeatability and agreement between tear meniscus height (TMH) measurements obtained with a keratograph and Fourier-domain optical coherence tomography (FD-OCT) and evaluates their efficacy in detecting dry eye disease (DED). The results reveal that the TMHs measured with the keratograph are slightly lower than those obtained with FD-OCT. FD-OCT TMH measurements are more reliable than keratograph data in the DED group, although there is poor agreement between the two devices in both groups.

Summary: This study investigated the morphological and functional changes of meibomian glands in pediatric and adult patients with allergic conjunctivitis. The results showed that meibomian glands in pediatric patients improved after treatment, while there was no improvement in adult patients. Age and baseline meibomian gland area ratio were predictors of improvement in meibomian glands.

The relationship between corneal biomechanical parameters and treatment outcomes of orthokeratology lenses

Xia Li, Jianjiang Xu, Jiaxu Hong, Jing Yao

Summary: Long-term orthokeratology treatment can improve corneal biomechanical properties in myopic eyes and slow down myopia progression. Smaller vertical axial deformation, reduced maximum eye movement, and decreased Ambrosio's relational thickness to the horizontal profile can predict treatment outcomes.

Summary: Dry eye disease (DED) is a common chronic eye disease that requires fast and reliable tools for diagnosis and monitoring. This study developed a biosensor based on silicon nanowire-based field-effect transistor (SiNW FET) for the quantitative measurement of MMP-9 in human tears. The biosensor demonstrated high agreement with enzyme-linked immunosorbent assay (ELISA) and showed good diagnostic sensitivity and specificity in real-world clinical settings.

CRISPR-free, programmable RNA pseudouridylation to suppress premature termination codons

Jinghui Song, Liting Dong, Hanxiao Sun, Nan Luo, Qiang Huang, Kai Li, Xiaowen Shen, Zhe Jiang, Zhicong Lv, Luxin Peng, Meifang Zhang, Kun Wang, Ke Liu, Jiaxu Hong, Chengqi Yi

Summary: Researchers have developed a programmable RNA base editor called RESTART to reverse premature translation termination caused by nonsense mutations. RESTART utilizes an engineered guide snoRNA and the endogenous H/ACA box snoRNP machinery to achieve precise pseudouridylation, enabling functional PTC readthrough in disease-relevant contexts. The tool also demonstrates robust pseudouridylation in primary cells.

Yinglei Zhang, Chunyan Hu, Cong Niu, Jiaxu Hong, Xujiao Zhou

Summary: This study investigated the effect of asiatic acid (AA) on the quantity and function of retinal ganglion cells (RGCs) in chronic glaucoma rat models and how AA regulates synaptic pathways. The rat model was created by electrocoagulation of the episcleral veins. AA reduced the loss of RGCs caused by chronic glaucoma, decreased excitatory neurotransmission to RGCs, and increased inhibitory neurotransmission.

Yue Li, Yuda Wei, Gang Li, Shiqian Huang, Jianjiang Xu, Qiurong Ding, Jiaxu Hong

Summary: This study developed a CRISPR/Cas9 system targeting the major HSV receptor NECTIN-1 on human corneal epithelial cells (HCECs).
